Morphological alterations and redox changes associated with hepatic warm ischemia-reperfusion injury

AIM To study the effects of warm ischemia-reperfusion(I/R) injury on hepatic morphology at the ultrastructural level and to analyze the expression of the thioredoxin(TRX)and glutaredoxin(GRX) *** Eleven patients undergoing liver resection were subjected to portal triad clamping(PTC). Liver biopsies were collected at three time points; first prior to PTC(baseline), 20 min after PTC(post-ischemia) and 20 min after reperfusion(post-reperfusion). Electron microscopy and morphometry were used to study and quantify ultrastructural changes, respectively. Additionally, gene expression analysis of TRX and GRX isoforms was performed by quantitative PCR. For further validation of redox protein status, immunogold staining was performed for the isoforms GRX1 and *** Post-ischemia, a significant loss of the liver sinusoidal endothelial cell(LSEC) lining was observed(P = 0.0003) accompanied by a decrease of hepatocyte microvilli in the space of Disse. Hepatocellular morphology was well preserved apart from the appearance of crystalline mitochondrial inclusions in 7 out of 11 patients. Postreperfusion biopsies had similar features as post-ischemia with the exception of signs of a reactivation of the LSECs. No changes in the expression of redox-regulatory genes could be observed at mR NA level of the isoforms of the TRX family but immunoelectron microscopy indicated a redistribution of TRX1 within the *** At the ultrastructural level, the major impact of hepatic warm I/R injury after PTC was borne by the LSECs with detachment and reactivation at ischemia and reperfusion, respectively. Hepatocytes morphology were well preserved. Crystalline inclusions in mitochondria were observed in the hepatocyte after ischemia.

This study illustrates an innovative way to fabricate inkjet-printed tracks by sequential printing of Zn nanoparticle ink and curing ink for low temperature in situ chemical *** chemical curing in place of standard sintering methods leads to the advantages of using flexible substrates that may not withstand the high thermal budgets of the standard methods.A general formulation engineering method is adopted to produce highly concentrated Zn ink which is cured by inkjet printing an over-layer of aqueous acetic acid which is the curing *** experimental results reveal that a narrow window of acid concentration of curing ink plays a crucial role in determining the electrical properties of the printed Zn *** conductive(~10^(5)S m^(−1))and mechanically flexible printed Zn features are *** addition,from systematic material characterization,we obtain an understanding of the curing ***,a touch sensor circuit is demonstrated involving all-Zn printed conductive tracks.

Mucosal-associated invariant T cells and oral microbiome in persistent apical periodontitis

Opportunistic bacteria in apical periodontitis (AP) may pose a risk for systemic ***-associated invariant T (MAIT) cells are innate-like T cells with a broad and potent antimicrobial activity important for gut mucosal *** was recently shown that MAIT cells are present in the oral mucosal tissue,but the involvement of MAIT cells in AP is ***,comparison of surgically resected AP and gingival tissues demonstrated that AP tissues express significantly higher levels of Vα7.2-Jα33,Vα7.2- Jα20,Vα7.2-Jα12,Cα and tumour necrosis factor (TNF),interferon (IFN)-γ and interleukin (IL)-17A transcripts,resembling a MAIT cell ***,in AP tissues the MR1-restricted MAIT cells positive for MR1–5-OP-RU tetramer staining appeared to be of similar levels as in peripheral blood but consisted mainly of CD4^+ *** gingival tissues,the AP microbiome was quantitatively impacted by factors like fistula and high patient age and had a prominent riboflavin-expressing bacterial *** merged in an integrated view,the examined immune and microbiome data in the sparse partial least squares discriminant analysis could identify bacterial relative abundances that negatively correlated with Vα7.2-Jα33,Cα,and IL-17A transcript expressions in AP,implying that MAIT cells could play a role in the local defence at the oral tissue *** conclusion,we describe the presence of MAIT cells at the oral site where translocation of oral microbiota could take *** findings have implications for understanding the immune sensing of polymicrobial-related oral diseases.

Northern Fulmars(Fulmarus glacialis)are a pelagic seabird species distributed at northern and polar *** are often used as an indicator of plastic pollution in the North Sea region,but data are lacking from higher latitudes,especially when it comes to ***,we investigated amounts of ingested plastic and their characteristics in fulmar chicks from the Faroe *** particles(≥1 mm)in chicks of two age classes were searched using a digestion method with *** addition,to evaluate if additive tissue burden reflects plastic ingestion,we measured liver tissue concentrations of two pollutant classes associated with plastic materials:polybrominated diphenyl ethers(PBDEs)and several dechloranes,using gas chromatography with high-resolution mass *** most common shape was hard fragment(81%)and the most common polymer was polyethylene(73%).Plastic contamination did not differ between either age class,and we found no correlation between neither the amount and mass of plastic particles and the concentration of *** comparison with previous studies on adult fulmars,we do not recommend using chicks for biomonitoring adults because chicks seem to ingest more plastics than adults.

Abscisic Acid Transport and Homeostasis in the Context of Stomatal Regulation

The discovery of cytosolic ABA receptors is an important breakthrough in stomatal research; signaling via these receptors is involved in determining the basal stomatal conductance and stomatal responsiveness. However, the source of ABA in guard cells is still not fully understood. The level of ABA increases in guard cells by de novo synthesis, recycling from inactive conjugates via β-glucosidases BG1 and BG2 and by import, whereas it decreases by hydroxylation, conjugation, and export. ABA importers include the NRT1/PTR family protein AIT1, ATP-binding cassette protein ABCG40, and possibly ABCG22, whereas the DTX family member DTX50 and ABCG25 function as ABA exporters. Here, we review the proteins involved in ABA transport and homeostasis and their physiological role in stomatal regulation. Recent experiments suggest that functional redundancy probably exists among ABA transporters between vascu- lature and guard cells and ABA recycling proteins, as stomatal functioning remained intact in abcg22, abcg25, abcg40, aitl, and bglbg2 mutants. Only the initial response to reduced air humidity was significantly delayed in abcg22. Considering the reports showing autonomous ABA synthesis in guard cells, we discuss that rapid stomatal responses to atmospheric factors might depend primarily on guard cellsynthesized ABA, whereas in the case of long-term soil water deficit, ABA synthesized in the vasculature might have a significant role.

Preparation of High Percentage α-Calcium Sulfate Hemihydrate via a Hydrothermal Method

α-calcium sulfate hemihydrate (α-HH) is known to be suitable for application as bone void filler. High percentage of α-HH is obviously needed for medical applications, especially for implantation. Three commercially available calcium sulfate dihydrates (DH, CaSO4·2H2O) with different sizes and surface morphologies were used as starting materials to synthesize high percentage α-HH via a hydrothermal method. The median particle sizes of the three types of DH were 946.7 μm, 162.4 μm and 62.4 μm, respectively. They were named as DH-L, DH-M and DH-S in this paper. The particle size distribution, morphology and phase composition of the raw materials were evaluated before synthesis. SEM results revealed that DH-L consisted of irregular large particles, while DH-M and DH-S were composed of plate-like particles with some small ones. High percentage HH can be obtained with proper synthesis parameters by hydrothermal method, specifically, 105 °C/90 min for DH-L (achieving 98.8% HH), 105°C/30 min for DH-M (achieving 96.7% HH) and 100°C/45 min for DH-S (achieving 98.4% HH). All the synthesized HH were hexagonal columns, demonstrating that they were α-phase HH. The particle size and morphology of starting material (DH) have significant influences on not only the rate of phase transition but also the morphology of the synthesized α-HH. Calcium sulfate dihydrate cements were prepared by the synthesized α-HH. The highest compressive strength of calcium sulfate dihydrate cement was 17.2 MPa. The results show that the preparation of high percentage α-HH is feasible via a hydrothermal method and the process can be further scaled up to industrial scale production.

Graphene’s unparalleled strength,chemical stability,ultimate surface-to-volume ratio and excellent electronic properties make it an ideal candidate as a material for membranes in micro-and nanoelectromechanical systems(MEMS and NEMS).However,the integration of graphene into MEMS or NEMS devices and suspended structures such as proof masses on graphene membranes raises several technological challenges,including collapse and rupture of the *** have developed a robust route for realizing membranes made of double-layer CVD graphene and suspending large silicon proof masses on membranes with high *** have demonstrated the manufacture of square graphene membranes with side lengths from 7µm to 110µm,and suspended proof masses consisting of solid silicon cubes that are from 5µm×5µm×16.4µm to 100µm×100µm×16.4µm in *** approach is compatible with wafer-scale MEMS and semiconductor manufacturing technologies,and the manufacturing yields of the graphene membranes with suspended proof masses were>90%,with>70%of the graphene membranes having>90%graphene area without visible *** measured resonance frequencies of the realized structures ranged from tens to hundreds of kHz,with quality factors ranging from 63 to *** graphene membranes with suspended proof masses were extremely robust,and were able to withstand indentation forces from an atomic force microscope(AFM)tip of up to~*** proposed approach for the reliable and large-scale manufacture of graphene membranes with suspended proof masses will enable the development and study of innovative NEMS devices with new functionalities and improved performances.

A Pleural mass biopsy was performed showing an invasive thymoma. Computed tomography (CT) scan of the chest performed on admission revealed a left sided pleural mass, anterior mediastinal mass, lymphadenopathy and pericardial effusion. Pleural mass biopsy showed an invasive thymoma. Due to her clinical presentation, a complete work-up was performed revealing paraneoplastic systemic lupus erythematous (SLE), myasthenia gravis (MG) and hypogammaglobulinemia. We reviewed the literature regarding thymoma and its relationship with paraneoplastic SLE, MG and hypogammaglobulinemia.

Is there an optimal environment in which to learn clinical anatomy? One man’s view

Learning anatomy is essential in medical schools, and even more so for budding surgeons. Much has been discussed regarding the updated curriculum, and whether the pedagogies should be based upon cadaveric, and/or multimedia, or hybrid approaches. Much debate has also been centered on who is best qualified to teach. While all these are important, the setting is also critically important for the medical students and surgical trainees. Therefore the overarching issue is whether all these activities should be held in the dissection room, the operating theatre or the classical “Theatrum Anatomicum” ? What are the key experiential learning differences in picking up anatomical knowledge in the various venues listed above? This paper will provide some insights for teachers and students of human anatomy, and some ideas for the future planners and developers of anatomy learning centers internationally.
