Assessment of the Bemisia tabaci C YP6CM l vQ transcript and protein levels in laboratory and field-derived imidacloprid-resistant insects and cross-metabolism potential of the recombinant enzyme

Over-expression of the cytochrome P450 CYP6CM1 gene has been associated with imidacloprid resistance in a number of Q and B biotype Bemisia tabaci laboratory strains from distinct geographical origins worldwide. We recently demonstrated that the Q biotype version of the CYP6CM 1 protein （CYP6CMlvQ） is capable of metabolizing imida- cloprid. Here, we show that the levels of BtCYP6CMlvQ were also elevated in laboratory- resistant strains and field-derived populations, with variable imidacloprid resistance levels, collected in Crete. High levels of CYP6CMlvQ transcripts were also determined in survivors of a heterogeneous field population, after exposure to discriminating imidacloprid dosage. Using peptide antibody-based detection assays, we demonstrated that in line with transcriptional data, the CYP6CMlvQ protein levels were higher in imidacloprid-resistant insects, which further implicates the gene as the causal factor of resistance. Finally, assess- ment of the cross-metabolism potential of CYP6CMlvQ against additional neonicotinoid molecules used for B. tabaci control revealed that clothianidin and thiacloprid, but not acetamiprid or thiamethoxam, are metabolized by the recombinant enzyme in vitro.

Improved estimation of PM_(2.5) brown carbon contributions to filter light attenuation

Izumol is a testis-specific gene product, whose function is essential for sperm-egg fusion. Throughout its lifespan, Izumol is posttranslationally modified, being both N-linked glycosylated on its extracellular domain and phosphorylated on the intracellular C-terminal tail. Within the caput regions of the rat epididymis, two phosphorylation events have been documented. However, as sperm pass through the epididymis, this cytoplasmic portion of Izumol has been shown to contain up to seven phosphorylation sites. Remarkably, in the rat, in correlation with these events, Izumol undergoes sub-cellular re-location, moving from the head/tail regions of the spermatozoa, to a predominantly equatorial segment location once they have reached the caudal end of the epididymis.

Feasibility of genomic prediction for brown rot(Monilinia spp.)resistance in peach

Brown rot,caused by Monilinia spp.,is one of the most important postharvest diseases of stone fruits *** rot resistance in peach is a polygenic trait controlled by multiple genes with a small *** this study,we assessed the feasibility of genomic prediction(GP)for brown rot resistance in peach using eight contrasting methods(GBLUP,rrBLUP,BayesA,BayesB,BayesC,Bayesian Ridge Regression,Bayesian Lasso and RKHS).A testing panel of 38 cultivars/advanced selections and 288 F1 individuals from 27 pedigree-related breeding families with'Bolinha'and/or'Contender'or almond source of resistance was phenotyped over six seasons(2015 to 2020).GP models outperformed MAS models under fivefold cross validation,and low to moderate predictive accuracy(PA)was achieved by fitting GP model for wounded(W)(0.092−0.449)and low PA for non-wounded(NW)disease severity index(0.129−0.295).An alternative cross validation approach using disease severity index recorded in lab to predict field disease incidence(FDI)in unphenotyped accessions revealed moderate correlation(0.548−0.553).Genomic predicted breeding value distinguished accessions with low FDI from those with high *** results presented here demonstrated feasibility of incorporating GP in peach breeding.

Knowledge of the stress field in the Earth＇s crust is a key issue for the understanding of geodynamic processes, seismic hazard assessment, and stability of underground openings such as waste disposals, tunnels, mines or wells, and reservoir management. The World Stress Map project is a collaborative project of academia, industry and governmental organizations that aims to understand the states and sources of tectonic stresses in the Earth＇s crust. We present the World Stress Map at 1：46,000,000 scale as a result of more than two decades of international collaboration. The map reveals that the first-order pattern of stress is of plate-wide scale, indi- cating that plate boundary forces are the major control of the stress orientations and the tectonic regime.

Geo-Hazard Susceptibility Assessment and Its Impacts on Livelihoods in Kerio Valley, Kenya

Geohazards are a recurrent issue in the Kerio River catchment of Kenya, which usually results in life and property loss. This research focuses on mapping geo-hazard risk zones of the region. The risk zones were developed from a combination of land use land cover maps, agroecological zones maps and soil erosion maps using the Analytical Hierarchy Process method of multi-criteria analysis. The final results depict the geohazard risk maps which show the susceptibility of different areas in the catchment (classified as risk zones) to hazards. The zones range from no risk zones to very high-risk zones. The results showed that the lowlands are most susceptible to hazards as they were classified as high-risk zones. These risk zone areas have impacts on the socio-economic development hence negatively impacting livelihoods in the area.

Proteomic changes in mammalian spermatozoa during epididymal maturation

Epididymal maturation is associated with the activation of a cAMP-induced tyrosine phosphorylation cascade, which is ultimately associated with the expression of capacitation-dependent sperm functions, such as hyperactivated movement and acrosomal exocytosis. As spermatozoa progress through the epididymis they first acquire the capacity to phosphorylate tyrosine on targets on the principal piece, followed by the midpiece. By the time these cells have reached the cauda epididymidis they can phosphorylate the entire tail from neck to endpiece. This particular pattern of phosphorylation is associated with the ontogeny of fully functional spermatozoa that are capable of fertilizing the oocyte. Proteomic analyses indicate that this change is associated with the phosphorylation of several mitochondrial proteins, creation of a mitochondrial membrane potential and activation of mitochondrial free radical generation. At least in rodent species, activation of sperm mitochondria appears to be a particularly important part of epididymal maturation. （Asian J Androl 2007 July; 9： 554-564）
