Str.mules a.0} highly dyna.ic str.ma.filled tubules extending fr.m the sur.a.e of pla.tids a.d occa.iona.ly inter.onnecting individua. pla.tids, a.lowing the movement of complex biologica. molecules between the inter.onnected pla.tids. Exper.ments with inhibitor. of cytoskeleton a.sembly ha.e indica.ed the involvement of a. a.tin-ba.ed system in str.mule movement. However. the motor.pr.tein a.socia.ed with the system ha. not been identified. Her., we pr.sent dir.ct evidence tha. myosin XI is involved in the for.a.ion a.d movement of str.mules in toba.co lea.es. a.plica.ion of 2,3-buta.edione 2-monoxime, a. inhibitor.of myosin a.Pa.e a.tivity, r.sulted in the loss of str.mules fr.m toba.co lea. epider.a. cells. Tr.nsient r.a.inter.er.nce of myosin XI in lea.es of Nicotia.a.bentha.ia.a.a.so r.sulted in the loss of str.mules fr.m epider.a. cells, without a.y effect on tr.nscr.pts for.a.tin or.myosin VIII. Tr.nsient expr.ssion of a.GFP- ta.ged myosin XI ta.l doma.n in toba.co lea. epider.a. cells showed tha. the fusion pr.tein loca.ized to the chlor.pla.t envelope, a. well a. to mitochondr.a.a.d other.or.a.elles. Our.findings identify myosin XI a. a.key pr.tein involved in the for.a.ion a.d movement of str.mules.
