Immunogenic antigens for vaccination are often created through the production of recombinant proteins using Escherichia coli.1 As a side product,aggregates called inclusion bodies(IBs)are formed,containing largely misfolded forms of the overexpressed recombinant protein.2 The effect of IBs on the immune system and whether they can be used as effective vaccine products remain unknown.The non-native conformation of proteins upon accumulation in IBs abrogates their use as vaccines aimed at generating high-affinity antibodies.3 However,IBs exhibit unique properties,including mechanical and thermal stability due to their intrinsic particulate nature,biocompatibility,high antigenic content,low toxicity and relative resistance to proteases.These characteristics make IBs attractive as an antigenic vaccine formulation for T cell responses to linear epitopes.Whether IBs can trigger adaptive cellular responses initiated via uptake by dendritic cells for presentation to T cells is unknown.

Ketodeoxynononic acid(Kdn)is a rather uncommon class of sialic acid in mammals.However,associations have been found between elevated concentrations of free or conjugated Kdn in relation to human cancer progression.Hitherto,there has been a lack of conclusive evidence that Kdn occurs on(specific)human glycoproteins(conjugated Kdn).Here,we report for the first time that Kdn is expressed on prostate-specific antigen(PSA)N-linked glycans derived from human seminal plasma and urine.Interestingly,Kdn was found only in an a2,3-linkage configuration on an antennary galactose,indicating a highly specific biosynthesis.This unusual glycosylation feature was also identified in a urinary PSA cohort in relation to prostate cancer(PCa),although no differences were found between PCa and nonPCa patients.Further research is needed to investigate the occurrence,biosynthesis,biological role,and biomarker potential of both free and conjugated Kdn in humans.

micro-computed tomography can be applied for the assessment of the micro-architectural characteristics of the cortical and trabecular bones in either physiological or disease conditions. However, reports often lack a detailed description of the methodological steps used to analyse these images, such as the volumes of interest, the algorithms used for image filtration, the approach used for image segmentation, and the bone parameters quantified, thereby making it difficult to compare or reproduce the studies. This study addresses this critical need and aims to provide standardized assessment and consistent parameter reporting related to quantitative jawbone image analysis. Various regions of the rat jawbones were screened for their potential for standardized micro-computed tomography analysis. Furthermore, the volumes of interest that were anticipated to be most susceptible to bone structural changes in response to experimental interventions were defined. In the mandible, two volumes of interest were selected, namely, the condyle and the trabecular bone surrounding the three molars. In the maxilla, the maxillary tuberosity region and the inter-radicular septum of the second molar were considered as volumes of interest. The presented protocol provides a standardized and reproducible methodology for the analysis of relevant jawbone volumes of interest and is intended to ensure global, accurate, and consistent reporting of its morphometry. Furthermore, the proposed methodology has potential, as a variety of rodent animal models would benefit from its implementation.

Reactive Extraction of Alcohols from Apolar Hydrocarbons with Aqueous Solutions

The aqueous solutions are evaluated as sustainable reactive extraction solvents for the recovery of monohydroxyl alcohols （benzyl alcohol, 1-hexanol, cyclohexanol） present in few-percent concentrations in apolar hydrocarbons （toluene, n-hexane, and cyclohexane） by considering two approaches. An aqueous solution containing a reactive extractant, like borate salts, borate complexes, a monosalt of dicarboxylic acid, hydroxypropyl-cyclodextrins, and silver nitrate, shows limited potential to be used. Another approach, in which the alcohol is chemically modified prior to the extraction into an easy-extractable form, in this case a monoester/carboxylic acid, shows much more potential. An environmentally benign aqueous solution of sodium hydrogen carbonate can provide a distribution ratio of benzyl alcohol up to 200, leaving the solubility of the organic solvent in the aqueous solution unchanged relative to pure water and therefore increasing the selectivity with two orders of magnitude. The modification of aromatic, cyclo-aliphatic, and linear aliphatic alcohols can be performed efficiently in the apolar organic solvent without need for a catalyst. The recovery of the modified alcohol can be performed by back-extraction in combination with a spontaneous hydrolysis.

Design Safe addresses the challenges of supporting integrative data-driven research in natural hazards engineering.It is an end-to-end data management,communications,and analysis platform where users collect,generate,analyze,curate,and publish large data sets from a variety of sources,including experiments,simulations,field research,and post-disaster reconnaissance.DesignSafe achieves key objectives through:(1)integration with high performance and cloud-computing resources to support the computational needs of the regional risk assessment community;(2)the possibility to curate and publish diverse data structures emphasizing relationships and understandability;and(3)facilitation of real time communications during natural hazards events and disasters for data and information sharing.The resultant services and tools shorten data cycles for resiliency evaluation,risk modeling validation,and forensic studies.This article illustrates salient features of the cyberinfrastructure.It summarizes its design principles,architecture,and functionalities.The focus is on case studies to show the impact of Design Safe on the disaster risk community.The Next Generation Liquefaction project collects and standardizes case histories of earthquake-induced soil liquefaction into a relational database—Design Safe—to permit users to interact with the data.Researchers can correlate in Design Safe building dynamic characteristics based on data from building sensors,with observed damage based on ground motion measurements.Reconnaissance groups upload,curate,and publish wind,seismic,and coastal damage data they gather during field reconnaissance missions,so these datasets are available shortly after a disaster.As a part of the education and community outreach efforts of Design Safe,training materials and collaboration space are also offered to the disaster risk management community.

目的为了解决溶酶体贮积症酶学诊断的标本在常温邮寄又不因外界条件的影响而导致酶的失活这一难题，探讨冷干技术处理标本后溶酶体酶的稳定性。方法建立正常人白细胞、血浆（各４０人份）和３株成纤维细胞标本库。对上述３个标本库分别进行了２０种、１４种和７种酶酶活性测定。比较了冷干后３种不同条件模拟邮寄与常规处理标本共４组酶活性的情况。结果经Ｆ检验：白细胞和成纤维细胞库４组之间酶活性无显著性差异（Ｐ＞０．０５）。血浆库４组之间酶活性有非常显著性差异（Ｐ＜０．０１）。经ｔ检验：白细胞和成纤维细胞库未冷干组与冷干后置室温３周组无显著性差异（Ｐ＞０．０５）。血浆库未冷干组与３周组有非常显著性差异（Ｐ＜０．０１）。
