AIM To determine whether three-dimensional(3D) reconstruction from conventional magnetic resonance imaging(MRI) is able to accurately detect a meniscal tear, and define the *** Thirty-three patients' 3T MRI scan data were collected and sagittal uni-planar 3D reconstructions performed from the preoperative MRI. There were 24 meniscal tears in 24 patients, and nine controls. All patients had arthroscopic corroboration of MRI findings. Two independent observers prospectively reported on all 33 reconstructions. Meniscal tear presence or absence was noted, and tear configuration subsequently categorised as either radial, bucket-handle, parrot beak, horizontal or *** Identification of control menisci or meniscal tear presence was excellent(Accuracy: observer 1 = 90.9%; observer 2 = 81.8%). This paper is concerned first with the behaviour of differences T（t） - T（s） near the origin, where （T（t）） is a semigroup of operators on a Banach space, defined either on the positive real line or a sector in the right half-plane （in which case it is assumed analytic）. For the non-quasinilpotent case extensions of results in the published literature are provided, with best possible constants; in the case of quasinilpotent semigroups on the half-plane, it is shown that, in general, differences such as T（t） -T（2t） have norm approaching 2 near the origin. The techniques given enable one to derive estimates of other functions of the generator of the semigroup; in particular, conditions are given on the derivatives near the origin to guarantee that the semigroup generates a unital algebra and has bounded generator.

Farnesoid X receptor （FXR） is a ligand-activated nuclear receptor that plays a central role in regulating genes involved in bile acid homeostasis, and fat and glucose metabolism. Here, we demonstrate a post-translational interplay between FXR phosphoryl- ation, SUMOylation, and ubiquitination that directs the receptor into an activation-degradation pathway in hepatocytes. We iden- tify a non-canonical SUMOylation motif termed pSuM that conjugates SUM02 at lys-325 of FXR under the direct control of casein kinase 2 （CK2）, which provides the required negative charge for Ubc9 and PIAS1 to perform SUMOylation, by phosphorylating Ser-327. lys-325 SUMOylation is indispensable to the promotion of efficient ligand activation and transcriptional coactivation of FXR. Constitutive pSuM activation using a phospho-mimic Ser-327 mutant or catalytic CK2 expression strongly induces SUM02 conjugation, which directs FXR ubiquitination and proteasome-dependent degradation. We also determine that such SUMOylation-dependent ubiquitination of FXR is mediated by the E3 ubiquitin ligase RNF4, which is required to achieve maximal induction of FXR and optimal up- or downregulation of responsive genes involved in bile acid homeostasis and liver regeneration. Our findings identify a highly regulated atypical SUMO conjugation motif that serves to coordinate FXR transcriptional compe- tence, thereby expanding the intricate dynamics of the SUMOylation process used by incoming signals to govern metabolic gene regulation.

Inflammatory bowel diseases are characterised by inflammation that compromises the integrity of the epithelial barrier. The intestinal epithelium is not only a static barrier but has evolved complex mechanisms to control and regulate bacterial interactions with the mucosal surface. Apical tight junction proteins are critical in the maintenance of epithelial barrier function and control of paracellular permeability. The characterisation of alterations in tight junction proteins as key players in epithelial barrier function in inflammatory bowel diseases is rapidly enhancing our understanding of critical mechanisms in disease pathogenesis as well as novel therapeutic opportunities. Here we give an overview of recent literature focusing on the role of tight junction proteins, in particular claudins, in inflammatory bowel diseases and inflammatory bowel disease associated colorectal cancer.

Plinian pumice fall from the Holocene eruption of Mount Mazama in the Cascade volcanic arc is an unconfined, perched aquifer in south-central Oregon. The pumice aquifer provides near-surface groundwater storage that maintains biologically diverse wetland environments. Wetland environments reflect post-eruption disruption of the once uniform pumice blanket by fluvial and lacustrine processes operating within the template of the pre-eruption landscape. In the 8.6 km2 Round Meadow watershed the pumice aquifer interacts with a seasonally flooded meadow, fen, springs, and perennial stream. The laterally uniform, isotropic pumice aquifer is disrupted by flat-bottomed ephemeral stream valleys that drain to the seasonally flooded meadow. Surface water levels in the seasonally flooded meadow are controlled by a knickpoint developed on bedrock. The underlying aquifer is confined by a layer of glass-rich diatomaceous silt grading upward to organic-rich silt. Here, the aquifer is comprised of remnants of the pumice deposit, lag sand, and reworked pumice. The water level in the confined aquifer is maintained by recharge from the unconfined pumice aquifer following flow pathways beneath ephemeral stream valleys. The fen is developed on a down-thrown block of welded tuff and pre-eruption diatomaceous silt. Water levels in the fen are sensitive to inter-annual variations in precipitation. low discharge, low temperature (5.0°C to 6.5°C), and low conductivity (30 to 50 μS/cm) springs appear to be fracture controlled and rising through welded tuff. Spring discharge and seepage through pumice from the welded tuff support perennial flow in the creek that also carries discharge from the seasonally flooded meadow when water levels are high enough to cross the knickpoint.
