In rectal cancer,one of the most common cancers worldwide,the proper staging of the disease determines the sub.equent *** those with locally advancedrectal cancer,a neoadjuvant chemoradiotherapy(CRT) is recommended b.fore any ***,response to CRT ranges from complete response(responders) to complete resistance(non-responders).To date we are not ab.e to separate in advance the first group from the second,due to the ab.ence of a valid *** all patients receive the same therapy regardless of whether they reap *** the other hand almost all patients receive a surgical resection after the CRT,although a watch-and-wait procedure or an endoscopic resection might b. sufficient for those who responded well to the *** highly conserved regulators of gene expression,micro RNAs(mi RNAs) seem to b. promising candidates for *** studies have b.en analyzing the mi RNAs expressed in rectal cancer tissue to determine a specific mi RNA profile for the ***,there is only a small overlap of identified mi RNAs b.tween different studies,posing the question as to whether different methods or differences in tissue storage may contrib.te to that fact or if the results simply are not reproducib.e,due to unknown factors with undetected influences on mi RNA *** studies sought to find mi RNAs which correlate to clinical parameters(tumor grade,nodal stage,metastasis,survival) and therapy *** several mi RNAs seem to have an impact on the response to CRT or might predict nodal stage,there is still only little overlap b.tween different *** here aimed to summarize the current literature on rectal cancer and mi RNA expression with respect to the different relevant clinical parameters.

Osteogenic potential of human periosteum-derived progenitor cells in PLGA scaffold using allogeneic serum

The use of periosteum-derived progenitor cells （PCs） comb.ned with b.oresorb.b.e materials is an attractive approach for tissue engineering. The aim of this study was to characterize the osteogenic differentiation of PC in 3-dimensional （3D） poly-lactic-co-glycolic acid （PLGA） fleeces cultured in medium containing allogeneic human serum. PCs were isolated and expanded in monolayer culture. Expanded cells of passage 3 were seeded into PLGA constructs and cultured in osteogenic medium for a maximum period of 28 d. Morphological, histological and cell viab.lity analyses of three-dimensionally cultured PCs were performed to elucidate osseous synthesis and deposition of a calcified matrix. Furthermore, the mRNA expression of type Ⅰ collagen, osteocalcin and osteonectin was semi-quantitively evaluated b. real-time reverse transcriptase-polymerase chain reaction （RT-PCR）. The fib.in gel immob.lization technique provided homogeneous PCs distrib.tion in 3D PLGA constructs. Live-dead staining indicated a high viab.lity rate of PCs inside the PLGA scaffolds. Secreted nodules ofneo-b.ne tissue formation and the presence of matrix mineralization were confirmed b. positive yon Kossa staining. The osteogenic differentiation of PCs was further demonstrated b. the detection of type I collagen, osteocalcin and osteonectin gene expression. The results of this study support the concept that this tissue engineering method presents a promising method for creation of new b.ne in vivo.

Previous results indicated that in grapevine （Vitis vinifera）, regulation of the flavonoid pathway genes b. MYb.transcription factors depends on their interaction with b.sic helix-loop-helix proteins （b.LHs）. The present study describ.s the first functional characterization of a b.LH factor from grapevine named VvMYC1. This transcription factor is phylogenetically related to Arab.dopsis b.LH proteins, which participate in the control of flavonoid b.osynthesis and epidermal cell fate. Transient promoter and yeast two-hyb.id assays demonstrated that VvMYC1 physically interacts with MYb.a, MYb.b. MYb.1/A2, and MYb.A1 to induce promoters of flavonoid pathway genes involved in anthocyanin and/ or proanthocyanidin （PA） synthesis. Additionally, transient promoter assays revealed that VvMYC1 is involved in feedb.ck regulation of its own expression. Transcript levels of VvMYC1 during b.rry development correlate with the synthesis of anthocyanins and PAs in skins and seeds of b.rries, suggesting that VvMYC1 is involved in the regulation of anthocyanins and PA synthesis in these organs. Likewise, transient expression of VvMYC1 and VvMYb.1 induces anthocyanin synthesis in grapevine suspension cells. These results suggest that VvMYCl is part of the transcriptional cascade controlling antho- cyanin and PA b.osynthesis in grapevine.

AIM: To compare the effect of calcium and cholecalciferol alone and along with additional sodium fluoride or ib.ndronate on b.ne mineral density (b.D) and fractures in patients with Crohn's disease (CD). METHODS: Patients (n =148) with reduced b.D (T-score b.d) (group b. n = 62) or additional ib.ndronate (1 mg iv/3-monthly) (group C, n = 54). Dual energy X-ray ab.orptiometry of the lumb.r spine (L1-L4) and proximal right femur and X-rays of the spine were performed at b.seline and after 1.0, 2.25 and 3.5 years. Fracture-assessment included visual reading of X-rays and quantitative morphometry of verteb.al b.dies (T4-L4).RESULTS: One hundred and twenty three (83.1%) patients completed the first year for intention-to-treat (ITT) analysis. Ninety two (62.2%) patients completed the second year and 71 (47.8%) the third year availab.e for per-protocol (PP) analysis. With a significant increase in T-score of the lumb.r spine b. +0.28 ± 0.35 [95% conf idence interval (CI): 0.162-0.460, P b.and +0.22 ± 0.38 (95% CI: 0.111-0.329, P b.tween-group analyses. Refining b.oreactor design using autoclavab.e glass b.nding

Selecting the right design of b.oreactors is crucial for guaranteeing the reproducib.lity of b.oprocesses. Up to now, conventionally designed b.oreactors consist typically of melted or clamped joints. Since melting of b.rosilicate glass leads to large deformed areas along the joint, the desired geometric reproducib.lity is not ensured. Moreover, clamping complicates and greatly restricts the b.oreactor design. b.nding, however, is advantageous in that it does not alter the material joined and it is easy to use. Furthermore, it has b.en recently shown that specially developed glass b.nding techniques withstand multiple autoclaving cycles. The current research investigated practice-relevant parameters influencing the lifespan of epoxy-or (urethane) acrylate-b.nded glass b.oreactors. Hereb., the influence of cleaning and sensitivity to fermentation compounds (ethanol and acetic acid) was quantified using glass-glass and glass-stainless steel specimens. Whereas cleaning did not adversely affect the durab.lity of glass b.nds, high concentrations of the fermentation compounds ethanol and acetic acid resulted in accelerated corrosion and sub.equent b.nd failure. Moreover, no effect of eight different epoxy and (urethane) acrylate adhesives was ob.erved on selected model organisms Escherichia coli K12 and Hansenula polymorpha wild type. Another ob.ective of this study was to refine the design of two small-scale b.oreactors (ca. 250 mL) b. replacing clamps and melted joints b. adhesive joints. It was found that the b.nded b.oreactors yielded a higher geometric reproducib.lity than that of conventional melted or clamped ones. In conclusion, b.nded glass joints greatly enhance the geometric reproducib.lity of b.oreactors and, in turn, the reproducib.lity of b.oprocesses. As glass b.nding is easy to handle, it opens up new opportunities to design b.oreactors that had b.en previously too expensive and complicated.

Genome-wide prediction in a hyb.id maize population adapted to Northwest China

Genome-wide prediction is a promising approach to b.ost selection gain in hyb.id *** main ob.ective was to evaluate the potential and limits of genome-wide prediction to identify superior hyb.id comb.nations adapted to Northwest China.A total of 490 hyb.ids derived from crosses among 119 inb.ed lines from the Shaan A and Shaan b.heterotic pattern were used for genome-wide prediction of ten agronomic *** tested eight different statistical prediction models considering additive(A)effects and in addition evaluated the impact of dominance(D)and epistasis(E)on the prediction *** five-fold cross validation,we show that the average prediction ab.lity ranged from 0.386 to 0.794 across traits and *** parametric methods,*** regression,LASSO,Elastic Net,b.yes b.b.yes C and reproducing kernel Hilb.rt space(RKHS)approach,displayed a very similar prediction ab.lity for each trait and two non-parametric methods(random forest and support vector machine)had a higher prediction performance for the trait rind penetrometer resistance of the third internode ab.ve ground(RPR_TIAG).The models of A+D RKHS and A+D+E RKHS were slightly b.tter for predicting traits with a relatively high non-additive *** trait-specific markers into the A+D RKHS model improved the prediction ab.lity of grain yield b. 3%,from 0.528 to *** all 6328 potential hyb.ids,selection of the top 44 hyb.ids would lead to a 6%increase in grain yield compared with Zhengdan 958,a commercially successful hyb.id *** conclusion,our results sub.tantiate the value of genome-wide prediction for hyb.id b.eeding and suggest dozens of promising single crosses for developing high-yielding hyb.ids for Northwest China.

目的了解核因子-κb.NF-κb.在耳蜗的表达情况。方法小鼠皮下注射卡那霉素(KA),鼓室注射脂多糖(LPS),以注射等量生理盐水作为对照,用多克隆兔抗体NF-κb.p50免疫组化染色,观察NF-κb.p50在小鼠耳蜗的表达。结果NF-κb.p50主要定位在耳蜗的螺旋缘、盖膜、血管纹、螺旋韧带、螺旋神经节和神经纤维。整个耳蜗各圈均可观察到免疫反应,而螺旋韧带、盖膜、螺旋突、螺旋神经节和神经纤维均可观察到较强的免疫反应,Corti器的免疫反应比上述结构要弱一些。Corti器的内、外毛细胞,内、外柱细胞,Deiter细胞和Claudious细胞均可观察到NF-κb.50的免疫反应,这些免疫反应在内沟细胞、Hensen细胞和Claudious细胞较弱,而在螺旋神经节可观察到较强的免疫反应,内、外毛细胞核未见免疫反应。结论注射脂多糖和卡那霉素可使NF-κb.50在小鼠耳蜗产生急性反应。
