Adenosine 5'triphosphate transport and accumulation during the cold preservation of rat hepatocytes in University of Wisconsin solution

AIM: We used isolated hepatocytes to investigate how different concentrations of ATP in the University of Wisconsin (UW) solution affected both cellular ATP content and cell viability during the cold storage and the rewarming step. The mechanism involved in ATP transport and accumulation in hypothermia was also ***: The cells were preserved up to 72 h in different conditions: UW solution without ATP (a-group),UW+5 mmol/L ATP (b-group), and UW+10 mmol/L ATP (c-group). The ATP content and the cell viability (LDH release) were determined during the cold storage and the rewarming step. In the groups a and c, the respiratory function of the cells at rewarming was studied. In addition,the cell volume of hepatocytes and the mechanism involved in ATP transport and accumulation were assessed. The extracellular degradation of exogenous nucleotides during transport experiments was investigated by a HPLC ***: After three days of cold storage a loss of cellular ATP content was observed in hepatocytes preserved either without nucleotides (a-group) or with 5 mmol/L ATP (b-group). In contrast, 10 mmol/L ATP (c-group) was able to maintain a normal ATP cellular content, with only a 6% diminution after 72 h of cold storage. The respiratory function was significantly different in hepatocytes preserved with 10 mmol/L ATP than without ATP. no significant change was detected for the three groups in cellular volume during the cold storage. We also report that the time course accumulation of [3H]-ATP by cold stored hepatocytes is a rapid process that is completed after 180 s with linear dependence on the extracellular ATP concentration (linear fitting results in a slope of 0.5624±0.1179 mmol/L ATP intracell/mmol/L ATP extracell).COnCLUSIOn: Our results show that, during hypothermic storage in UW solution, hepatocytes are permeable to ATP by a diffusive mechanism. Pharmacodynamic Study of Parallel Groups Comparing the Effect of Rivaroxaban 20 Mg (Laboratorios Leti, S.A.v.) vs Rivaroxaban 20 Mg (Bayer Laboratories) on Prothrombin Time

Background: The prevalence of both atrial fibrillation (FA) and diabetes mellitus (DM) is increasing and they often occur together and constitute a high risk of thrombosis. Rivaroxaban is a Factor Xa inhibitor with a rapid onset and disappearance of action after oral administration;it acts by inhibiting the active form of the coagulation factor. In order to reflect the effect of the action of Rivaroxaban, we used the prothrombin time (PT);however, it′s not the most accurate, but it is the one available in our community. Methods: This was a prospective, randomized, analyst-blinded, parallel group clinical study to verify the efficacy of Rivaroxaban Leti 20 mg (RL) (12 volunteers vs Rivaroxaban Bayer 20 mg (RB) (13 volunteers). The variables were determination of PT and Partial Thromboplastin Time (aPTT) at baseline and at 24, 48 and 72 hours after administering a daily dose of 20 mg for three days. The determination was carried out with the IDG method (Integrated Diagnostics Group Sanzay Corporation) with an International Sensitivity Index (ISI) of 1.17 PT and aPTT were taken before the first dose, and then, every day during the next 3 days, three hours after the ingestion of their daily dose at 7 am. Results: The 25 healthy volunteers were similar in age, BMI, and SBP/DBP level with a greater number of men in the Bayer group. The efficacy of rivaroxaban was similar in both groups with prolongation of PTT to the 2nd day of treatment with PT, and percentage changes from baseline (14.46 ± 0.97 for RB vs 14.17 ± 0.94 RL p: 0.45), PTT results and percentage changes from the base (RB: 34 ± 4.53 RL: 33.46 ± 2.82). The safety of rivaroxaban was good in both groups with no serious adverse events. The equivalence in the logarithmically transformed PT result (ln) on day two, Mean and CI (90%) 99.2 (94.4-104) and 100 (99.5-100.8);neither the means nor the 90% confidence intervals of the PT variable transformed logarithmically to ensure its normality, were far from the 80%-125%

In-hand manipulation of a circular dynamic object by soft fingertips without angle measurement

A multi-fingered robotic hand with curved fingertips enables contact re-positioning without reattaching at the expense of fingertip rolling. This rolling stands for a characteristic that facilitates dexterous manipulation but results in an algebraically complex dynamic model subject to such constraints. The hemispherical shape of fingertips allows a dexterous manipulation when controlling the tangent forces, which are essential to rotate object. However, the measurement of the object angle in practice requires tactile-optical sensing. In this paper, considering robotic fingers with curved soft tips, we propose a feedback control that ensures optimal dynamical grasping of a circular rigid object. It is shown that the collaboration of the contact forces, to get a minimum pose of internal forces, and the tangential forces, to induce the conditions for assuring the grasp closure, is necessary to get a skillful manipulation. In this case, the orientation control of a circular object to the desired angle while avoiding direct measurement of the object angle is presented. Stability conditions of the system are presented in the sense of stability-in-the-manifold. Finally, representative simulations are shown and discussed.

Improving surface integrity when drilling CFRPs and Ti-6Al-4v using sustainable lubricated liquid carbon dioxide

In the quest for decreasing fuel consumption and resulting gas emissions in the aeronautic sector,lightweight materials such as Carbon Fiber Reinforced Polymers(CFRPs)and Ti-6Al-4v alloys are being *** materials,with excellent weight-to-strength ratios,are widely used for structural applications in aircraft *** date,several studies have been published showing that the use of metalworking fluids(MWFs),special tool geometries,or advanced machining techniques is required to ensure a surface quality that meets aerospace component *** MWFs pose a number of environmental and worker health hazards and also degrade the mechanical properties of CFRPs due to water absorption in the ***,a transition to more environmentally friendly cooling/lubrication techniques that prevent moisture problems in the composite is *** research shows that lubricated LCO_(2) is a viable option to improve the quality of drilled CFRP and titanium aerospace components compared to dry machining,while maintaining clean work *** results show that the best combination of tool geometry and cooling conditions for machining both materials is drilling with Brad point drills and lubricated LCO_(2).Drilling under these conditions resulted in a 90%improvement in fiber pullout volume compared to dry machined CFRP *** addition,a 33% reduction in burr height and a 15% improvement in surface roughness were observed compared to dry drilling of titanium.

According to World Health Organization(WHO)estimates,in 2020,there were 1.8 million human immunodeficiency virus(HIv)-infected children below 15 years old living with HIv and 150,000 children who acquired the infection[1].Every year,110,000 children die as a consequence of acquired immune deficiency syndrome-related causes[2].As in adults,optimal management of infected children depends on early diagnosis and treatment with antiretroviral drugs based on their viral genotype.

Anisotropic iron-doping patterns in two-dimensional cobalt oxide nanoislands on Au(111)

An in teg rated approach combi ning den sity functional theory(DFT)calculati ons and atomic resol utio n sea nning tun neling microscopy(STM)is used to study well-defined iron-doped cobalt oxide nanoislands supported on Au(111).The focus is on the structure and distribution of Fe dopants within these nanoislands of CoO as a function of Fe to Co *** DFT and STM results agree strongly and complement each other to allow for a more complete un dersta nding of the dopa nt structure trends on the nano *** Fe as a marker,we first find that the stacki ng seque nee of the moire structure of the host cobalt oxide nano islands can be ide ntified un ambiguously through a combinati on of DFT and *** the distinct con trast of the embedded Fe dopa nt atoms as observed with atom-resolved STM,we find correlatio ns betwee n Fe dopa nt positi on and the CoO/Au(111)moire patter n at varyi ng Fe dopa nt den *** n of Fe-dopa nt clusters within the nano islands is investigated in detail through DFT and found to agree with the dopant patter ns observed in *** find that the structural effects of Fe dopants throughout the nanoislands with the basal planes and the two types of edges—the oxygen and metal edges—have different *** DFT calculations and STM images show a strong preferenee for Fe dopants to be located directly on or near the oxygen edge of the nanoislands as opposed to being directly on or n ear the metal *** n together,our results illustrate that Fe dopa nt incorporati on and distributi on within CoO nanoislands are highly an isotropic and gover ned by both the moire structure of the basal planes as well as nano-size effects present at the under-coordinated edges of different local geometry and chemistries.

Assessment of the exploitable biomass of thread herring(Opisthonema spp.)in northwestern Mexico

In recent years,the small pelagic fishery on the Pacific northwest coast of Mexico has significantly increased fishing pressure on thread herring Opisthonema *** fishery is regulated using a precautionary approach(acceptable biological catch(ABC)and minimum catch size).However,due to fishing dynamics,fish aggregation habits and increased fishing mortality,periodic biomass assessments are necessary to estimate ABC and assess the resource *** Catch-MSY approach was used to analyze historical series of thread herring catches off the western Baja California Sur(BCS,1981–2018)and the Gulf of California(GC,1972–2018)to estimate exploitable biomass and target reference points in order to obtain catch *** to the results,in GC,the maximum biomass reached in 1972(at the beginning of fishery)and minimum biomass reached in 2015;the estimated exploitable biomass for 2019 was 42.2×10^(4) t;and the maximum sustainable yield(MSY)was 15.4×10^(4) *** the western BCS coast,the maximum biomass was reached in 1981(at the beginning of fishery)and minimum biomass was reached in 2017;the estimated exploitable biomass for 2019 was 3.2×10^(4) t;and the MSY was 1.2×10^(4) *** stocks showed a decrease in biomass over the past years and were currently near to point of full *** results suggest that the use of the Catch-MSY method is suitable to obtain annual biomass estimates,in order to establish an ABC,to know the current state of the resource,and to avoid overcoming the potential recovery of the stocks.
