Host-associated probiotics(HAPs)are bacteria originally isolated from rearing water or the host's gastrointestinal tract in order to enhance the host's growth and health.This study investigated the HAP potential of Bacillus sp.PM8313,isolated from wild red sea bream(Pagrus major),through characterization and feeding trials.Results based on in vitro tests showed that PM8313 is safe,confirming its hemolytic,cytotoxic,and antibiotic resistance.In addition,PM8313 showed advantages as a probiotic with high viability in the gastrointestinal model and a high cell adhesion rate.Whole-genome sequencing demonstrated that PM8313 has a 4,615,871 bp single circular chromosome and a guaninecytosine content of 45.25%.It also showed the absence of genes encoding virulence factors,such as cytotoxin,enterotoxin,hemolysin,sphingomyelinase,and phospholipase.In the feeding trial,a supplemental diet of 1×10^(8)CFU/g PM8313 positively altered the weight gain,digestive enzyme activity,and intestinal microbiota composition of red sea bream.Analysis of nonspecific immune parameters and immune-related gene expression,and a challenge test showed that PM8313 supplementation increases immunity and pathogenic bacteria resistance.Our findings suggest that PM8313 should be considered for application as a novel HAP to red sea bream aquaculture.

Ameloblasts are specialized cells derived from the dental epithelium that produce enamel,a hierarchically structured tissue comprised of highly elongated hydroxylapatite(OHAp)crystallites.The unique function of the epithelial cells synthesizing crystallites and assembling them in a mechanically robust structure is not fully elucidated yet,partly due to limitations with in vitro experimental models.Herein,we demonstrate the ability to generate mineralizing dental epithelial organoids(DEOs)from adult dental epithelial stem cells(aDESCs)isolated from mouse incisor tissues.DEOs expressed ameloblast markers,could be maintained for more than five months(11 passages)in vitro in media containing modulators of Wnt,Egf,Bmp,Fgf and Notch signaling pathways,and were amenable to cryostorage.When transplanted underneath murine kidney capsules,organoids produced OHAp crystallites similar in composition,size,and shape to mineralized dental tissues,including some enamel-like elongated crystals.DEOs are thus a powerful in vitro model to study mineralization process by dental epithelium,which can pave the way to understanding amelogenesis and developing regenerative therapy of enamel.

随着全球能源需求增长和环境污染加剧,发展可持续能源减少对化石燃料(如石油、天然气和煤炭等)的消耗成为实现人类社会可持续发展的关键.氢能因其能量密度高、燃烧无污染、应用形式多样被认为是最理想的替代能源.电解水制氢包括阴极析氢反应(HER)和阳极析氧反应(OER),具有绿色环保、生产灵活和纯度高等特点,是理想的绿色生产技术之一.然而,阳极电解水产氧反应动力学缓慢,导致阴极的产氢效率低.此外,在电解水过程中,会产生高氧化性的过氧化氢(H_(2)O_(2)),降低电解水膜的寿命,阻碍电解水技术的实际应用.因此,亟待开发新型高效、稳定且具有高附加值的电解水催化剂.目前,电化学整体水分解(OWS)制氢技术存在安全风险、投资回报低、阳极OER动力学慢和电能消耗大等问题,将阳极氧化反应与混合电解水(HWE)装置中的HER相结合,借助热力学较好的电氧化反应取代缓慢的传统OER反应协同产氢,可以有效克服传统电解水的产率不足,解决污染排放和生物质回收问题.本文综述了协同电催化用于联产氢气和低能耗、高法拉第效率高价值产品的催化剂结构设计,揭示不同协同电催化系统的催化途径和意义,以实现更高效、零碳排放的目标.首先,介绍了HWE系统的发展现状,重点关注各种富氢物质的协同电解,例如酒精、生物质衍生物、葡萄糖和在阳极形成的高附加值化学品.与传统阳极OER工艺相比,有机/生物质底物小分子的OER表现出较低的热力学需求,降低产氢能耗.随后,详细介绍了基于阴极HER和阳极OER协同电解反应、协同催化HWE高效电极/电催化剂的合理设计,以实现高催化活性、高选择性和良好的电化学稳定性.重点讨论了新型电极/电催化剂设计、活性改进以及结构-催化活性关系提升的合成策略.再后,讨论了基于有机/生物质小分子协同HWE系统电催化的代表性研究进展和突破,强调了其在促进可持续低压制氢方面的重要作用,并回顾了近年来HWE的研究突破,同时,对一些可行性分析和机理探索进行比较,为制氢提供了新的研究方向.最后,提出了协同电催化制氢面临的挑战并展望未来的研究方向.综上,大多数电催化剂存在催化活性低、稳定性差等问题,要实现可持续、经济高效和清洁的产氢技术,仍有很多方面需要进一步的深入研究.本文综述了高效多功能HWE系统发展现状和催化剂结构设计,为电解水制氢和高附加值产品的节能联产提供一定的参考.

Trimetallic Au@PdPb nanowires for oxygen reduction reaction

The development of highly efficient and stable Pd-based catalysts is crucial to improve their sluggish oxygen reduction reaction(ORR)kinetics in acid media.To improve ORR activity and utilization efficiency of Pd,an ideal catalyst should have ORR-favorable chemical environment,optimized geometric structure,and long periods of operation.In this work,we first synthesize a novel trimetallic Au@PdPb core–shell catalyst consisting of PdPb alloy nano-layers grown on the surface of ultrathin Au nanowires(NWs)by a two-step water-bath method.The Au@PdPb NWs have the merits of anisotropic one-dimensional nanostructure,high utilization efficiency of Pd atoms and doping of Pb atoms.Because of the structural and multiple compositional advantages,Au@PdPb NWs exhibit remarkably enhanced ORR activity with a high haIf-wave potential(0.827 V),much better than those of commercial Pd black(0.788 V)and bimetallic Au@Pd NWs(0.803 V).Moreover,Au@PdPb NWs display better electrocatalytic stability for the ORR than those of Pd black and Au@Pd NWs.This study demonstrates the validity of our approach for deriving highly ORR-active Pd-based catalysts by modifying their structure and composition.

Generation of tumor spheroids using a droplet-based microfluidic device for photothermal therapy

Despite their simplicity,monolayer cell cultures are not able to accurately predict drug behavior in vivo due to their inability to accurately mimic cell-cell and cell-matrix interactions.In contrast,cell spheroids are able to reproduce these interactions and thus would be a viable tool for testing drug behavior.However,the generation of homogenous and reproducible cell spheroids on a large scale is a labor intensive and slow process compared to monolayer cell cultures.Here,we present a droplet-based microfluidic device for the automated,large-scale generation of homogenous cell spheroids in a uniform manner.Using the microfluidic system,the size of the spheroids can be tuned to between 100 and 130μm with generation frequencies of 70 Hz.We demonstrated the photothermal therapy(PTT)application of brain tumor spheroids generated by the microfluidic device using a reduced graphene oxide-branched polyethyleneimine-polyethylene glycol(rGO-BPEI-PEG)nanocomposite as the PTT agent.Furthermore,we generated uniformly sized neural stem cell(NSC)-derived neurospheres in the droplet-based microfluidic device.We also confirmed that the neurites were regulated by neurotoxins.Therefore,this droplet-based microfluidic device could be a powerful tool for photothermal therapy and drug screening applications.

In order to explore the possibility of enhanced creep resistance by grain size control, creep tests of stress-relieved and annealed Zr-1Nb-0.7Sn-0.1Fe cladding tubes were carried out at intermediate temperatures. The creep rate decreased significantly with increase of grain size from .2 μm to 4 μm with annealing. The stress exponent was observed to be in the range of 5.0.5 for both stress-relieved and annealed Zr-1Nb-0.7Sn- 0.1Fe, suggesting that the dislocation climb is the rate controlling creep mechanism. It was shown that not only the creep rate decreased but also creep life increased in the annealed Zr-1Nb-0.7Sn-0.1Fe, suggesting the grain size control may provide an effective way to enhance the creep resistance in Zr alloys for nuclear application.

Effect of characteristics and distribution of Mg_(17)Al_(12)precipitates on tensile and bending properties of high-Al-containing Mg alloys

This study investigates the effect of characteristics and distribution of Mg_(17)Al_(12)precipitates on the uniaxial tensile and three-point bending properties of extruded Mg alloys containing high Al contents.The extruded Mg–9Al–1Zn–0.3Mn(AZ91)alloy contains lamellar-structured Mg_(17)Al_(12)discontinuous precipitates along the grain boundaries,which are formed via static precipitation during natural air cooling.The extruded Mg–11Al–1Zn–0.3Mn(AZ111)alloy contains spherical Mg_(17)Al_(12)precipitates at the grain boundaries and inside the grains,which are formed via dynamic precipitation during extrusion.Due to inhomogeneous distribution of precipitates,the AZ111 alloy consists of two different precipitate regions:precipitate-rich region with numerous precipitates and finer grains and precipitate-scarce region with a few precipitates and coarser grains.The AZ111 alloy exhibits a higher tensile strength than the AZ91 alloy because its smaller grain size and more abundant precipitates result in stronger grain-boundary hardening and precipitation hardening effects,respectively.However,the tensile elongation of the AZ111 alloy is lower than that of the AZ91 alloy because the weak cohesion between the dynamic precipitates and the matrix facilitates the crack initiation and propagation.During bending,a macrocrack initiates on the outer surface of bending specimen in both alloys.The AZ111 alloy exhibits higher bending yield strength and lower failure bending strain than the AZ91 alloy.The bending specimens of the AZ91 alloy have similar bending formability,whereas those of the AZ111 alloy exhibit considerable differences in bending formability and crack propagation behavior,depending on the distribution and number density of precipitates in the specimen.In bending specimens of the AZ111 alloy,it is found that the failure bending strain(ε_(f,bending))is inversely proportional to the area fraction of precipitates in the outer zone of bending specimen(A_(ppt)),with a relationship ofε_(f,bending)=–0.1A_(ppt)+5.86.

Long-term outcome of stem cell transplantation with and without anti-tumor necrotic factor therapy in perianal fistula with Crohn’s disease

BACKGROUND Stem cell transplantation is a promising therapeutic option for curing perianal fistula in Crohn’s disease(CD).Anti-tumor necrotic factor(TNF)therapy combined with drainage procedure is effective as well.However,previous studies are limited to proving whether the combination treatment of biologics and stem cell transplantation improves the effect of fistula closure.AIM This study aimed to evaluate the long-term outcomes of stem cell transplantation and compare Crohn’s perianal fistula(CPF)closure rates after stem cell transplantation with and without anti-TNF therapy,and to identify the factors affecting CPF closure and recurrence.METHODS The patients with CD who underwent stem cell transplantation for treating perianal fistula in our institution between Jun 2014 and December 2022 were enrolled.Clinical data were compared according to anti-TNF therapy and CPF closure.RESULTS A total of 65 patients were included.The median age of females was 26 years(range:21-31)and that of males was 29(44.6%).The mean follow-up duration was 65.88±32.65 months,and complete closure was observed in 50(76.9%)patients.The closure rates were similar after stem cell transplantation with and without anti-TNF therapy(66.7%vs 81.6%at 3 year,P=0.098).The patients with fistula closure had short fistulous tract and infrequent proctitis and anorectal stricture(P=0.027,0.002,and 0.008,respectively).Clinical factors such as complexity,number of fistulas,presence of concurrent abscess,and medication were not significant for closure.The cumulative 1-,2-,and 3-year closure rates were 66.2%,73.8%,and 75.4%,respectively.CONCLUSION Anti-TNF therapy does not increase CPF closure rates in patients with stem cell transplantation.However,both refractory and non-refractory CPF have similar closure rates after additional anti-TNF therapy.Fistulous tract length,proctitis,and anal stricture are risk factors for non-closure in patients with CPF after stem cell transplantation.

BACKGROUND Studies on varicose veins have focused its effects on physical function;however,whether nonsurgical treatments alter muscle oxygenation or physical function remains unclear.Moreover,the differences in such functions between individuals with varicose veins and healthy individuals remain unclear.AIM To investigate changes in physical function and the quality of life(QOL)following nonsurgical treatment of patients with varicose veins and determine the changes in their muscle oxygenation during activity.METHODS We enrolled 37 participants(those with varicose veins,n=17;healthy individuals,n=20).We performed the following measurements pre-and post-nonsurgical treatment in the varicose vein patients and healthy individuals:Calf muscle oxygenation during the two-minute step test,open eyes one-leg stance,30 s sit-to-stand test,visual analog scale(VAS)for pain,Pittsburgh sleep quality index,physical activity assessment,and QOL assessment.RESULTS Varicose veins patients and healthy individuals differ in most variables(physical function,sleep quality,and QOL).Varicose veins patients showed significant differences between pre-and post-nonsurgical treatment—results in the 30 sit-to-stand test[14.41(2.45)to 16.35(4.11),P=0.018],two-minute step test[162.29(25.98)to 170.65(23.80),P=0.037],VAS for pain[5.35(1.90)to 3.88(1.73),P=0.004],and QOL[39.34(19.98)to 26.69(17.02),P=0.005];however,no significant difference was observed for muscle oxygenation.CONCLUSION Nonsurgical treatment improved lower extremity function and QOL in varicose veins patients,bringing their condition close to that of healthy individuals.Future studies should include patients with severe varicose veins requiring surgery to confirm our findings.

Short- and long-term outcomes of surgical treatment in patients with intestinal Behcet’s disease

BACKGROUND Behcet’s disease(BD),a chronic vasculitic disorder affecting multiple organs,is characterized by recurrent oral and genital ulcers,arthritis,vasculitis,and intes-tinal ulcers.Although intestinal involvement of BD is common in East Asia,the efficacy and long-term outcomes of surgical treatment of intestinal BD still remain to be established.AIM To evaluate the postoperative clinical course of intestinal BD and determine factors associated with its recurrence.METHODS Data from patients who underwent surgical treatment for intestinal BD between January 2010 and August 2021 were retrospectively reviewed.Patients’demo-graphics,clinical features,postoperative course,complications,and follow-up data were evaluated.RESULTS We analyzed 39 surgeries in 31 patients.The mean patient age was 45.1 years,and the mean interval between the diagnosis of intestinal BD and surgical treatment was 4.9 years(range 1.0-8.0 years).The most common indication for surgery was medical intractability(n=16,41.0%),followed by fistula or abscess(n=11,28.2%).Laparoscopic approaches were used in 19 patients(48.7%),and 5 patients(12.8%)underwent emergency surgeries.The most common surgical procedure was ileocecal resection(n=18,46.2%),followed by right colectomy(n=11,28.2%).A diverting stoma was created in only one patient(2.6%).During a mean follow-up period of 45(range 8-72)months,eight cases(20.5%)of recurrence in five patients required reoperation.The interval between operations was 12.1 months(range 6.3-17.8 mo).Four patients(10.3%)experienced recurrence within 1 year postoperatively,and all eight recurrences occurred within 2 years of the initial surgery.The reoperation rates at 1 and 3 years were 10.3%and 20.5%,respectively.A redo ileocolic anastomosis was performed in all recurrent cases.In multivariate Cox regression analysis,emergency surgery[hazard ratio(HR)9.357,95%confidence interval(CI):1.608-54.453,P=0.013]and elevated C-reactive protein(CRP)levels(HR 1.154,95%CI:1.002–1.328,P=0.047),but not medication use,were predictors
