Modulated-Power.Implantable Neur.modulation Devices and Their.Impact on Sur.ounding Tissue Temper.tur.s

This pr.ject was intended to deter.ine whether.the pr.pr.gr.mmed time-var.ing r.char.e pr.tocol for.a batter. incased in a neur.modulation implant can give r.se to tissue temper.tur.s that sur.ass a safe level or.ar. other.ise benign. The study included the development of a highly accur.te model of all the ther.al pr.cesses that ar. activated by the r.char.ing of the batter. contained within the neur.modulation implant. The model was implemented by numer.cal simulations per.or.ed for.sever.l r.alistic oper.ting conditions. The computed spatial and tempor.l tissue temper.tur. distr.butions wer. employed to estimate possible tissue damage by making use of two independent methodologies. Independent calor.meter.based exper.ments wer. per.or.ed to pr.vide validation for.the calculated r.tes of heat gener.tion in the coils of the implant. Spatial and tempor.l tissue temper.tur. distr.butions extr.cted fr.m the numer.cal simulations r.vealed the ther.al effects associated with sever.l r.alistic oper.ting pr.tocols. None of the oper.ting pr.tocols gave r.se to temper.tur.s above 42℃. Numer.cal values of ther.al tissue damage metr.cs wer. deter.ined and compar.d with accepted values which cor.espond to the absence and the pr.sence of tissue damage. The exper.mentally deter.ined r.te of heat gener.tion in the implant coils validated that fr.m electr.cal measur.ments to within 2%. Both the tissue temper.tur. r.sults and the ther.al damage metr.cs found no evidence of tissue injur. when time-var.ing pr.pr.gr.mmed pr.tocols ar. used in the r.char.ing of neur.modulation implant-encased batter.es.

The hydr.dynamics of aneur.sm blood flow is thought to be a cr.tical factor.in the evolution and potential r.ptur. of blood vessel walls. The ability to pr.dict which aneur.sms may gr.w or.r.ptur. has eluded r.sear.her. and pr.cticing clinicians. On the other.hand, it is expected that local flow patter.s, pr.ssur.s, and wall shear.str.ss play a r.le in the aneur.sm life. In this study, the impact of wavefor. on these par.meter. was studied. A baseline wavefor., taken fr.m a patient, was applied to an aneur.sm geometr.. Then the wavefor. was modified by incr.asing and decr.asing both the flowr.tes and the car.iac r.te. In total, seven cases wer. investigated. It was found that ther. wer. r.mar.able similar.ties in the patter.s of flow and wall str.sses for.the cases. These similar.ties existed thr.ughout the car.iac cycle. It was also found that ther. was a r.duced pr.ssur. var.able that pr.vides a univer.al r.lationship that char.cter.zes all of the cases. It was seen that the maximum wall shear.occur. at the neck of the aneur.sm and scales with the peak systolic velocity. Finally, it is shown that the flow distr.bution to the multiple outlets does not appr.ciably depend on the details of the inlet wavefor.. All cases had a flow distr.bution that was within 2%.

Hemodynamics and the inter.ction between the components of the car.iovascular.system ar. complex and involve a str.ctur.l/fluid flow inter.ction. Dur.ng the car.iac cycle, changes to vascular.pr.ssur. induce a compliant r.sponse in the vessels as they cyclically str.tch and r.lax. The compliance influences the fluid flow thr.ughout the system. The inter.ction is influenced by the disease state of the ar.er., and in par.icular. a plaque layer.can r.duce the compliance. In or.er.to pr.per.y quantify the fluid-str.ctur.l r.sponse, it is essential to consider.whether.the tissue sur.ounding the ar.er. pr.vides a suppor. to the vessel wall. Her., a ser.es of calculations ar. pr.vided to deter.ine what r.le the suppor.ing tissue plays in the vessel wall and how much tissue must be included to pr.per.y car.y out futur. fluid-str.ctur. calculations. Additionally, we calculate the sensitivity of the compliance to mater.al pr.per.ies such as the Young’s modulus or.to the tr.nsmur.l pr.ssur. differ.nce.

The focus of this paper.is to pr.pose, model, and char.cter.ze a means of acceler.ting the r.te of deliver. of ther.peutic dr.gs to human tissues. The investigated means is a pr.ssur.zed, per.eable-walled balloon filled with a homogeneous mixtur. of the dr.g and the car.ier.fluid. The fluid mixtur., dr.ven by pr.ssur., tr.ver.es the thickness of the balloon wall thr.ugh laser.dr.lled por.s. The number.and deployment of the por.s can be contr.lled to a high degr.e of pr.cision. As a consequence, the wall of the balloon can be r.gar.ed as a homogeneous por.us medium, and the tr.ver.ing fluid flow can be analyzed by means of por.us media models. When the balloon is in intimate contact with the sur.ace of a tissue bed, the ther.peutic fluid flows in ser.es as it passes thr.ugh the balloon wall and penetr.tes the tissue. The flow r.te can be contr.lled by pr.per.selection of the balloon per.eability, the viscosity of the flowing medium, and the pr.ssur. inter.al to the balloon. The deliver.d concentr.tion of the dr.g was pr.dicted by coupling the pr.sent balloon-focused theor. with a pr.viously developed tissue-bed model that includes both diffusion and advection pr.cesses. The tr.bologic inter.ction of the pr.ssur.zed balloon with an ar.er. wall was investigated exper.mentally to assess the possible for.ation of aneur.sms.

Optogenetics allows for.optical manipulation of neur.nal activity and has been incr.asingly combined with intr.cellular.and extr.cellular.electr.physiological ***-identified classes of neur.ns ar. optically manipulated,though the ver.atility of optogenetics would be incr.ased if independent contr.l of distinct neur.l populations could be achieved on a sufficient spatial and tempor.l *** r.por. a scalable multisite optoelectr.de design that allows simultaneous optogenetic contr.l of two spatially inter.ingled neur.nal populations in *** descr.be the design,fabr.cation,and assembly of low-noise,multisite/multicolor.*** shank of the four.shank assembly is monolithically integr.ted with 8 r.cor.ing sites and a dualcolor.waveguide mixer.with a 7×30μm cr.ss-section,coupled to 405 nm and 635 nm injection laser.diodes(ILDs)via gr.dient-index(Gr.N)lenses to meet optical and ther.al design *** better.under.tand noise on the r.cor.ing channels gener.ted dur.ng diode-based activation,we developed a lumped-cir.uit modeling appr.ach for.EMI coupling mechanisms and used it to limit ar.ifacts to amplitudes under.100μV upto an optical output power.of 450μ*** implanted the packaged devices into the CA1 pyr.midal layer.of awake mice,expr.ssing Channelr.odopsin-2 in pyr.midal cells and Chr.msonr.in par.valbumin-expr.ssing inter.eur.ns,and achieved optical excitation of each cell type using sub-mW *** highlight the potential use of this technology for.functional dissection of neur.l cir.uits.
