N-acetylserotonin alleviates retinal ischemia-reperfusion injury via HMGB1/RAGE/NF-κB pathway in rats

AIM:To observe the effects of N-acetylserotonin(naS)administration on retinal ischemia-reperfusion(RIR)injury in rats and explore the underlying mechanisms involving the high mobility group box 1(HMGB1)/receptor for advanced glycation end-products(RAGE)/nuclear factor-kappa B(NF-κB)signaling ***:A rat model of RIR was developed by increasing the pressure of the anterior chamber of the *** male Sprague Dawley were randomly divided into five groups:sham group(n=8),RIR group(n=28),RIR+naS group(n=28),RIR+FPS-ZM1 group(n=8)and RIR+naS+FPS-ZM1 group(n=8).The therapeutic effects of naS were examined by hematoxylin-eosin(H&E)staining,and retinal ganglion cells(RGCs)*** expression of interleukin 1 beta(IL-1β),HMGB1,RAGE,and nod-like receptor 3(NLRP3)proteins and the phosphorylation of nuclear factorkappa B(p-NF-κB)were analyzed by immunohistochemistry staining and Western blot *** expression of HMGB1 protein was also detected by enzyme-linked immunosorbent assay(ELISA).RESULTS:H&E staining results showed that naS significantly reduced retinal edema and increased the number of RGCs in RIR *** naS therapy,the HMGB1 and RAGE expression decreased significantly,and the activation of the NF-κB/NLRP3 pathway was antagonized along with the inhibition of p-NF-κB and NLRP3 protein ***,naS exhibited an anti-inflammatory effect by reducing IL-1β*** inhibitory of RAGE binding to HMGB1 by RAGE inhibitor FPS-ZM1 led to a significant decrease of p-NF-κB and NLRP3 expression,so as to the IL-1βexpression and retinal edema,accompanied by an increase of RGCs in RIR ***:naS may exhibit a neuroprotective effect against RIR via the HMGB1/RAGE/NF-κB signaling pathway,which may be a useful therapeutic target for retinal disease.

In this paper,high-energy Ne ions were used to irradiate Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) metallic glass(MG)and crystalline W to investigate their difference in mechanical response after *** results showed that with the irradiation dose increased,the tensile micro-strain increased,nano-hardness increased from 7.11 GPa to 7.90 GPa and 8.62 GPa,young’s modulus increased,and H3/E2 increased which indicating that the plastic deformability decreased in crystalline *** the same irradiation conditions,the Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) MG still maintained the amorphous structure and became more disordered despite the longer range and stronger displacement damage of Ne ions in Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) MG than in crystalline *** the irradiation hardening and embrittlement behavior of crystalline W,Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) MG showed the gradual decrease in hardness from 6.02 GPa to 5.89 GPa and 5.50 GPa,the decrease in modulus and the increase in plastic deformability with the increasing ***,the irradiation softening and toughening phenomenon of Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) MG could provide new ideas for the design of nuclear materials.

Diagnostic evaluation of endoscopic ultrasonography with submucosal saline injection for differentiating between T1a and T1b early gastric cancer

BACKGROUND Endoscopic ultrasonography(EUS)has become a reliable method for predicting the invasion depth of early gastric cancer(EGC).However,diagnostic accuracy of EUS is affected by several *** particular,it is difficult to differentiate between T1a and T1b EGC through *** To confirm whether submucosal saline injection(SSI)could improve the accuracy of EUS in distinguishing T1a and T1b lesions in *** Twenty-four patients with EGC were examined by EUS and subsequently by SSI combined EUS to compare the degree of tumor ***,they underwent endoscopic or surgical resection within 7 *** diagnostic accuracy of EUS and SSI combined EUS was evaluated based on the final pathological findings *** injected into the submucosa acted as an echoic contrast enhancing agent and had the effect of distinguishing the mucosal and submucosal layers *** Of total 24 patients,23 were diagnosed with EGC(T1 cancer:13 as T1a,and 10 as T1b).Standard EUS identified 6 of 13 T1a cancer patients and 3 of 10 T1b cancer ***,EUS-SSI identified 12 of 13 T1a cancer patients and 6 of 10 T1b cancer *** this study,SSI combined EUS was more accurate than EUS alone in diagnosing T1a and T1b lesions of EGC(75.0%and 37.5%,respectively).CONCLUSION SSI improved the diagnostic accuracy of EUS in distinguishing between the T1a and T1b stages in EGC.

Effect of Lycopus lucidus *** on gut microflora and short chain fatty acid composition in Crj:CD-1 mice

We investigated the diversity and composition of microflora in feces of Lycopus lucidus Turcz.-fed *** addition,we evaluated the production of major cytokines(Interleukin-6 and-10)which are related to inflammation and fatty acid composition of several tissues.16S ribosomal Dna sequencing-based microbiome taxonomic profiling analysis was performed utilizing the EzBioCloud data *** mice fed on *** showed a significantly reduced number of lactic acid bacteria and coliform in the feces compared with the control group(pna sequencing analysis of fecal samples showed that *** supplementation decreased the community of harmful microflora(Enterobacteriaceae including Escherichia coli and Bacteroides sp.)in feces compared with the control group(pna expression of cytokine IL-6 and IL-10 between the control and *** fed *** fecal fatty acid composition in the *** group had percentages of 4:0,6:0,8:0 and 10:0 in the intestine but those short chain fatty acids were not detected in the control *** results showed that *** supplementation influenced gut environment by decreasing harmful microflora and increased the percentages of several short fatty acids.

We have conducted a comprehensive investigation into the bright single pulse emission from PSR B1133+16using the Giant Metrewave Radio *** time resolution data(61μs)were obtained at a center frequency of 322 MHz with a bandwidth of 32 MHz over a continuous observation period of 7.45 hr.A total of 1082 bright pulses were sporadically detected with peak flux densities ranging from 10 to 23 times stronger than the average pulse ***,no giant pulse-like emission with a relative pulse energy larger than 10 and extremely short duration was detected,indicating that these bright pulses cannot be categorized as giant pulse *** majority of these bright pulses are concentrated in pulse phases at both the leading and trailing windows of the average pulse profile,with an occurrence ratio of approximately *** pulse energy distribution for all individual pulses can be described by a combination of two Gaussian components and a cutoff power-law with an index of α=-*** updated nulling fraction of 15.35%±0.45% was determined from the energy *** emission of individual pulses follows a log-normal distribution in peak flux density *** is imperative that regular phase drifting in bright pulse sequence is identified in both the leading and trailing components for the first *** physical mechanisms are discussed in detail to provide insights into these observations.

Improving thermal efficiency and stability of laser welding process for magnesium alloy by combining power modulation and subatmospheric pressure environment

The laser welding(LW)process of highly reflective materials presents low thermal efficiency and poor *** solve the problem,the effects of subatmospheric environment on LW process,technological parameters in subatmospheric environment on weld formation and welding with sinusoidal modulation of laser power on the stability of LW process in subatmospheric environment were *** AZ31magnesium(Mg)alloy was used as the test *** test result revealed that the weld penetration in subatmospheric environment can increase by more than ten times compared with that under normal *** the keyhole depth greatly rises,significantly periodic local bulge is observed on the backwall surface of the keyhole and the position of the bulge shifts along the direction of the keyhole ***,the hump-shaped surface morphology of the welded seam is formed;moreover,the weld width in local zones in the lower part of the welded seam remarkably *** LW in subatmospheric environment,the weld penetration can be further greatly increased through power ***,power modulation can inhibit the occurrence of bulges in local zones on the backwall of the keyhole during LW in subatmospheric environment,thus further curbing the significant growth of the weld widths of hump-shaped welding beads and local zones in the lower part of welded ***,the mechanism of synchronously improving the thermal efficiency and stability of LW process of highly reflective materials through power modulation in subatmospheric environment was *** was conducted according to theoretical analysis of recoil pressure and observation results of dynamic behaviors of laser induced plasma clouds and keyholes in the molten pool through high speed photography.
