Background： This experiment was conducted to test the hypothesis that vitamin e（Vit e） and acetylsalicylic acid（ASA）, a cyclooxygenase-2（COX-2） inhibitor, will additively reduce the production of the immunosuppressive molecule prostaglandin e_2（PGe_2） and hence reduce inflammatory responses in weaner pigs experimentally infected with an enterotoxigenic strain of e. ***： The experiment was conducted in a research facility with 192 individually-housed male weaner pigs（Landrace × Large White） weighing 6.6 ± 0.04 kg（mean ± SeM）. The pigs were experimentally infected with an enterotoxigenic strain of e. coli and were allocated to a 2 × 3 factorial design with the respective factors being without and with 125 ppm ASA and three levels of Vit e supplementation（50, 100 or 200 IU/kg diet, dl-α-tocopheryl acetate）.Results： Acetylsalicylic acid supplementation improved average daily gain（P 〈 0.05） and tended to improve feed：gain ratio（P 〈 0.10） during the first 14 d after weaning. Acetylsalicylic acid supplementation also improved（P 〈 0.001） amino acid utilization efficiency（as assessed by plasma urea level） and tended to decrease（P 〈 0.10） PGe2 production in the liver without affecting smal intestinal histology and tight junction protein mR NA expression in the jejunal epithelium. Vitamin e supplementation greater than 100 IU/kg diet sustained both the plasma Vit e concentration（P 〈 0.001） and plasma haptoglobin content（P 〈 0.001） after weaning. However, there was no additive effects of the combined supplementation of ASA and Vit e on performance, intestinal barrier function and inflammatory responses of weaned ***： Although ASA and vitamin e improved amino acid utilization efficiency and reduced acute inflammatory responses, ASA and vitamin e did not additively reduce production of PGe2 and inflammatory responses in weaner pigs experimental y infected with an enterotoxigenic strain of e. coli.

AIM: To establish stable tetracycline-inducible pancreatic cancer cell lines. MeTHODS: Suit-2, MiaPaca-2, and Panc-1 cells were transfected with a second generation reverse tetracycline-controlled transactivator protein (rtTA2S-M2), under the control of either a cytomegalovirus (CMV) or a chicken β-actin promoter, and the resulting clones were ***: Use of the chicken (β-actin) promoter proved superior for both the production and maintenance of doxycycline-inducible cell lines. The system proved versatile, enabling transient inducible expression of a variety of genes, including GST-P, CYP2e1, S100A6, and the actin capping protein, CapG. To determine the physiological utility of this system in pancreatic cancer cells, stable inducible CapG expressors were established. Overexpressed CapG was localised to the cytoplasm and the nuclear membrane, but was not observed in the nucleus. High CapG levels were associated with enhanced motility, but not with changes to the cell cycle, or cellular proliferation. In CapG-overexpressing cells, the levels and phosphorylation status of other actin-moduating proteins (Cofilin and ezrin/Radixin) were not altered. However, preliminary analyses suggest that the levels of other cellular proteins, such as ornithine aminotransferase and enolase, are altered upon CapG induction. CONCLUSION: We have generated pancreatic-cancer derived cell lines in which gene expression is fully controllable.

Phototropins （phot1 and phot2） are blue-light receptor kinases controlling a range of responses that optimize the photosynthetic efficiency of plants. Light sensing is mediated by two flavin-binding motifs, known as LOVl and LOV2, located within the N-terminal region of the protein. Photoexcitation via LOV2 leads to activation of the C-terminal kinase domain and consequently receptor autophosphorylation. However, knowledge of the in-vivo phosphorylation sites for Arabidopsis phototropins is lacking and has impeded progress in elucidating the functional significance of receptor phos- phorylation. We have purified photl from Arabidopsis and identified the in-vivo sites of receptor phosphorylation by liquid chromatography tandem mass spectrometry. Arabidopsis-derived photl binds flavin mononucleotide as chromophore and is phosphorylated at four major sites located upstream of LOV2 （Ser^58, Ser^85, Ser^350, and Ser^410）, three of which are induced by blue light. Nevertheless, structure-function analysis indicates that the biological activity of photl can be attributed to a modular unit comprising the LOV2-kinase region of the protein. Thus, peptide regions upstream of LOV2, including the sites of receptor phosphorylation identified here, do not appear to be important for receptor signaling. By contrast, these regions may be necessary for maximizing stomatal performance and possibly light-induced relocalization of photl.

Numerous clinical and research applications necessitate the ability to interface with peripheral nerve fibers to read and control relevant neural pathways. Visceral organ modulation and rehabilitative prosthesis are two areas which could benefit greatly from improved neural interfacing approaches. Therapeutic neural interfacing, or ‘bioelectronic medicine’, has potential to affect a broad range of disorders given that all the major organs of the viscera are neurally innervated. However, a better understanding of the neural pathways that underlie function and a means to precisely interface with these fibers are required. existing peripheral nerve interfaces, consisting primarily of electrode-based designs, are unsuited for highly specific （individual axon） communication and/or are invasive to the tissue. Our laboratory has explored an optogenetic approach by which optically sensitive reporters and actuators are targeted to specific cell （axon） types. The nature of such an approach is laid out in this short perspective, along with associated technologies and challenges.

evolution in geoscientific data provides the mineral industry with new opportunities.A direction of geochemical data generation evolution is towards big data to meet the demands of data-driven usage scenarios that rely on data *** direction is more significant where traditional geochemical data are not ideal,which is the case for evaluating unconventional resources,such as tailing storage facilities(TSFs),because they are not static due to sedimentation,compaction and changes associated with hydrospheric and lithospheric processes(e.g.,erosion,saltation and mobility of chemical constituents).In this paper,we generate big secondary geochemical data derived from Sentinel-2 satellite-remote sensing data to showcase the benefits of big geochemical data using TSFs from the Witwatersrand Basin(South Africa).Using spatially fused remote sensing and legacy geochemical data on the Dump 20 TSF,we trained a machine learning model to predict in-situ gold ***,we deployed the model to the Lindum TSF,which is 3 km away,over a period of a few years(2015-2019).We were able to visualize and analyze the temporal variation in the spatial distributions of the gold grade of the Lindum ***,we were able to infer extraction sequencing(to the resolution of the data),acid mine drainage formation and seasonal *** findings suggest that dynamic mineral resource models and live geochemical monitoring(e.g.,of elemental mobility and structural changes)are possible without additional physical sampling.

针对大变形非线性结构拓扑优化问题,提出了基于混合细胞自动机(HCA,Hybrid CellularAutomata)多空间域连续体结构拓扑优化方法;采用密度法,建立了单元相对密度表示的材料弹-塑性模型;以单元相对密度和应变能作为细胞自动机(CA,Cellular Automata)的状态信息,利用CA局部控制规则,修改相对密度,迭代实现各设计空间域应变能均匀分布;设计了多空间域拓扑优化HCA算法,采取多个对象同时耦合优化迭代,各自收敛策略,解决了多空间域优化迭代算法收敛稳定性问题;最后,以汽车保险杆结构横梁和支撑等两个设计空间为例,施加大变形动态载荷作用,对提出的多空间结构优化算法进行了验证,优化后结构有效地降低了碰撞作用力峰值达54%,提高了结构安全性.

Tree peony is a unique traditional f lower in China,with large,fragrant,and colorful f ***,a relatively short and concentrated f lowering period limits the applications and production of tree peony.A genome-wide association study(GWAS)was conducted to accelerate molecular breeding for the improvement of f lowering phenology traits and ornamental phenotypes in tree peony.A diverse panel of 451 tree peony accessions was phenotyped for 23 f lowering phenology traits and 4 f loral agronomic traits over 3 *** by sequencing(GBS)was used to obtain a large number of genome-wide single-nucleotide polymorphisms(SNPs)(107050)for the panel genotypes,and 1047 candidate genes were identified by association ***-two related genes were observed during at least 2 years for f lowering,and seven SNPs repeatedly identified for multiple f lowering phenology traits over multiple years were highly significantly associated with five genes known to regulate f lowering *** validated the temporal expression profiles of these candidate genes and highlighted their possible roles in the regulation of f lower bud differentiation and f lowering time in tree *** study shows that GWAS based on GBS can be used to identify the genetic determinants of complex traits in tree *** results expand our understanding of f lowering time control in perennial woody *** of markers closely related to these f lowering phenology traits can be used in tree peony breeding programs for important agronomic traits.

Symptoms of gastroenteropancreatic located neuroendocrine neoplasms(GePNeNs) are often related to food intake and manifest as abdominal pain or diarrhoea which can influence patients nutritional status. Malnutrition is common in cancer patients and influences quality of life, treatment options and survival but is also present in up to 40% of patients with GeP-NeNs. As part of malnutrition there are often deficiencies in fat-soluble vitamins, mainly vitamin D. Little knowledge exists on trace elements. Several factors influence the development of malnutrition such as size and localisation of the primary tumour as well as metastases, side effects from treatment but also hormone production of the tumour itself. One of the main influencing factors leading to malnutrition is diarrhoea which leads to dehydration and electrolyte disturbances. Treatment of diarrhoea should be guided by its cause. Screening for malnutrition should be part of routine care in every GeP-NeN patient. Multidisciplinary treatment including dietician support is necessary for all malnourished patients with GeP-NeNs.

endoscopic retrograde cholangiopancreatography (eRCP) has evolved from a largely diagnostic to a largely therapeutic modality. Cross-sectional imaging, such as computed tomography (CT) and magnetic resonance imaging (MRI), and less invasive endoscopy, especially endoscopic ultrasound (eUS), have largely taken over from eRCP for diagnosis. However, eRCP remains the "first line" therapeutic tool in the management of mechanical causes of acute recurrent pancreatitis, including bile duct stones (choledocholithiasis), ampullary masses (benign and malignant), congenital variants of biliary and pancreatic anatomy (e.g. pancreas divisum, choledochoceles), sphincter of Oddi dysfunction (SOD), pancreatic stones and strictures, and parasitic disorders involving the biliary tree and/or pancreatic duct (e.g Ascariasis, Clonorchiasis).
