Microbial markers in colorectal cancer detection and/or prognosis

Colorectal cancer(CRC) is the second leading cause of cancer worldwide. CRC is still associated with a poor prognosis among patients with advanced disease. On the contrary, due to its slow progression from detectable precancerous lesions, the prognosis for patients with early stages of CRC is encouraging. While most robust methods are invasive and costly, actual patient-friendly screening methods for CRC suffer of lack of sensitivity and specificity. Therefore, the development of sensitive, non-invasive and cost-effective methods for CRC detection and prognosis are necessary for increasing the chances of a cure. Beyond its beneficial functions for the host, increasing evidence suggests that the intestinal microbiota is a key factor associated with carcinogenesis. Many clinical studies have reported a disruption in the gut microbiota balance and an alteration in the faecal metabolome of CRC patients, suggesting the potential use of a microbialbased test as a non-invasive diagnostic and/or prognostic tool for CRC screening. This review aims to discuss the microbial signatures associated with CRC known to date, including dysbiosis and faecal metabolome alterations, andthe potential use of microbial variation markers for noninvasive early diagnosis and/or prognostic assessment of CRC and advanced adenomas. We will finally discuss the possible use of these markers as predicators for treatment response and their limitations.

Background:Liver resection and local ablation are the only curative treatment for non-cirrhotic hepatocellular carcinoma(HCC).Few data exist concerning the prognosis of patients resected for non-cirrhotic *** objectives of this study were to determine the prognostic factors of recurrence-free survival(RFS)and overall survival(OS)and to develop a prognostication algorithm for non-cirrhotic ***:French multicenter retrospective study including HCC patients with non-cirrhotic liver without underlying viral hepatitis:F0,F1 or F2 ***:A total of 467 patients were included in 11 centers from 2010 to ***-cirrhotic liver had a fibrosis score of F0(n=237,50.7%),F1(n=127,27.2%)or F2(n=103,22.1%).OS and RFS at 5 years were 59.2%and 34.5%,*** multivariate analysis,microvascular invasion and HCC differentiation were prognostic factors of OS and RFS and the number and size were prognostic factors of RFS(P10 cm(n=176,Group 2);>2 HCC regardless of size Conclusions:We developed a prognostication algorithm based on the number(≤or>2)and size(≤or>10 cm),which could be used as a treatment decision support concerning the need for perioperative *** case of bifocal HCC,surgery should not be a contraindication.

Association of colorectal cancer with pathogenic Escherichia coli: Focus on mechanisms using optical imaging

AIM: To investigate the molecular or cellular mechanisms related to the infection of epithelial colonic mucosa by pks-positive Escherichia coli(E. coli) using optical ***: We choose to evaluate the tumor metabolic activity using a fluorodeoxyglucose analogue as 2-deoxyglucosone fluorescent probes and to correlate it with tumoral volume(mm^3). Inflammation measuring myeloperoxidase(MPO) activity and reactive oxygen species production was monitored by a bioluminescent(BLI) inflammation probe and related to histological examination and MPO levels by enzyme-linked immunosorbent assay(ELISA) on tumor specimens. The detection and quantitation of these two signals were validated on a xenograft model of human colon adenocarcinoma epithelial cells(HCT116) in nude mice infected with a pks-positive E. coli. The inflammatory BLI signal was validated intra-digestively in the colitisCEABAC10 DSS models, which mimicked Crohn's disease. reSULTS: Using a 2-deoxyglucosone fluorescent probe, we observed a high and specific HCT116 tumor uptake in correlation with tumoral volume(P = 0.0036). Using the inflammation probe targeting MPO, we detected a rapid systemic elimination and a significant increase of the BLI signal in the pks-positive E. coli-infected HCT116 xenograft group(P re significantly higher(1556 ± 313.6 vs 234.6 ± 121.6 ng/m L P = 0.001) in xenografts infected with the pathogenic E. coli strain. Moreover, histological examination of tumor samples confirmed massive infiltration of pks-positive E. coli-infected HCT116 tumors by inflammatory cells compared to the uninfected group. These data showed that infection with the pathogenic E. coli strain enhanced inflammation and ROS production in tumors before tumor growth. Moreover, we demonstrated that the intra-digestive monitoring of inflammation is feasible in a reference colitis murine model(CEABAC10/DSS).CONCLUSION: Using BLI and fluorescence optical imaging, we provided

Massive hepatic necrosis with toxic liver syndrome following portal vein ligation

Right portal vein ligation (PVL) is a safe and widespread procedure to induce controlateral liver hypertrophy for the treatment of bilobar colorectal liver metastases. We report a case of a 60-year-old man treated by both right PVL and ligation of the glissonian branches of segment 4 for colorectal liver metastases surrounding the right and median hepatic veins. After surgery, the patient developed massive hepatic necrosis with secondary pulmonary and renal insufficiency requiring transfer to the intensive care unit. This so-called toxic liver syndrome finally regressed after hemofiltration and positive oxygen therapy. Diagnosis of acute congestion of the ligated lobe was suspected. The mechanism suspected was an increase in arterial inflow secondary to portal vein ligation concomitant with a decrease in venous outflow due to liver metastases encircling the right and median hepatic vein. This is the first documented case of toxic liver syndrome in a non-cirrhotic patient with favorable issue, and a rare complication of PVL.

Mapping and understanding degradation of alpine wetlands in the northern Maloti-Drakensberg, southern Africa

The alpine terrestrials of the Maloti-Drakensberg in southern Africa play crucial roles in ecosystem functions and livelihoods,yet they face escalating degradation from various factors including overgrazing and climate *** study employs advanced Digital Soil Mapping(DSM)techniques coupled with remote sensing to map and assess wetland coverage and degradation in the northern *** model achieved high accuracies of 96%and 92%for training and validation data,respectively,with Kappa statistics of 0.91 and 0.83,marking a pioneering automated attempt at wetland mapping in this *** attributes such as terrain wetness index(TWI)and valley depth(VD)exhibit significant positive correlations with wetland coverage and erosion gully density,Channel Network Depth and slope were negative *** density analysis revealed terrain attributes as dominant factors driving degradation,highlighting the need to consider catchment-specific susceptibility to *** challenge traditional assumptions which mainly attribute wetland degradation to external forces such as livestock overgrazing,ice rate activity and climate *** sensitivity map produced could serve as a basis for Integrated Catchment Management(ICM)projects,facilitating tailored conservation *** research should expand on this work to include other highland areas,explore additional covariates,and categorize wetlands based on hydroperiod and sensitivity to *** comprehensive study underscores the potential of DSM and remote sensing in accurately assessing and managing wetland ecosystems,crucial for sustainable resource management in alpine regions.

The gut microbiota acts as a real organ. The symbiotic interactions between resident micro-organisms and the digestive tract highly contribute to maintain the gut homeostasis. However, alterations to the microbiome caused by environmental changes(e.g., infection, diet and/or lifestyle) can disturb this symbiotic relationship and promote disease, such as inflammatory bowel diseases and cancer. Colorectal cancer is a complex association of tumoral cells, non-neoplastic cells and a large amount of micro-organisms, and the involvement of the microbiota in colorectal carcinogenesis is becoming increasingly clear. Indeed, many changes in the bacterial composition of the gut microbiota have been reported in colorectal cancer, suggesting a major role of dysbiosis in colorectal carcinogenesis. Some bacterial species have been identified and suspected to play a role in colorectal carcinogenesis, such as Streptococcus bovis, Helicobacter pylori, Bacteroides fragilis, Enterococcus faecalis, Clostridium septicum, Fusobacterium spp. and Escherichia coli. The potential pro-carcinogenic effects of these bacteria are now better understood. In this review, we discuss the possible links between the bacterial microbiota and colorectal carcinogenesis, focusing on dysbiosis and the potential pro-carcinogenic properties of bacteria, such as genotoxicity and other virulence factors, inflammation, host defenses modulation, bacterial derived metabolism, oxidative stress and anti-oxidative defenses modulation. We lastly describe how bacterial microbiota modifications could represent novel prognosis markers and/or targets for innovative therapeutic strategies.

Thermal Property Enhancement of a Novel Shape-Stabilized Sodium Acetate Trihydrate-Acetamide/Expanded Graphite-Based Composite Phase Change Material

Phase change materials(PCMs)are a kind of highly efficient thermal storage materials which have a bright application prospect in many fields such as energy conservation in buildings,waste heat recovery,battery thermal management and so *** inorganic hydrated salt PCMs have received increasing attention from researchers due to their advantages of being inexpensive and ***,inorganic hydrated salt PCMs are still limited by the aspects of inappropriate phase change temperature,liquid phase leakage,large supercooling and severe phase separation in the application *** this work,sodium acetate trihydrate was selected as the basic inorganic PCM,and a novel shape-stabilized composite phase change material(CPCM)with good thermal properties was prepared by adding various functional *** first,the sodium acetate trihydrate-acetamide binary mixture was prepared and the melting point was adjusted using *** the binary mixture was incorporated into expanded graphite to synthesize a novel shape-stabilized *** thermophysical properties of the resultant shape-stabilized CPCM were systematically *** microscopic morphology and chemical structure of the obtained shape-stabilized CPCM were characterized and *** experiment results pointed out that acetamide could effectively lower the melting point of sodium acetate *** obtained shape-stabilized CPCM modified with additional 18%(mass fraction)acetamide and 12%(mass fraction)expanded graphite exhibited good shape stability and thermophysical characteristics:a low supercooling degree of 1.75℃and an appropriate melting temperature of 40.77℃were obtained;the latent heat of 151.64 kJ/kg and thermal conductivity of 1.411 W/(m·K)were also ***,after 50accelerated melting-freezing cycles,the obtained shape-stabilized CPCM represented good thermal reliability.

Exploring well-being disparities between urban and rural areas:A case study in the Stavropol Territory,Russia

Rural areas are crucial for a country’s sustainable *** strategies are needed to develop rural areas to improve the well-being of rural population and generate new job *** is especially important in countries where agricultural production accounts for a significant share of the gross product,such as *** this study,we identified the key indicators of satisfaction and differences between rural and urban citizens based on their social,economic,and environmental backgrounds,and determined whether there are well-being disparities between rural and urban areas in the Stavropol Territory,*** collected primary data through a survey based on the European Social Survey framework to investigate the potential differences between rural and urban *** computing the regional well-being index using principal component analysis,we found that there was no statistically significant difference in well-being between rural and urban *** of key indicators showed that rural residents felt psychologically more comfortable and safer,assessed their family relationships better,and adhered more to traditions and ***,urban residents showed better economic and social conditions(e.g.,infrastructures,medical care,education,and Internet access).The results of this study imply that we can better understand the local needs,advantages,and unique qualities,thereby gaining insight into the effectiveness of government ***-makers and local authorities can consider targeted interventions based on the findings of this study and strive to enhance the well-being of both urban and rural residents.

The number and composition of species in a community can be quantified withα-diversity indices,including species richness(R),Simpson’s index(D),and the Shannon-Wiener index(H΄).In forest communities,there are large variations in tree size among species and individu-als of the same species,which result in differences in eco-logical processes and ecosystem ***,tree size inequality(TSI)has been largely neglected in studies using the available diversity *** TSI in the diameter at breast height(DBH)data for each of 99920 m×20 m forest census quadrats was quantified using the Gini index(GI),a measure of the inequality of size *** generalized performance equation was used to describe the rotated and right-shifted Lorenz curve of the cumulative proportion of DBH and the cumulative proportion of number of trees per *** also examined the relationships ofα-diversity indices with the GI using correlation *** generalized performance equation effectively described the rotated and right-shifted Lorenz curve of DBH distributions,with most root-mean-square errors(990 out of 999 quadrats)beingre significant positive correlations between each of threeα-diversity indices(i.e.,R,D,and H’)and the ***,the total abundance of trees in each quadrat did not significantly influence the *** means that the TSI increased with increasing spe-cies ***,two new indices are proposed that can balanceα-diversity against the extent of TSI in the com-munity:(1−GI)×D,and(1−GI)×H’.These new indices were significantly correlated with the original D and H΄,and did not increase the extent of variation within each group of *** study presents a useful tool for quantifying both species diversity and the variation in tree sizes in forest communities,especially in the face of cumulative species loss under global climate change.
