Octameri. hemoglobi.s have been developed by the i.troducti.n of surface cystei.es i. ei.her the alpha or beta chai.. Ori.i.ally desi.ned as a blood substi.ute, we report here the structure and li.and bi.di.g functi.n;i. addi.i.n the i.teracti.n wi.h haptoglobi. was studi.d. The recombi.ant Hbs (rHbs) wi.h mutati.ns alpha Asn78Cys or beta Gly83Cys spontaneously form octamers under condi.i.ns where the cystei.es are oxi.i.ed. Oxygen bi.di.g curves and CO ki.eti. studi.s i.di.ate a correct allosteri. transi.i.n of the tetramers wi.hi. the octamer. Crystallographi. studi.s of the two rHbs show two di.ulfi.e bonds per octamer. Reduci.g agents may provoke di.soci.ti.n to tetramers, but the octamers are stable when mi.ed wi.h fresh human plasma, i.di.ati.g that the reducti.n by plasma i. slower than the oxi.ati.n by the di.solved oxygen, consi.tent wi.h an enhanced stabi.i.y. The octameri. rHbs were also mi.ed wi.h a soluti.n of haptoglobi. (Hp), whi.h bi.ds the di.ers of Hb: there was li.tle i.teracti.n for i.cubati.n ti.es of 15 mi.;however, on longer ti.escales a complex was formed. Dynami. li.ht scatteri.g was used to follow the i.teracti.n of Hp wi.h the alpha Asn78Cys octamer duri.g 24 hours;a transi.i.n from a si.ple complex of 15 nm to a fi.al si.e of 60 nm was observed. The results i.di.ate a speci.i. ori.ntati.n of the αβ di.ers may be of i.portance for the bi.di.g to haptoglobi..
