Germline activating variants in WWP1,which encodes an E3 ubiquitin ligase that antagonizes PTEN tumor suppressive function,have been proposed as an alternative mechanism of PTEN inactivation in PTEN-hamartoma-tumor syndrome(PHTS)-like patients with wildtype PTEN.1 More specifically,heterozygous,potentially activating wwP1 variants were first identified by Lee et al in patients affected with gastrointestinal oligopolyposis,including adenomatous,hyperplastic/serrated,and hamartomatous polyps,and occasionally with colorectal cancer(Table 1).Subsequently,based on the PHTS phenotypic features,wWP1 mutational screening was performed in patients with thyroid nodules,2 or normocephalic autism spectrum disorder(ASD),3 where germline WWP1 variants were also identified(Table S1).

In this work the influence of the density of the molten metal on the emergence of the raining phenomenon in the horizontal centrifugal casting process is numerically studied. Transient 2D numerical simulations were carried out using Computational Fluid Dynamics software. Three molten metals with different density, namely aluminum, iron and lead, and three angular frequencies, namely 50, 66 and 77 rad/s were considered. It is found that the density of the molten metal significantly affects the emergence, transient or permanent, of the rain phenomenon. However, the magnitude and duration of the rain phenomenon depend on the angular frequency of the rotating mold. Likewise, since gravitational forces affect the metal according to its density, the value of the critical rotation speed of the mold is also affected.

The BETA application-specific integrated circuit(ASIC)is a fully programmable chip designed to amplify,shape and digitize the signal of up to 64 Silicon photomultiplier(SiPM)channels,with a power consumption of approximately~1 mW/*** to its dual-path gain,the BETA chip is capable of resolving single photoelectrons(phes)with a signal-to-noise ratio(SNR)>5 while simultaneously achieving a dynamic range of~4000 ***,BETA can provide a cost-effective solution for the readout of SiPMs in space missions and other applications with a maximum rate below 10 *** this study,we describe the key characteristics of the BETA ASIC and present an evaluation of the performance of its 16-channel version,which is implemented using 130 nm *** ASIC also contains two discriminators that can provide trigger signals with a time jitter down to 400 ps FWHM for 10 *** linearity error of the charge gain measurement was less than 2%for a dynamic range as large as 15 bits.

The temperature drop of molten metal flowing in open channels is numerically determined. Rectangular, trapezoidal and triangular geometries are considered. The overall heat transfer coefficients for the bottom, side walls and free surface of the channel have been taken from the literature. For each geometry, the volumetric flow rate, mean residence time and temperature drop as a function of the channel inclination angle were determined. The rectangular and trapezoidal geometries present the smallest temperature drops, while the triangular geometry presents the greatest temperature drop. The factors that most affect this drop are the value of the free surface area of the channel, and the average residence time of the molten metal in the channel.

The performance of bridge projects in Kenya is poor in terms of completion by schedule, cost, and quality (scope). Yet, there is less evidence of empirical research on what factors contribute to this outcome. This study aimed to bridge this gap by examining the effects of contractor-related factors on the performance of bridge construction projects in Kenya through a case study of the Bridge projects Implemented by the Kenya National Highway Agency (KeNHA). The theory of constraints (ToC) was adopted as its theoretical framework. Descriptive research was used, and the target population was 18 bridge construction projects, which were the units of analysis from 2012 to 2022. In each of these projects, 18 respondents, namely clients, consultants, contractors, engineers, environment and social guards, project managers, stakeholders, subcontractors, technical advisors, and inspectors, were included in a target population of 144 respondents. A census was conducted and a structured questionnaire was administered from which a response rate of 68% was achieved. The information was analyzed using descriptive, correlation, and multiple linear regression analysis. The contractor-related factors considered in the study were staff and management factors. The findings indicated that staff and management factors had a positive and significant outcome on performance of bridge construction projects. The study recommends continuous training and a safe learning environment for staff to improve their skills and performance in future projects. The study also recommends that a special category for bridge contractors be created in Kenya’s National Construction Authority rankings to ensure that only qualified contractors implement the Bridge projects.

Deep phenotyping and genetic characterization of individuals are fundamental to assessing the metabolic status and determining nutrition-specific requirements. This study aimed to ascertain the utmost effectiveness of personalized interventions by aligning dietary adjustments with both the genotype and metabolotype of individuals. Therefore， we assessed here the usefulness of a polygenic score (PGS) characterizing a potential pro-inflammatory profile (PGSi) as a nutrigenetic tool to discern individuals from the Danish PREVENToMICS cohort that could better respond to precision nutrition (PN) plans， specifically targeted at counteracting the low-grade inflammatory profile typically found in obesity. The cohort followed a PN plan to counteract the pro-inflammatory profile (PNi group) or generic dietary recommendations (Control) for 10 weeks. PGSi was applied for genetic stratification (Low/High). The effects of the intervention on anthropometrics and biomarkers related to inflammatory profile and carbohydrate metabolism were assessed. Around 30% of subjects had a high genetic predisposition to pro-inflammatory status (high-PGSi). These individuals demonstrated the most effective response to the dietary plan， experiencing improved body composition， with significant decreases in body weight (△: -4.84%; p=0.039) and body fat (△: -4.86%; p=0.007)， and beneficial changes in pro- and anti-inflammatory biomarkers， with significant increases in IL-10 (△: 71.3%; p=0.025) and decreases in TNF-α (△: -3.0%; p=0.048)， CRP (△: -31.1%)， ICAM1 (△: -5.8%)， and MCP1 (△: -4.2%) circulating levels， compared to low-PGSi individuals. Both phenotypic and genetic stratification contributed to a better understanding of metabolic heterogeneity in response to diet. This approach allows for refinement of the prediction of individual requirements and potentially for better management of obesity.

Darcy’s law is widely used to describe the flow in porous media in which there is a linear relationship between fluid velocity and pressure gradient. However, it has been found that for high numbers of Reynolds this law ceases to be valid. In this work, the Ergun equation is employed to consider the non-linearity of air velocity with the pressure gradient in casting sands. The contribution of non-linearity to the total flow in terms of a variable defined as a non-Darcy flow fraction is numerically quantified. In addition, the influence of the shape factor of the sand grains on the non-linear flow fraction is analyzed. It is found that for values of the Reynolds number less or equal than 1, the contribution of non-linearity for spherical particles is around 1.15%.
