Evaluation of a chitosan-polyethylene glycol paste as a local antibiotic delivery device

AIm To investigate the efficacy of a chitosan/polyethylene glycol blended paste as a local antibiotic delivery device, particularly in musculoskeletal wounds.mETHODS Acidic(A) chitosan sponges and neutralized(N) chitosan/polyethylene glycol(PEG) blended sponges were combined in ratios of 3A:2N, 1A:1N, and 2A:3N; then hydrated with phosphate buffered saline to form a chitosan/PEG paste(CPP). Both in vitro and in vivo studies were conducted to determine the potential CPP has as a local antibiotic delivery device. In vitro biocompatibility was assessed by the cytotoxic response of fibroblast cells exposed to the experimental groups. Degradation rate was measured as the change in dry mass due to lysozyme based degradation over a 10-d period. The antibiotic elution profiles and eluate activity of CPP were evaluated over a 72-h period. To assess the in vivo antimicrobial efficacy of the CPP, antibiotic-loaded paste samples were exposed to subcutaneously implanted murine catheters inoculated with Staphylococcus aureus. material properties of the experimental paste groups were evaluated by testing the ejection force from a syringe, as well as the adhesion to representative musculoskeletal tissue samples.RESULTS The highly acidic CPP group, 3A:2N, displayed significantly lower cell viability than the control sponge group. The equally distributed group, 1A:1N, and the highly neutral group, 2A:3N, displayed similar cell viability to the control sponge group and are deemed biocompatible. The degradation studies revealed CPP is more readily degradable than the chitosan sponge control group. The antibiotic activity studies indicated the CPP groups released antibiotics at a constant rate and remained above the minimum inhibitory concentrations of the respective test bacteria for a longer time period than the control chitosan sponges, as well as displaying a minimized burst release. As the global population grows,the demand for textiles is increasing rapidly.However,this puts immense pressure on manufacturers to produce more fiber.While synthetic fibers can be pro-duced cheaply,they have a negative impact on the environment.On the other hand,fibers from wool,sisal,fique,wood pulp(viscose),and man-made cellulose fibers(mmCFs)from cotton can-not alone meet the growing fiber demand without major stresses on land,water,and existing markets using these materials.With a greater emphasis on transparency and circular economy practices,there is a need to consider natural non-wood alternative sources for mmCFs to supple-ment other fiber types.However,introducing new feedstocks with different compositions may require different biomass conversion methods.Therefore,based on existing work,this review ad-dresses the technical feasibility of various alternative feedstocks for conversion to textile-grade fibers.First,alternative feedstocks are introduced,and then conventional(dissolving pulp)and emerging(fibrillated cellulose and recycled material)conversion technologies are evaluated to help select the most suitable and promising processes for these emerging alternative sources of cellulose.It is important to note that for alternative feedstocks to be adopted on a meaningful scale,high biomass availability and proximity of conversion facilities are critical factors.In North America,soybean,wheat,rice,sorghum,and sugarcane residues are widely available and most suitable for conventional conversion through various dissolving pulp production methods(pre-hydrolysis kraft,acid sulfite,soda,SO2-ethanol-water,and potassium hydroxide)or by emerging cellulose fibrillation methods.While dissolving pulp conversion is well-established,fibrillated cel-lulose methods could be beneficial from cost,efficiency,and environmental perspectives.Thus,the authors strongly encourage more work in this growing research area.However,conducting thorough cost and sustainability assessments is important to determine the best feedstock a

Luminescence constitutes a unique source of insight into hot carrier processes in metals,including those in plasmonic nanostructures used for sensing and energy applications.However,being weak in nature,metal luminescence remains poorly understood,its microscopic origin strongly debated,and its potential for unraveling nanoscale carrier dynamics largely unexploited.Here,we reveal quantum-mechanical effects in the luminescence emanating from thin monocrystalline gold flakes.Specifically,we present experimental evidence,supported by first-principles simulations,to demonstrate its photoluminescence origin(i.e.,radiative emission from electron/hole recombination)when exciting in the interband regime.Our model allows us to identify changes to the measured gold luminescence due to quantum-mechanical effects as the gold film thickness is reduced.Excitingly,such effects are observable in the luminescence signal from flakes up to 40 nm in thickness,associated with the out-of-plane discreteness of the electronic band structure near the Fermi level.We qualitatively reproduce the observations with first-principles modeling,thus establishing a unified description of luminescence in gold monocrystalline flakes and enabling its widespread application as a probe of carrier dynamics and light-matter interactions in this material.Our study paves the way for future explorations of hot carriers and charge-transfer dynamics in a multitude of material systems.

Systematic Radio Telescope Alignment Using Portable Fringe Projection Profilometry

In 2019,the Event Horizon Telescope(EHT)released the first-ever image of a black hole event horizon.Astronomers are now aiming for higher angular resolutions of distant targets,like black holes,to understand more about the fundamental laws of gravity that govern our universe.To achieve this higher resolution and increased sensitivity,larger radio telescopes are needed to operate at higher frequencies and in larger quantities.Projects like the next-generation Very Large Array(ngVLA)and the Square-Kilometer Array(SKA)require building hundreds of telescopes with diameters greater than 10 ms over the next decade.This has a twofold effect.Radio telescope surfaces need to be more accurate to operate at higher frequencies,and the logistics involved in maintaining a radio telescope need to be simplified to support them properly in large quantities.Both of these problems can be solved with improved methods for surface metrology that are faster and more accurate with a higher resolution.This leads to faster and more accurate panel alignment and,therefore,a more productive observatory.In this paper,we present the use of binocular fringe projection profilometry as a solution to this problem and demonstrate it by aligning two panels on a 3-m radio telescope dish.The measurement takes only 10 min and directly delivers feedback on the tip,tilt,and piston of each panel to create the ideal reflector shape.

Collaborative Efforts and Strategies for Cholera Outbreak Control in Garissa County, Kenya: Implementation of Water Quality monitoring Interventions

A multi-faceted Case Area Targeted Intervention (CATI) approach emphasizing the integration of Water, Sanitation and Hygiene (WASH) interventions and Oral Cholera Vaccine (OCV) campaign was employed to respond to the outbreak of cholera in Garissa County. Drinking water sources in areas heavily impacted by cholera were systematically mapped and tested for microbiological quality. The quality assessment was carried out in April 2023 during an ongoing cholera outbreak in the county. A total of 109 samples were collected and tested for thermotolerant coliforms and other in situ parameters. The finding revealed that more than 87% of the samples did not meet the World Health Organization (WHO) standard for thermotolerant coliforms;and 30% had turbidity values above the recommended threshold values. None of the 109 samples had any traceable residual chlorine. Following these findings, the county government implemented the targeted interventions which resulted in a positive impact in the fight against cholera. The WHO supported key interventions which included capacity building in water quality monitoring and prepositioning of critical WASH commodities to the cholera affected areas.

Diabetic foot ulcer (DFU) is a debilitating and severe manifestation ofuncontrolled and prolonged diabetes that presents as ulceration, usually locatedon the plantar aspect of the foot. Approximately 15% of individuals with diabeteswill eventually develop DFU, and 14%-24% of them will require amputation ofthe ulcerated foot due to bone infection or other ulcer-related complications. Thepathologic mechanisms underlying DFU are comprise a triad: Neuropathy,vascular insufficiency, and secondary infection due to trauma of the foot.Standard local and invasive care along with novel approaches like stem celltherapy pave the way to reduce morbidity, decrease amputations, and preventmortality from DFU. In this manuscript, we review the current literature withfocus on the pathophysiology, preventive options, and definitive management ofDFU.

Exploring the tectonostratigraphy of the Pakistan Himalaya with new detrital zircon geochronology and neodymium isotope data from the Kaghan Valley

In the central Himalaya,the Greater and Lesser Himalayan tectonostratigraphic zones can be distinguished from each other by combining detrital zircon U-Pb ages and whole-rock Nd isotopic data.In the Hazara-Kashmir syntaxis of northern Pakistan,rock units in the Kaghan valley have been variously assigned to the Tethyan,Greater,and Lesser Himalayan zones.To determine which tectonostratigraphic zones are represented,we sampled across the Batal thrust in the northern Kaghan valley because this structure is identified in some research as the main Central thrust.Samples from the footwall yield Paleo-to mesoproterozoic maximum depositional ages(-1.8-1.1 Ga)and an averageεNd(0)value of-14.3.Samples from the hanging wall yield Neoproterozoic maximum depositional ages(~1000-600 ma)andεNd(0)values of-17.3.The contrasting detrital zircon ages show that the Batal thrust in the Kaghan valley is a major structural division.However,these data lack the characteristic detrital zircon age spectra andεNd(0)values of Greater and Lesser Himalayan rocks north and south of the main Central thrust in the central Himalaya,respectively.Therefore,either the Batal thrust is not the main Central thrust,or the main Central thrust in the Kaghan valley cuts a different structural level and carries rock units not seen outside the Hazara-Kashmir syntaxis.

Electromyographic and mechanomyographic Responses During Isokinetic Leg Extensions in Children Versus Adolescents

Purpose The purpose of this study was to examine peak torque(PT),mean power(mP),electromyographic(EmG)amplitude,mechanomyographic(mmG)amplitude,and neuromuscular efficiency from EmG and mmG amplitude(NmE_(EmG)and NmE_(mmG),respectively)across the velocity spectrum in children versus adolescents.methods Seventeen children(n=8 males,n=9 females)and 22 adolescents(n=11 males,n=11 females)participated.Participants completed maximal concentric isokinetic leg extensions at 60,120,180,240 and 300°/s.PT,mP,EmG amplitude,and mmG amplitude were quantified during all muscle actions.NmE_(EmG)and NmE_(mmG)were quantified by expressing PT relative to EmG and mmG amplitude,respectively,for each muscle action.Results PT,mP,NmE_(EmG),and NmE_(mmG)were greater for the adolescents than the children collapsed across sex and veloc-ity(P≤0.021).There were no significant differences for EmG amplitude(P≥0.051),while mmG amplitude increased significantly from 60 to 240°/s(Pm 240 to 300°/s(PmG and mmG amplitude(NmE_(EmG)and NmE_(mmG),respectively),in conjunction with measurements of torque and power production,may provide practitioners a unique method of examining growth and development-related augmentations in neuromuscular function during dynamic movements.

Autonomously self-propelled nanoswimmers represent the nextgeneration nano-devices for bio-and environmental technology.However,current nanoswimmers generate limited energy output and can only move in short distances and duration,thus are struggling to be applied in practical challenges,such as living cell transportation.Here,we describe the construction of biodegradable metal-organic framework based nanobots with chemically driven buoyancy to achieve highly efficient,long-distance,directional vertical motion to“find-and-fetch”target cells.Nanobots surface-functionalized with antibodies against the cell surface marker carcinoembryonic antigen are exploited to impart the nanobots with specific cell targeting capacity to recognize and separate cancer cells.We demonstrate that the self-propelled motility of the nanobots can sufficiently transport the recognized cells autonomously,and the separated cells can be easily collected with a customized glass column,and finally regain their full metabolic potential after the separation.The utilization of nanobots with easy synthetic pathway shows considerable promise in cell recognition,separation,and enrichment.

Effects of intermittent sprint and plyometric training on endurance running performance

Purpose:The purpose of this study was to compare the effects of intermittent sprint training and plyometric training on endurance running performance.methods:Fourteen moderately trained male endurance runners were allocated into either the intermittent sprint training group(n = 7)or the plyometric training group(n = 7).The preliminary tests required subjects to perform a treadmill graded exercise test,a countermovement jump test for peak power measurement,and a 10-km time trial.Training included 12 sessions of either intermittent sprint or plyometric training carried out twice per week.On completion of the intervention,post-tests were conducted.Results:Both groups showed significant reduction in weekly training mileage from pre-intervention during the intervention period.There were significant improvements in the 10-km time trial performance and peak power.There was also significant improvement in relative peak power for both groups.The 10-km time trial performance and relative peak power showed a moderate inverse correlation.Conclusion:These findings showed that both intermittent sprint and plyometric training resulted in improved 10-km running performance despite reduction in training mileage.The improvement in running performance was accompanied by an improvement in peak power and showed an inverse relationship with relative peak power.
