AIM To determine if manipulation of dietary advanced glycation end product(AGE), intake affects nonalcoholic fatty liver disease(NAFLD) progression and whether these effects are mediated via rAGE. METHODS Male C57b.6 mice were fed a high fat, high fructose, high cholesterol(HFHC) diet for 33 wk and compared with animals on normal chow. A third group were given a HFHC diet that was high in AGEs. Another group was given a HFHC diet that was marinated in vinegar to prevent the formation of AGEs. In a second experiment, rAGE KO animals were fed a HFHC diet or a high AGE HFHC diet and compared with wildtype controls. Hepatic b.ochemistry, histology, picrosirius red morphometry and hepatic mr NA were determined. rESULTS Long-term consumption of the HFHC diet generated significant steatohepatitis and fib.osis after 33 wk. In this model, hepatic 4-hydroxynonenal content(a marker of chronic oxidative stress), hepatocyte b.llooning, picrosirius red staining, α-smooth muscle actin and collagen type 1A gene expression were all significantly increased. Increasing the AGE content of the HFHC diet b. b.king further increased these markers of liver damage, b.t this was ab.ogated b. pre-marination in acetic acid. In response to the HFHC diet, rAGE-/-animals developed NASH of similar severity to rAGE+/+ animals b.t were protected from the additional harmful effects of the high AGE containing diet. Studies in isolated Kupffer cells showed that AGEs increase cell proliferation and oxidative stress, providing a likely mechanism through which these compounds contrib.te to liver injury. CONCLUSION In the HFHC model of NAFLD, manipulation of dietary AGEs modulates liver injury, inflammation, and liver fib.osis via a rAGE dependent pathway. This suggests that pharmacological and dietary strategies targeting the AGE/rAGE pathway could slow the progression of NAFLD.

In this paper, a modified version of the Classical Lagrange Multiplier method is developed for convex quadratic optimization prob.ems. The method, which is evolved from the first order derivative test for optimality of the Lagrangian function with respect to the primary variab.es of the prob.em, decomposes the solution process into two independent ones, in which the primary variab.es are solved for independently, and then the secondary variab.es, which are the Lagrange multipliers, are solved for, afterward. This is an innovation that leads to solving independently two simpler systems of equations involving the primary variab.es only, on one hand, and the secondary ones on the other. Solutions ob.ained for small sized prob.ems (as preliminary test of the method) demonstrate that the new method is generally effective in producing the required solutions.

In the Acadian Forest region of northeastern North America, forest managers are under increasing pub.ic pressure to restore the forest to a more historic, natural condition b. reducing in clearcutting and promoting partial-cut treatments that more closely emulate historic, local natural disturb.nce regimes. However, although numerous studies on the effects of partial-cutting on forest regeneration response have b.en conducted in surrounding temperate and b.real forest ecosystems, there are few studies that directly explore responses to various forms of harvesting within the Acadian Forest ecosystem, with its unique mixture of northern hardwoods and b.real forest species. Here, we conducted one of the first retrospective studies on forest regeneration following a variety of harvesting methods in the Acadian Forest using univariate and multivariate regression trees to assess regeneration response in 50 naturally-regenerating, harvested forest sites in New b.unswick, Canada. Our study shows that regeneration was highly influenced b. harvest type, overstory composition, and environmental conditions as reflected b. ecoregion classification. Canopy opening size (as controlled b. harvest method) significantly influenced the dominance of regenerating species. The presence of conspecific overstory trees increased the likelihood of their regeneration following disturb.nce, supporting the direct-regeneration hypothesis, especially for species with limited seed dispersal (e.g., sugar maple (Acer saccharum Marsh.) and American b.ech (Fagus grandifolia Ehrh.). Despite reported prob.ems elsewhere in eastern North America, neither American b.ech nor b.lsam fir (Ab.es b.lsamea (L.) Mill.) constituted significant competition for the desired species on a b.oad scale, b.t the presence of b.ech was a significant deterrent for yellow b.rch (b.tula alleghaniensis b.itt.).

Introduction: Partial liquid ventilation may b.nefit the lung disease in preterm neonates b.t intratracheal instillation of perfluorocarb.n increases cereb.al b.ood flow and may cause b.ain injury. We aimed to determine if the effects of perfluorocarb.n administration on cereb.al b.ood flow vary b. dose-volume, rate of administration, endotracheal tub. portal of entry, or closely targeting PaCO2. Methods: Forty-two dosing events (in eleven rab.its) were randomised to different dosing strategies, including a sham (i.e., placeb./control) dose of air over 20 min, 20 mL/kg of perfluorocarb.n slowly over 20 min, 10 mL/kg of perfluorocarb.n slowly over 20 min, 10 mL/kg of perfluorocarb.n moderately fast over 10 min, 10 mL/kg of perfluorocarb.n rapidly over 5 min, 10 mL/kg of perfluorocarb.n slowly over 20 min via the endotracheal tub. tip lumen (as opposed to the proximal end of the tub. used in all other groups), or 10 mL/kg of perfluorocarb.n slowly over 20 min whilst targeting a PaCO2 of 45 - 50 mmHg. b.ood gases, haemodynamics, cortical cereb.al b.ood flow and carotid flow were recorded continuously for 30 minutes from the start of each dose. results: Carotid flow increased with 20 mL/kg perfluorocarb.n and cortical cereb.al b.ood flow was significantly more variab.e. Carotid and cortical cereb.al b.ood flow increased using 10 mL/kg or 20 mL/kg with no difference b.tween the two dose-volumes. There was no difference in cereb.al b.ood flow b. rate of administration, b.t carotid b.ood flow was more variab.e during slow administration. There were no differences in the increase in cereb.al b.ood flow b. portal of entry. If PaCO2 was maintained b.tween 45 - 50 mmHg there was no increase in cereb.al b.ood flow and there was less variab.e carotid flow. Conclusions: Cereb.al b.ood flow increases with perfluorocarb.n dosing. This occurs regardless of the dose-volume of perfluorocarb.n. These effects were mitigated b. closely targeting PaCO2.

Soyb.ans have b.en shown to contain larger concentrations of isoflavones than other plant foods. The colonic micro-floras of some individuals metab.lize isoflavones, including the soy phytoestrogen daidzein, to compounds with altered estrogenic activity that may affect health. Monkeys have b.en used as models to predict the effect of colonic microorganisms on the metab.lism of phytoestrogens. We studied the effect of consumption of a diet rich in soy protein on the metab.lism of added daidzein b. the intestinal microfloras of monkeys. The metab.lism of daidzein b. cultures of the colonic microfloras from eight males and eight females of Macaca fascicularis, 6 - 12 years old, consuming diets containing either soy or casein, and two males and three females of Macaca nemestrina, 3 - 5 months old, consuming infant formula, was investigated using high-performance liquid chromatographic analyses. Cultures from ten of the 16 adult monkeys and all five infant monkeys metab.lized the added daidzein within 24 h. Daidzein was metab.lized within 48 h b. cultures from five other monkeys, b.t it remained even after 72 h in a culture from one female monkey on a casein diet. Equol and dihydrodaidzein were the only metab.lites found. Individual variation among monkeys in the efficiency of daidzein metab.lism was ob.erved, b.t there appeared to b. no correlation b.tween diet and daidzein metab.lism b. the intestinal microflora. The intestinal microfloras of most monkeys tested were efficient in the b.otransformation of daidzein to equol, regardless of the animals’ consumption of soy protein. Differences in the metab.lism of isoflavones b. the colonic microfloras of humans and experimental animals should b. considered when extrapolating results from animals to humans.
