Inhibition effect of curcumin on UVb-induced secretion of pro-inflammatory cytokines from corneal limbus epithelial cells

~ AIM： To study the effects of curcumin on the secretion of interleukin （IL） -6 and IL-8 by corneal limbus epithelial cells. METHODS： Human corneal Iimbus epithelial cells were isolated and cultured from donor eyes and irradiated by UVb at different dosages with or without curcumin. MTT test was used for studying the effects of UVb and curcumin on the cell viability. The role of mitogen-activated protein kinase （MAPK） and nuclear factor-kappa b （NF-Kb） pathways on the UVb-induced secretion of IL.6 and IL-8 were tested by addition of their inhibitors to the culture with or without UVb-radiation. Levels of various signal pathways, IL-6 and IL-8 in the cells and in the conditioned culture medium were measured by ELISA analysis. RESULTS： UVb at 20 mJ/cm2 or less and curcumin at 20 IJmol/L or less did not affect the cell viability of culturedlimbus epithelial cells （P〉0.05）. UVb irradiation at 10 and 20 mJ/cm2 induced a significant increase of secretion of IL-6 and IL-8 and upregulated NF-Kb and phosphorylated MAPK pathways of cultured limbus epithelial cells （P〈0.05）. Various signal pathway inhibitors, including SP600125 （JNK inhibitor）, Sb203580 （p38 MAPK inhibitor） and bAY11-7082 （NF-Kb inhibitor） significantly decreased the UVb-induced secretion of IL-6 and IL-8 secretion （P〈0.05）. Curcumin at 5-20 pmol/L significantly inhibited UVb- induced secretion of IL-6 and IL-8 by limbus epithelial cells in a dose-dependent manner; while curcumin alone did not affect the secretion of IL-6 and IL-8. The upregulation of NF-Kb and MAPK pathways induced by UVb treatment was significantly inhibited by curcumin, suggesting that NF-Kb and MAPK pathways are involved in the inhibitory effect of curcumin on UVb-induced production of IL-6 and IL-8.

While stromal interactions are essential in cancer adaptation to hormonal therapies,the effects of bone stroma and androgen deprivation on cancer progression in bone are poorly understood.Here,we tissue-engineered and validated an in vitro microtissue model of osteoblastic bone metastases,and used it to study the effects of androgen deprivation in this microenvironment.The model was established by culturing primary human osteoprogenitor cells on melt electrowritten polymer scaffolds,leading to a mineralized osteoblast-derived microtissue containing,in a 3D setting,viable osteoblastic cells,osteocytic cells,and appropriate expression of osteoblast/osteocyte-derived mRNA and proteins,and mineral content.Direct co-culture of androgen receptordependent/ independent cell lines (LNCaP,C4-2b,and PC3) led cancer cells to display functional and molecular features as observed in vivo.Co-cultured cancer cells showed increased affinity to the microtissues,as a function of their bone metastatic potential.Cocultures led to alkaline phosphatase and collagen-I upregulation and sclerostin downregulation,consistent with the clinical marker profile of osteoblastic bone metastases.LNCaP showed a significant adaptive response under androgen deprivation in the microtissues,with the notable appearance of neuroendocrine transdifferentiation features and increased expression of related markers (dopa decarboxylase,enolase 2).Androgen deprivation affected the biology of the metastatic microenvironment with stronger upregulation of androgen receptor,alkaline phosphatase,and dopa decarboxylase,as seen in the transition towards resistance.The unique microtissues engineered here represent a substantial asset to determine the involvement of the human bone microenvironment in prostate cancer progression and response to a therapeutic context in this microenvironment.

Geriatric trauma constitutes an increasingly recognized problem. Aging results in a progressive decline in cellular function which leads to a loose of their capacity to respond to injury. Some medications commonly used in this population can mask or blunt the response to injury. Falls constitute the most common cause of trauma and the leading cause of trauma-related deaths in this population. Falls are complicated by the widespread use of antiplatelets and anticoagulants, especially in patients with brain injury. Under-triage is common in this population. Evaluation of frailty could be helpful to solve this issue. Appropriate triaging and early aggressive management with correction of coagulopathy can improve outcome. Limitation of care and palliative measures must be considered in cases with a clear likelihood of poor prognosis.

From leaf to whole-plant water use efficiency(WUE)in complex canopies:Limitations of leaf WUE as a selection target

Plant water use efficiency(WUE) is becoming a key issue in semiarid areas, where crop production relies on the use of large volumes of water. Improving WUE is necessary for securing environmental sustainability of food production in these areas. Given that climate change predictions include increases in temperature and drought in semiarid regions,improving crop WUE is mandatory for global food production. WUE is commonly measured at the leaf level, because portable equipment for measuring leaf gas exchange rates facilitates the simultaneous measurement of photosynthesis and transpiration. However,when those measurements are compared with daily integrals or whole-plant estimates of WUE, the two sometimes do not agree. Scaling up from single-leaf to whole-plant WUE was tested in grapevines in different experiments by comparison of daily integrals of instantaneous water use efficiency [ratio between CO2assimilation(AN) and transpiration(E); AN/E] with midday AN/E measurements, showing a low correlation, being worse with increasing water stress. We sought to evaluate the importance of spatial and temporal variation in carbon and water balances at the leaf and plant levels. The leaf position(governing average light interception) in the canopy showed a marked effect on instantaneous and daily integrals of leaf WUE. Night transpiration and respiration rates were also evaluated, as well as respiration contributions to total carbon balance. Two main components were identified as filling the gap between leaf and whole plant WUE: the large effect of leaf position on daily carbon gain and water loss and the large flux of carbon losses by dark respiration. These results show that WUE evaluation among genotypes or treatments needs to be revised.

Growth of Shorea contorta Vid.Inoculated with Eucalypt Ectomycorrhizal Fungi in the Nursery and in a Logged-Over Dipterocarp Forest in Surigao,Philippines

This study was conducted to determine the compatibility of ectomycorrhizal (ECM) fungi associated with eucalypts on dipterocarps. Two nodal cuttings of Shorea contorta (Vid.) were rooted in a non-mist system for two months, and later inoculated with vegetative mycelia of three strains of Pisolithus collected under eucalypts and a strain of Scleroderma from dipterocarps. Inoculated rooted cuttings were planted in irradiated potting mix and raised in the nursery for five months. Root colonization prior to outplanting ranged from 31% to 38% and ECM fungi did not colonize the uninoculated ones. The cuttings were outplanted in a logged-over dipterocarp forest in bislig, Surigao Sur following a Randomized Complete block Design with four blocks;each block was planted with 50 seedlings. Two years after outplanting, Pisolithus strain from New Caledonia (H6394) promoted the highest height (46 cm) and diameter (0.48 cm) increment. Height was increased by 17% and diameter by 7% relative to the uninoculated control. by contrast, Pisolithus strain from the Philippines (H615) gave the shortest (26 cm) height increment and smallest (0.42 cm) diameter. Diameter growth of cuttings inoculated with Scleroderma D01 (from the Philippines) and a Pisolithus strain from Australia (H445) was bigger (0.47 cm) than the uninoculated treated cuttings (0.45 cm). In terms of survival, the uninoculated cuttings gave the highest (90%) percent survival while the lowest (60%) was those inoculated with Pisolithus strain from New Caledonia. It is possible that the indigenous ECM fungi infected easily the roots of the uninoculated cuttings thus contributing to the high survival rate (90%). During outplanting, fruit bodies of S. columnare were present in the field site. The results, however, show that ECM fungi were able to colonize the roots and that some strains promoted the growth and survival of S. contorta planted in a logged-over dipterocarp forest in bislig, Surigao, Philippines.

In Silico Evaluation of Anti-Malarial Agents from Hoslundia opposita as Inhibitors of Plasmodium falciparum Lactate Dehydrogenase (PfLDH) Enzyme

Malaria has continued to be a health and economic problem in Africa and the world at large. Many anti-malarial drugs have been rendered ineffective due to the emergence of resistant strains of Plamodium falciparum. A key malaria parasite enzyme in glycolytic pathway, P. falciparum lactate dehydrogenase (PfLDH) is specially targeted for anti-malarial drugs development. Thus, the aim of this investigation was to determine the in silico inhibition effects of antimalarial compounds from Hoslundia opposita Vahl. namely hoslundin, hoslundal and hoslunddiol on PfLDH enzyme. The compounds were docked to the three-dimensional structure of PfLDH as enzyme using AutoDock Vina in PyRx virtual screening software. binding affinity and position of the inhibitors were evaluated using PyMol software. The PfLDH enzyme showed two binding sites: the cofactors binding site (Site A) and secondary binding site (Site b). In the absence of the cofactor all ligands showed higher affinity than NADH, and were bound to the cofactors binding site (Site A). When docked in the presence of the cofactor, site b was the preferred binding site. binding to cofactor site with higher binding energy than NADH suggests that these ligands could act as preferential competitive inhibitors of PfLDH. However, the binding to site b also suggests that they may be non-competitive allosteric inhibitors. Amino acid residues Gly99, Asn140, Phe100 and Thr97 were indicated to form hydrogen bonds with Hoslundin. Hoslunddiol showed hydrogen bonding with Thr97 and Met30, while Hoslundal formed hydrogen bond with Thr101 and Asn140.

Rose Hip Powder That Contains the Natural Amount of Shells and Seeds Alleviates Pain in Osteoarthritis of the Dominant Hand—A Randomized, Double-blind, Placebo-Controlled, Cross-Over Clinical Trial

Aim: A standardized preparation of seeds and shells of selected sub-species of Rosa canina L, trade name Hyben Vital, reduces discomfort from osteoarthritis of the knee and hip. This study aims to investigate the impact of the same rose-hip powder (RHP) on discomfort and the consumption of rescue medication, in patients with osteoarthritis of the hand. Methods: The double blind, placebo-controlled, crossover trial included 30 patients with osteoarthritis of the dominant hand. Patients were randomly allocated to treatment with either five gram encapsulated RHP or placebo, for three months (Phase 1), after which they switched to the corresponding treatment for a further three months period (Phase 2). before entering the study, after 3 weeks and following three months of each of the study phases, scores for pain, stiffness and general feeling of discomfort were evaluated using a 10 step categorical scale, focusing on 16 different daily activities of the hand. The consumption of rescue medication was also calculated at the beginning and at the end of each study phase. Data are based on the intention to treat. Results: At the end of Phase 1, 90% of patients in the group receiving RHP first (group A), showed a reduction in pain, as compared to 36% in the group b initially given placebo (p 0.029). In line with this observation, stiffness and the general feeling of discomfort from the disease declined during RHP treatment (p 0.032). In group A, symptom reduction was still indicated by the study subjects 3 weeks after the switch to placebo. The consumption of rescue medication such as paracetamol, codeine and tramadol also declined significantly in group A when compared to group b (p 0.013). Conclusion: The present data suggest that administration of RHP, containing seeds and shells can reduce symptoms of osteoarthritis of the hand and consumption of rescue medication.

Case report of acute-on-chronic liver failure secondary to diffuse large b-cell lymphoma

Acute liver failure is a rare presentation of hematologic malignancy. Acute on chronic liver failure(ACLF) is a newly recognized clinical entity that describes acute hepatic decompensation in persons with preexisting liver disease. Diffuse large b-cell lymphoma(DLbCL) is an aggressive non-Hodgkin's lymphoma(NHL) with increasing incidence in older males, females and blacks. However, it has not yet been reported, to present with acute liver failure in patients with preexisting chronic liver disease due to human immunodeficiency virus(HIV)/hepatitis C virus(HCV) co-infection. We describe a case of ACLF as the presenting manifestation of DLbCL in an elderly black man with HIV/HCV coinfection and prior Hodgkin's disease in remission for three years. The rapidly fatal outcome of this disease is highlighted as is the distinction of ACLF from decompensated cirrhosis. Due to the increased prevalence of HIV/HCV co-infection in the African American 1945 to 1965 birth cohort and the fact that both are risk factors for chronic liver disease and NHL we postulate that the incidence of NHL presenting as ACLF may increase.
