Resistance to pentavalent antimonial drugs and the lack of vaccines make it urgent to find novel therapeutic options to treat Leishmaniasis, a tropical disease caused by the Leishmania protozoan parasite. The study reported here is to investigate if Streptomycin, an aminoglycoside, and Amphotericin B, the second-line treatment drug, exhibit antileishmanial activity through a similar mechanism. By using mOE (molecular Operating Environment), we performed molecular docking studies on these drugs binding to a range of targets including ribosome targets in Leishmania and H. sapiens. Our study shows that the two drugs do not bind to the same pockets in Leishmania targets but to the same pockets in the human ribosome, with some differences in interactions. moreover, our 2D maps indicated that Amphotericin B binds to the A-site in the human cytoplasmic ribosome, whereas streptomycin does not.

青藏高原及其周缘褶皱-冲断带变形方式和时限是验证高原形成众多大陆岩石圈变形动力学端员模型的关键。近年发展起来的断层泥自生伊利石K-Ar定年技术为精确限定褶皱-冲断带变形时限提供了有效手段。鲜水河—安宁河—小江断裂系作为伴随印度-欧亚板块碰撞造山和高原物质侧向挤出过程形成的大型左旋走滑断裂系,其变形过程可为解译印度-欧亚板块碰撞远程应力向东传递方式提供关键证据。本文选取安宁河断裂冕宁—西昌段作为研究对象,在详细构造解析以明确其构造运动学特征的基础上,开展了断层泥自生伊利石K-Ar定年研究,以期精确限定安宁河断裂脆性变形的时间。构造解析表明安宁河断裂在近EW向挤压作用下经历了冲断变形,断层带发育碎斑岩和断层泥,指示为脆性变形。安宁河断裂不同粒级断层泥样品的黏土矿物学和伊利石K-Ar定年分析揭示,随着样品粒级减小,高温2m1型伊利石含量相对低温1m/1md型伊利石含量逐渐减少,其K-Ar年龄总体呈变年轻趋势,表明不同粒级伊利石K-Ar年龄是由碎屑2m1型和自生1m/1md型两个多型端员组成的混合年龄。伊利石年龄分析获得自生1m/1md型伊利石的年龄为(42.6±9.4)ma,表明安宁河断裂经历了始新世中期的冲断变形。结合前人构造变形、沉积学、低温热年代学和古地磁等研究,我们认为始新世中期青藏高原腹地及其周缘褶皱-冲断带发生准同期的构造挤压变形,其动力学机制可能与印度-欧亚板块硬碰撞以及青藏高原地块的陆内俯冲所导致的先存构造带活化有关。始新世中期安宁河断裂发生冲断变形直接证明印度-欧亚板块碰撞初期的远程应力已传递至青藏高原东南缘地区。

Formation potential and analysis of 32 regulated and unregulated disinfection by-products: Two new simplified methods

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic matter naturally present in water, leading to the formation of disinfection by-products(DBPs). multi-analyte methods based on mass spectrometry(mS) are preferred to quantify multiple DBP classes at once however, most require extensive sample pre-treatment and significant resources. In this study, two analytical methods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap(P&T) coupled with gas chromatography mass spectrometry(GC-mS) method was optimized that automated sample pre-treatment and analyzed volatile and semi-volatile compounds, including trihalomethanes(THms), iodinated trihalomethanes(I-THms), haloacetonitriles(HANs), haloketones(HKTs) and halonitromethanes(HNms). LOQs were between 0.02-0.4 μg/L for most DBPs except for 8 analytes that were in the low μg/L range. A second method with liquid chromatography(LC) tandem mass spectrometry(mS/mS) was developed for the quantification of 10 haloacetic acids(HAAs) with a simple clean-up and direct injection. The LC-mS/mS direct injection method has the lowest detection limits reported(0.2-0.5 μg/L). Both methods have a simple sample pre-treatment, which make it possible for routine analysis. Hyperchlorination and uniform formation conditions(UFC) formation potential tests with chlorine were evaluated with water samples containing high and low TOC. Hyperchlorination formation potential test maximized THms and HAAs while UFC maximized HANs. Ascorbic acid was found to be an appropriate quencher for both analytical methods. Disinfected drinking water from four water utilities in Alberta, Canada were also evaluated.

Predicting Ailanthus altissima presence across a managed forest landscape in southeast Ohio

Background: The negative impacts of the exotic tree, Ailanthus altissima(tree-of-heaven, stink tree), is spreading throughout much of the Eastern United States. When forests are disturbed, it can invade and expand quickly if seed sources are ***: We conducted studies at the highly dissected Tar Hollow State Forest(THSF) in southeastern Ohio USA,where Ailanthus is widely distributed within the forest, harvests have been ongoing for decades, and prescribed fire had been applied to about a quarter of the study area. Our intention was to develop models to evaluate the relationship of Ailanthus presence to prescribed fire, harvesting activity, and other landscape characteristics, using this Ohio location as a case study. Field assessments of the demography of Ailanthus and other stand attributes(e.g., fire, harvesting, stand structure) were conducted on 267 sample plots on a 400-m grid throughout THSF,supplemented by identification of Ailanthus seed-sources via digital aerial sketch mapping during the dormant season. Statistical modeling tools Random Forest(RF), Classification and Regression Trees(CART), and maxent were used to assess relationships among attributes, then model habitats suitable for Ailanthus ***: In all, 41 variables were considered in the models, including variables related to management activities, soil characteristics, topography, and vegetation structure(derived from LiDAR). The most important predictor of Ailanthus presence was some measure of recent timber harvest, either mapped harvest history(CART) or LiDARderived canopy height(maxent). Importantly, neither prescribed fire or soil variables appeared as important predictors of Ailanthus presence or absence in any of the models of the ***: These modeling techniques provide tools and methodologies for assessing landscapes for Ailanthus invasion, as well as those areas with higher potentials for invasion should seed sources become available. Electrodeposition from a lyotropic liquid crystal template medium was used to produce nanostructured platinum microelectrodes with high specific surface area and high mass transport efficiency. Compared to polished and conventional platinized microelectrodes, well-ordered nanostructured platinum microelectrodes exhibited enhanced electrocatalytic properties for oxygen and ascorbic acid, whilst well-ordered nanostructured platinum microelectrodes offered improved electrocatalytic properties for oxygen reduction compared to disordered nanostructured platinum microelectrodes.

Angling of hybrid sunfish from a hatchery pond with potential implications for community fisheries

Hatcheries are frequently called upon to produce catchable-sized fish for stocking community fishing *** attributes of fish selected for stocking into community ponds are that they are easy to produce in a hatchery system to sizes anglers are interested in catching and they provide anglers with high catch rates once *** sunfish[male Bluegill(Lepomis macrochirus)×female Green Sunfish(***)]have attributes that potentially make them attractive for use in community fishing *** assessed initial angler catch rates of 100 stocked hybrid sunfish in a 0.12-ha hatchery pond and after being subjected to angling(four,1-hr catch-and-release fishing events with five anglers).We also investigated whether catch rates would change following a supplemental stocking of an additional 100 hybrid sunfish(four,1-hr catch-and-release fishing events with five anglers).The anal fin of each fish in the second stocking was hole punched to differentiate them from those of the first stocking and fish caught by anglers were hole punched in the caudal fin each time they were caught before being released back into the *** catch rates were highest during initial fishing events that followed stocking(9.2 fish/angler hr and 18.0 fish/angler hr)and substantially declined in subsequent events(≤3.4 fish/angler hr).Catches of the newly stocked fish and previously stocked fish contributed to the high catch following the supplemental ***(80%)of the fish were caught in the first 30 min of each event and 45%were caught during the first 10 *** were able to catch 88%of the fish from the first stocking and 67%from the second stocking at least *** mortality occurred during the study as all fish were recovered when the pond was *** results suggest that hybrid sunfish will potentially provide high initial catch rates following stocking into community ponds,but managers should expect reduced catch rates following initial fishing even without h
