Control of APN/CD13 and NEP/CD10 on sperm.m.tility

Am.nopeptidase N （APN/CD13） and neutral endopeptidase （NEP/CD10） are enzym.s present in hum.n sperm.cells and involved in regulation of sperm.m.tility of noncapacitated sperm.tozoa. We investigated the involvem.nt of APN/CD 13 and NEP/CD 10 in m.tility and in kinem.tic param.ters of hum.n capacitated sperm.tozoa. Sperm.cells isolated by a discontinuous Percoll gradient （40%-80%） followed up by swim.up techniques were incubated with the APN/CD 13 -specific inhibitor, leuhistin （100 μm.l L^-1）, and the NEP/CD 10-specific inhibitor, thiorphan （1 μm.l L^-1）. The com.lete inhibition of both APN/CD 13 and NEP/CD 10 im.roved sperm.m.tility. Sperm.tozoa incubated with the APN/CD13-specific inhibitor lenhistin showed asym.etrical trajectories, whereas sperm.trajectories were m.re regular after treatm.nt with the NEP/CD 10-specific inhibitor thiorphan. In conclusion, APN/CD 13 and NEP/CD 10 m.dulate the m.tility of capacitated sperm.tozoa, although each of the enzym.s seem. to participate in the control of different aspects of sperm.m.tility. Therefore, both inhibitors m.y be useful for sperm.activation at different functional stages of sperm.tozoa.

Surface-active polym.rs are of substantial im.ortance in m.ny diverse aspects of m.dern technology, with applications ranging from.solid phase chem.cal synthesis related to drug discovery and chem.cal catalysis to biocom.atible/bioactive m.dical im.lants and prostheses, and to surface-m.dified fabrics. Whilst there are a num.er of existing physical (e. g. corona or plasm. discharge, ion beam.irradiation[1] ) and chem.cal (e. ***, oxidation, chlorination, acylation and quaternisation[2-4]) m.thods for the surface m.dification of polym.rs, the frequent requirem.nt for significant infrastructure, harsh reaction conditions, and lim.tation to specific polym.r types (e. g. polybutadiene[5] ), which m.st possess suitable chem.cal functionality capable of direct m.dification, led us to consider alternative chem.cal m.thods. Desirable was an alternative that would be am.nable to a large range of polym.rs, perm.tting direct chem.cal m.dification under m.ld conditions and using inexpensive reagents.

High purity(>99.9% com.osition) nickel m.tal specim.ns were used in electropolishing treatm.nts with an acid-free ionic liquid electrolyte prepared from.quaternary am.onium.salts as a green polishing solution. Voltam.etry and chronoam.erom.try tests were conducted to determ.ne the optim.m.conditions for electrochem.cal polishing. Atom.c force m.croscopy(AFm. revealed nanoscale effectiveness of each polishing treatm.nt. Atom.c force m.croscopy provided an overall observation of the m.terial interface between the treated and unpolished regions. Surface m.rphology com.arisons sum.arized electrochem.cal polishing efficiency by providing root-m.an-square roughness averages before and after electrochem.cal polishing to reveal a m.rror finish six tim.s sm.other than the sam. nickel m.tal surface prior to electropolishing. This transition m.nifested in a m.rked change in root-m.an-squared roughness from.112.58 nm.to 18.64 nm.and producing a sm.oth m.rror finish. Finally, the m.chanism.of the ionic liquid during electropolishing revealed decom.osition of choline in the form.of a transient choline radical by acceptance of an electron from.the nickel-working electrode to decom.ose to trim.thylam.ne and ethanol.

Ten years after the initial generation of induced pluripotent stem.cells(hiPSCs)from.hum.n tissues,their potential is no longer questioned,with over 15000 publications listed on Pubm.d,covering various fields of research;including disease m.deling,cell therapy strategies,pharm.cology/toxicology screening and 3D organoid ***,despite evidences that the presence of m.tations in hiPSCs should be a concern,publications addressing genom.c integrity of these cells represent less than 1%of the *** a first overview of the m.tation types currently reported in hiPSCs,including karyotype abnorm.lities,copy num.er variations,single point m.tation as well as uniparental disom.,this review will discuss the im.act of reprogram.ing param.ters such as starting cell type and reprogram.ing m.thod on the m.intenance of the cellular genom.c ***,a specific focus will be placed on culture conditions and subsequent differentiation protocols and how their m.y also trigger genom.c aberrations within the cell population of ***,in a last section,the im.act of genom.c alterations on the possible usages of hiPSCs and their derivatives will also be exem.lified and *** will also discuss which techniques or com.ination of techniques should be used to screen for genom.c abnorm.lities with a particular focus on the necessary quality controls and the potential alternatives.
