Determination of Optimal Hormone and Mineral Salts Levels in Tissue Culture Media for Callus Induction and Growth of Industrial Hemp (<i>Cannabis sativa</i>L.)

Historically, industrial hemp (Cannabis sativa L.) has been a valuable source of metabolites and compounds, such as cannabidiols. There is a need for large amounts of plant tissue to be grown under controlled environments, and plant tissue culture is one unique way to yield this tissue. The purposes of this study were to determine: 1) the optimal concentrations (μM)/ratios of auxin:cytokinin in media and;2) the optimal mineral salts formulation for callus induction and callus growth in select hemp cultivars. To find the optimal concentration/ratios, 16 different combinations of auxin:cytokinin and three different mineral salts formulations were evaluated. The three mineral salts formulations tested were MS salts, MB5D1K and an MTSU formulation. The top performing hormone formulations were determined to be equal concentrations (1:1, 2:2, 3:3 μM) of auxin and cytokinin. The top performing media hormone formulations for callus induction were determined to be 2:1, 2:2, 2:3, and 3:2 μM (auxin:cytokinin). The optimal mineral salts formulation was determined to be MD5D1K. Therefore, the overall optimal media formulation for hemp callus production would be MB5D1K salts with the concentration/ratio of 2:2 μM (auxin:cytokinin).

Vitis aestivalis “Norton/Cynthiana” is a grape native to North America and utilized in the commercial wine industry. The purpose of this research was to determine the appropriate time of year and/or time after last freeze that will yield the most successful propagation rate for Vitis aestivalis. It was hypothesized that the highest rate of propagation would be in early summer. During a year-long study (2015-2016), forty cuttings were taken semi-monthly from a vineyard, treated with 0.1% IBA and placed in a plant growth room set to spring conditions. Daily temperatures were recorded every day as well. In a follow-up study, cuttings were taken weekly from early spring before vine budding until late fall (2017) and treated as previous. After six weeks, cuttings were evaluated for root production. In both studies, rooting success rates were the highest in the month of June at 10.0% - 27.5%, with rates less than 7.5% for all other sampling dates. It was determined that the best time to propagate Vitis aestivalis is in June, 10 - 12 weeks after the last temperature below 0°C or eight to ten weeks after the first budding.

The fauna of the Pleistocene Homo-bearing sites of Java has been well known for more than a century.A recent revision of the crocodylian remains confirmed both the validity of Gavialis bengawanicus and the synonymiza-tion of Crocodylus ossifragus with *** we report on a still unpublished crocodylian specimen collected by Eugene dubois in the latest Early Pleistocene of Kali Gedeh that can be tentatively referred to the genus *** size of the specimen,the approximately 1 m long lower jaw in particular,indicat-ed that this crocodile attained a total length of approximately 6 or 7 *** with specimens from the Plio-Pleistocene of Africa,this material provides evidence for gigantism in *** is not clear whether or not the‘temperature-size rule’applies to fossil crocodylians,but due to the growing interest in predicting future temperature-related size changes of the extant organisms,it would be interesting to study in detail the past reac-tion to temperature changes of crocodylians and other terrestrial ectothermic animals.

Long-term genetic studies utilizing backcross and congenic strain analyses coupled with positional cloning strategies and functional studies identified Cdkn2a,Mtor,and Mndal as mouse plasmacytoma susceptibility/resistance *** incidence data in congenic strains carrying the resistance alleles of Cdkn2a and Mtor led us to hypothesize that drug combinations affecting these pathways are likely to have an additive,if not synergistic effect in inhibiting tumor cell *** and novel systems-level genomic approaches were used to assess combination activity,disease specificity,and clinical potential of a drug combination involving rapamycin/everolimus,an Mtor inhibitor,with entinostat,an histone deacetylase *** combination synergistically repressed oncogenic MYC and activated the Cdkn2a tumor *** identification of MYC as a primary upstream regulator led to the identification of small molecule binders of the G-quadruplex structure that forms in the NHEIII region of the MYC *** studies highlight the importance of identifying drug combinations which simultaneously upregulate tumor suppressors and downregulate oncogenes.

本文简要回顾了四个世纪的海外中国研究,论述了学术结构的历史需求和路径依赖如何在不同时期围绕不断变化的中心范式关注学术研究。本文将海外中国研究划分为五波浪潮:古典汉学的文化、语言和宗教研究,民国时期的改良主义社会科学,冷战的政治问题,80年代的后现代主义社会史,以及当代40岁以下学者的新关注点。每一个时期不仅主要研究范式不同,学术界性质也产生了变化,研究中心从传教士转向社会科学家、从欧洲转向美国、从学术刊物转向基于互联网的出版物。
