Different apoptotic reactions of dorsal root ganglion A- and B-cells after sciatic nerve axotomy： effect of p75 neurotrophin receptor

Background By unbiased stereological methods, we have observed preferential dorsal root ganglion （DRG） B-cell loss in rodents after nerve injury, and caspase-3 activation and cell loss were related to the present of p75 receptor （p75^NTR）. We hypothesized that DRG B-cells express higher levels of pro-apoptotic proteins as compared to A-cells and the expressions of pro-apoptotic proteins can be reduced by depletion of p75^NTR. This study aimed to identify the p75NTR involved apoptotic pathway in DRG neurons after nerve injury. Methods The p75NTR knockout mice （p75-/-） and wildtype Balb/C mice （p75＋/＋） were used in this study. The expressions of pro-apoptotic proteins, c-Jun-N-terminal kinase （JNK）, c-jun and p38 in DRG were evaluated with immunohistochemistry 2 and 7 days following unilateral sciatic nerve transection. In addition, extra-cellular related kinase （ERK）, a transducer of survival signals, was also tested with immunohistochemistry and Western blotting methods in these animal models. Results Phosphorylated JNK （P-JNK） and phosphorylated p38 （P-p38） were mainly located in small B-cells, whereas phosphorylated c-jun （P-c-jun） was located in both A- and B-cells. Phosphorylated ERK （P-ERK） was located in both B-cells and satellite cells. Axotomy dramatically increased the expressions of P-JNK and P-c-jun （paired t-test）, with no influence on the expressions of P-p38 and P-ERK. Furthermore, the increase of P-JNK in p75＋/＋ mice 2 days after nerve axotomy was approximately 2.2-folds of that in p75-/- mice （P=-0.001, unpaired t-test）. Conclusion p75NTR-dependent JNK-caspase-3 pathway is involved in DRG B-cell loss after nerve injury and JNK is not the unique upstream of c-jun activation.

Current treatment of ulcerative colitis

Ulcerative colitis (UC) is a chronic disease featuring re- current inflammation of the colonic mucosa. The goal of medical treatment is to rapidly induce a steroid-free remission while at the same time preventing complica- tions of the disease itself and its treatment. The choice of treatment depends on severity, localization and the course of the disease. For proctitis, topical therapy with 5-aminosalicylic acid (5-ASA) compounds is used. More extensive or severe disease should be treated with oral and local 5-ASA compounds and corticosteroids to induce remission. Patients who do not respond to this treatment require hospitalization. Intravenous steroids or, when refractory, calcineurin inhibitors (cyclosporine, tacrolimus), tumor necrosis factor-α antibodies (infliximab) or immunomodulators (azathioprine, 6-mercaptopurine) are then called for. Indications for emergency surgery include refractory toxic megacolon, perforation, and continuous severe colorectal bleeding. Close collaboration between gastroenterologist and surgeon is mandatory in order not to delay surgical therapy when needed. This article is intended to give a general, practice-orientated overview of the key issues in ulcerative colitis treatment. Recommendations are based on published consensus guidelines derived from national and international guidelines on the treatment of ulcerative colitis.

1|INTRODUCTION Capecitabine is an oral prodrug of 5-fluorouracil(5-FU),which is widely used for adjuvant and neoadjuvant chemotherapy of different solid tumors,particularly breast and colorectal cancers.1 Neurotoxicity of capecitabine has been consistently reported as capecitabine-induced toxic leukoencephalopathy,which includes bilateral lesions in the corpus callosum and corticospinal tract presenting as acute or delayed central nervous system toxicity.2 This side effect requires discontinuation of chemotherapy3;however,neurological symptoms due to capecitabine are reported to be usually reversible upon drug withdrawal.

Wind is an environmental stimulus that stresses plants of all growth forms at all life-stages by influencing the development,architecture,and morphology of roots and ***,comparative studies are scarce and no study directly investigated whether shoot and root morphological traits of trees,grasses and forbs differ in their response to short wind pulses of different wind *** this study,we found that across species,wind stress by short wind pulses of increasing intensity consistently changed root morphology,but did not affect shoot morphological traits,except plant height in four *** effects in roots were generally weak in tree species but consistent across growth ***,plant height of species was correlated with changes in specific root length and average *** results indicate that short-pulse wind treatments affect root morphology more than shoot morphology across growth *** further suggest that wind stress possibly promotes root anchorage in young plants and that these effects might depend on plant height.

To the Editor,Marfan syndrome is an autosomal dominant disorder of connective tissue caused by mutations in the gene encoding a matrix component of microfibrils;fibrillin-1(FBN1),with an incidence of about 1-3 per 10,000 individuals.2.3 Cardinal manifestations include proximal aortic aneurysm,dislocation of the ocular lens,and long-bone *** ectasia,which is defined as a ballooning or significant widening of the dural sac or neural root sleeves,is also a common finding in these patients,which is reported to present in more than 60%of cases and is considered a major criterion for the clinical diagnosis of this syndrome.

On the applicability of the single parabolic band model to advanced thermoelectric materials with complex band structures

Due to the complex interplay between composition,synthesis parameters and the performance of thermoelectric materials,the optimization of thermoelectric materials needs to be complemented by modelling.A relatively simple and thus popular approach is the so called single parabolic band model,which allows for an efficient optimization of the material properties and a benchmarking of different materials based on relatively few,well available experimental *** complex band structures are common for high performance materials,single parabolic band modelling is also employed with apparent success for material systems where the underlying assumptions are not well *** order to assess the validity of a single parabolic band analysis for such systems,the thermoelectric properties for two model systems are calculated:one with a single band that is twofold degenerate and one with a light and a heavy *** if the density of states masses and the scattering potentials are kept identical,the transport properties and in particular the Hall coefficients differ significantly,which leads to an incorrectly determined carrier *** the carrier concentration is the base for the single parabolic band analysis,all the quantities obtained from it(optimum carrier concentration,effective mass,deformation potential)are determined incorrectly as well.

The molecular mechanism of shade avoidance in crops--How data from Arabidopsis can help to identify targets for increasing yield and biomass production

In order to prevent or counteract shading,plants enact a complex set of growth and developmental adaptations when they sense a change in light quality caused by other plants in their *** shade avoidance response（SAR）typically includes increased stem elongation at the expense of plant fitness and yield,making it an undesirable trait in an agricultural *** the molecular factors involved in SAR can potentially improve productivity by increasing tolerance to higher planting ***,most of the investigations of the molecular mechanism of SAR have been carried out in Arabidopsis thaliana,and it is presently unclear in how far results of these investigations apply to crop *** this review,current data on SAR in crop plants,especially from members of the Solanaceae and Poaceae families,are integrated with data from Arabidopsis,in order to identify the most promising targets for biotechnological ***,which detect the change in light caused by neighboring plants,and early signaling components can be targeted to increase plant ***,they control various photomorphogenic processes not necessarily related to shade *** factors involved in SAR signaling could be better targets to specifically enhance or suppress *** integration from Arabidopsis and crop plants also indicates factors that could facilitate the control of specific aspects of *** are provided for the regulation of plant architecture,flowering induction and carbohydrate *** to-be-elucidated factors that control SAR-dependent changes in biotic resistance and cell wall composition are pointed *** review also includes an analysis of publicly available gene expression data for maize to augment the sparse molecular data available for this important species.
