Wind is a climat. variable wit. major impact. on humans, ecosyst.ms and infrast.uct.re, especially in coast.l regions wit. cold climat.s. Climat.-relat.d changes in high-wind event. t.erefore have major implicat.ons for high-lat.t.de resident., yet.t.ere has heret.fore been no syst.mat.c evaluat.on of such changes in a framework spanning hist.rical and fut.re t.meframes. In t.is st.dy, hourly winds from surface st.t.on report. and from dynamical downscaling of winds simulat.d by t.o different.global climat. models have been synt.esized int. hist.rical and fut.re wind climat.logies for Alaska. Quant.le mapping procedures are used t. calibrat. wind simulat.ons driven by an at.ospheric reanalysis, and t.e calibrat.d winds are t.en used t. bias-adjust.t.e full dist.ibut.ons of hist.rical and fut.re winds downscaled from t.e global climat. models. In t.e result.ng climat.logies, winds are generally st.onger at.coast.l and offshore (island) locat.ons t.an at.int.rior sit.s, where calm condit.ons are frequent.in wint.r. t.e season of peak wind speed varies from wint.r in t.e coast.l and offshore locat.ons t. summer in int.rior areas. High-wind event. det.rmined from t.e hourly dat. are most.frequent.during wint.r at.coast.l locat.ons. Project.d changes for t.e lat. 21st.cent.ry are st.t.st.cally significant.at.many locat.ons, and t.ey show a qualit.t.vely similar seasonalit. in t.e out.ut.from t.e t.o models: an increase of mean wind speeds in t.e cold season and a decrease of mean wind speeds in t.e warm season. High-wind event. are project.d by bot. models t. become more frequent.in t.e nort.ern and west.rn Alaska coast.l regions, which are precisely t.e regions in which t.e prot.ct.ve sea ice cover has decreased (and is project.d t. decrease furt.er), point.ng t. increased risks of coast.l flooding and erosion.

t.e major seed st.rage prot.in phaseolin of common bean (Phaseolus vulgaris L.) is deficient.in met.ionine, an essent.al amino acid for human and animal healt.. t. improve t.e nut.it.onal qualit. of common bean, we designed met.ionine enhancement.of phaseolin based on t.e t.ree-dimensional st.uct.re of prot.in, de novo design principles and genet.c informat.on. Amino acid subst.t.t.on and loop insert.on were t.rget.d t. t.e int.rior and ext.rior, respect.vely, of t.e prot.in’s β-barrels. First. we int.oduced t.e met.ionine enhancement.mut.t.ons int. phaseolin cDNA, expressed cDNA in Escherichia coli and purified monomeric non-glycosylat.d prot.ins. Biophysical analysis of E. coli-expressed prot.ins demonst.at.d a similar st.uct.ral st.bilit. of wild-t.pe and mut.nt.phaseolin monomers. Here, we at.empt.d t. t.st.t.e st.uct.ral st.bilit. of t.e met.ionine-enhanced phaseolin by int.oducing phaseolin cDNA t. t.bacco via Agrobact.rium t.mefaciens-mediat.d t.ansformat.on of leaf disks, regenerat.ng t.ansgenic t.bacco plant., and examining t.e accumulat.on of phaseolin prot.in in mat.re t.ansgenic t.bacco seeds. We used seven const.uct. cont.ining different.ext.nt. of met.ionine enhancement. ranging from t.e original 3 t. maximum 33 met.ionines per 397 amino acid residues. ELISA analyses indicat.d t.at.t.e met.ionine-enhanced phaseolins did not.accumulat. as st.bly in mat.re t.ansgenic t.bacco seeds as t.e wild-t.pe phaseolin. It.seems likely t.at.t.e met.ionine-enhanced phaseolin prot.ins were under t.e st.ingent.scrut.ny of t.e prot.in qualit. cont.ol mechanism in t.e endoplasmic ret.culum (ER), Golgi complex and/or vacuolar prot.in bodies. t.e prot.in degradat.on is probably t. occur in t.e vacuolar prot.in bodies due t. t.e inst.bilit. of t.e t.imer assembly caused by t.e met.ionine enhancement.mut.t.ons t.rget.ng eit.er amino-acids subst.t.t.ons or/and loop insert.ons t. t.e int.rior β-sheet. and t.m/loop regions, respect.vely, of N- and C-barrel st.uct.res.
