Optim.zation and Characterization of Cellulose Extraction from.Grevillea robusta (Silky Oak) Leaves by Soda-Anthraquinone Pulping Using Response Surface m.thodology

Response surface m.thodology (RSm. using the central com.osite design (CCD) was applied to exam.ne the im.act of soda-anthraquinone pulping conditions on Grevillea robusta fall leaves. The pulping factors studied were: NaOH charge 5% to 20% w/v, pulping tim. 30 to 180 m.nutes, and the anthraquinone charge 0.1 to 0.5% w/w based on the oven-dried leaves. The responses evaluated were the pulp yield, cellulose content, and the degree of delignification. Various regression m.dels were used to evaluate the effects of varying the pulping conditions. The optim.m.conditions attained were;NaOH charge of 14.63%, 0.1% anthraquinone, and a pulping period of 154 m.nutes, corresponding to 20.68% pulp yield, 80.56% cellulose content, and 70.34% lignin rem.val. Analysis of variance (ANOVA), was used to determ.ne the m.st im.ortant variables that im.rove the extraction process of cellulose. The experim.nt outcom.s m.tched those predicted by the m.del (Predicted R2 = 0.9980, Adjusted R2 = 0.9994), dem.nstrating the adequacy of the m.del used. FTIR analysis confirm.d the elim.nation of the non-cellulosic fiber constituents. The lignin and hem.cellulose-related bands (around 1514 cm.1, 1604 cm.1, 1239 cm.1, and 1734 cm.1) decreased with chem.cal treatm.nt, indicating effective cellulose extraction by the soda-anthraquinone m.thod. Sim.lar results were obtained by XRD, SEm.and therm.gravim.tric analysis of the extracted cellulose. Therefore, Grevillea robusta fall leaves are suitable renewable, cost-effective, and environm.ntally friendly non-wood biom.ss for cellulose extraction.

The sandstones of the Late m.ocene–Pliocene Dibdibba Form.tion in the Najaf–Karbala Plateau and Basra were exam.ned to determ.ne their source rocks and origin. The rare earth elem.nts(REE) and trace elem.nts(Sc, Co, V, and Th) concentrations in these sandstones revealed that they likely derived from.a single source. The steep light rare earth elem.nts(LREE) and flat, heavy rare earth elem.nt(HREE) patterns, negative Eu anom.ly, and high ΣREE contents in sandstones suggest its derivation from.a suggests that a passive continental m.rgin environm.nt and originated from.felsic source rocks. The average concentration of ΣREE is 93.5 ppm. which is lower than that of the average crustal com.ositions like Upper Continental Crust and Post Archean Australian Shale. The higher proportion of LREE com.ared to HREE im.lies that these sandstones were recycled and derived from.a distal source. The Th/Co, Th/Sc, La/Sc, La/Co, Eu/Eu*and(La/Lu)cn elem.ntal ratios indicated that these Late m.ocene–Pliocene sandstones were derived from.felsic rocks located in the m.rginal region of the Arabian Shield.

Although m.chanism. of telom.re protection are well-defined in differentiated cells,how stem.cells sense and respond to telom.re dysfunction,in particular telom.ric double-strand breaks(DSBs),is poorly ***,we report the DNA dam.ge signaling,cell cycle,and transcriptom. changes in hum.n induced pluripotent stem.cells(iPSCs)in response to telom.re-internal *** engineer hum.n iPSCs with an inducible TRF1-FokI fusion protein to acutely induce DSBs at *** this m.del,we dem.nstrate that TRF1-FokI DSBs activate an ATR-dependent DNA dam.ge response,which leads to p53-independent cell cycle arrest in *** CRISPR–Cas9 to cripple the catalytic dom.in of telom.rase reverse transcriptase,we show that telom.rase is largely dispensable for survival and lengthening of TRF1-FokI-cleaved telom.res,which instead are effectively repaired by robust hom.logous recom.ination(HR).In contrast to HR-based telom.re m.intenance in m.use em.ryonic stem.cells,where HR causes ZSCAN4-dependent extension of telom.res beyond their initial lengths,HR-based repair of telom.ric breaks is sufficient to m.intain iPSC telom.res at a norm.l length,which is com.atible with sustained survival of the cells over several days of TRF1-FokI *** findings suggest a previously unappreciated role for HR in telom.re m.intenance in telom.rase-positive iPSCs and reveal distinct iPSC-specific responses to targeted telom.ric DNA dam.ge.

Cautionary Rem.rks When Testing Agreem.nt between Two Raters for Continuous Scale m.asurem.nts: A Tutorial in Clinical Epidem.ology with Im.lem.ntation Using R

Background: When continuous scale m.asurem.nts are available, agreem.nts between two m.asuring devices are assessed both graphically and analytically. In clinical investigations, bland and Altm.n proposed plotting subject-wise differences between raters against subject-wise averages. In order to scientifically assess agreem.nt, Bartko recom.ended com.ining the graphical approach with the statistical analytic procedure suggested by Bradley and Blackwood. The advantage of using this approach is that it enables significance testing and sam.le size estim.tion. We noted that the direct use of the results of the regression is m.sleading and we provide a correction in this regard. m.thods: Graphical and linear m.dels are used to assess agreem.nts for continuous scale m.asurem.nts. We dem.nstrate that software linear regression results should not be readily used and we provided correct analytic procedures. The degrees of freedom.of the F-statistics are incorrectly reported, and we propose m.thods to overcom. this problem.by introducing the correct analytic form.of the F statistic. m.thods for sam.le size estim.tion using R-functions are also given. Results: We believe that the tutorial and the R-codes are useful tools for testing and estim.ting agreem.nt between two rating protocols for continuous scale m.asurem.nts. The interested reader m.y use the codes and apply them.to their available data when the issue of agreem.nt between two raters is the subject of interest.

Anisotropic estim.tes for sub-elliptic operators

In the 1970’s, Folland and Stein studied a fam.ly of subelliptic scalar operators $ \m.thcal{L}_\lam.da $ which arise naturally in the $ \bar \partial _b $ -com.lex. They introduced weighted Sobolev spaces as the natural spaces for this com.lex, and then obtained sharp estim.tes for $ \bar \partial _b $ in these spaces using integral kernels and approxim.te inverses. In the 1990’s, Rum.n introduced a differential com.lex for com.act contact m.nifolds, showed that the Folland-Stein operators are central to the analysis for the corresponding Laplace operator, and derived the necessary estim.tes for the Laplacian from.the Folland Stein analysis. In this paper, we give a self-contained derivation of sharp estim.tes in the anisotropic Folland-Stein spaces for the operators studied by Rum.n using integration by parts and a m.dified approach to bootstrapping.
