Genetic variation at the p53 locus of two ecologically divergent Microtus pine voles:identification of molecular markers for species assignment

The Lusitanian(Microtus lusitanicus)and the Mediterranean(Microtus duodecimcostatus)pine voles are recently diverged sister species endemic of the Iberian Peninsula that can be identiﬁed with ecological and morphological characters,but in areas where the 2 species co-occur,species designation may be difﬁ*** discrimination between *** and *** has not been achieved yet possibly because of their estimated re-cent split and an evolutionary history that includes inter-species geneﬂ*** our previous observations on exons 5–7 of the p53 gene,here we analyze the potential use of the p53 genomic region as a discrimination marker of these species by extending our analyses to several kb upstream and downstream of the p53 gene and charac-terizing the degree of genetic differentiation in 7 markers within this ***,we fully sequenced the P53 protein of both *** observed:(i)generally high differentiation in this region;(ii)*** showed in general higher values of nucleotide and haplotype diversities;(iii)the concatenated phylogenetic tree separates the 2 species;(iv)the 2 P53 proteins only differ in 1 amino acid;(v)4 of the markers,2 in p53,one in Atp1b2,and another in Wrap53,contain species-speciﬁc genetic variation thus allowing a reliable discrimination between specimens from both species,irrespective of sampling location or introgression *** provide ad-ditional data on the putative role of p53 in the evolution of these species and present researchers with a fast and cost-effective resource for *** and *** identiﬁcation.

Burrow morphology of Uca uruguayensis and Uca leptodactylus(decapoda:ocypodidae)from a subtropical mangrove forest in the western Atlantic

The continuous excavation of burrows by fiddler crabs generates bioturbation in the sediment,which can be estimated from burrow *** aim of the present study was to describe the burrow morphology of Uca uruguayensis and *** and its relationship with demography of resident individuals and to estimate the level of bioturbation in the sediment generated by each *** all individuals from each of the 2 species,sex was determined and the carapace width(CW;mm)*** were characterized according to burrow diameter(BD;mm),maximum burrow depth(MBD;mm)and burrow volume(BV;cm^(3)).The density of each species in the study area was also *** both species,the males were larger and occupied burrows with higher BV compared to *** between sexes in relation to the burrow characteristics might reflect sexual dimorphism within the group and are probably related to the fact that males use the burrows for *** and BV showed significant positive relationships with the size of resident *** amount of sediment removed per burrow was estimated from mean BV:10.78 cm^(3)of sediment/burrow for *** and 12.38 cm^(3)of sediment/burrow for *** the density and depth differences between the 2 species,the similarity in burrow volume suggests that *** and *** present the same importance in terms of the bioturbation *** morphology is highly associated with characteristics of the occupant,although extrinsic factors should also be considered,and its description can provide estimates on the bioturbation generated by Uca species in mangrove forests.

Genomic characterization of multidrug-resistant extraintestinal pathogenic Escherichia coli isolated from grain culture soils

Multidrug-resistant(MDR)Escherichia coli,mainly extraintestinal pathogenic ***(ExPEC),has been widely reported in infections *** agricultural soils,manure is a hotspot for the dissemination of antimicrobial resistance genes(ARGs)and pathogenic bacteria;however,MDR bacteria have also been reported in soils with no history of manure *** addition,cross-resistance and co-resistance have been described as responsible for the metal-driven selection of bacteria resistant to ***,the aim of this study was to analyze three MDR *** isolates obtained from Brazilian grain culture soil samples with no history of manure use by whole-genome *** MDR *** isolates were recovered from soils from corn and coffee fields,and presented resistance toβ-lactams,quinolones,aminoglycosides,tetracyclines,sulphonamides,and dihydrofolate reductase *** analysis showed ARGs to several antimicrobials(i.e.,β-lactams,tetracyclines,aminoglycosides,sulphonamides,trimethoprim,phenicols,fosfomycin,and macrolides)as well as several metal resistance genes and antibacterial biocide resistance *** addition,known mutations in quinolone-resistance-determining regions of GyrA(Ser83Leu and Asp87Asn),ParE(Ser458Thr),and ParC(Ser80Ile)were also *** analysis showed the presence of virulence genes(lpfA,mcmA,gad,mchF,iroN,cma,and iss)associated with ***-resistant ExPEC isolates were assigned to phylogenetic group *** presence of MDR B1-ExPEC in soil samples shows the ability of these isolates to survive in *** study reports for the first time some sequence types(i.e.,ST345,ST448,and ST1146)of MDR *** in Brazilian ***,these findings contribute to the monitoring of antimicrobial resistance and surveillance studies based on whole-genome sequencing worldwide.

This review shows the importance of Wo_(3)photoanode as a potentially low-cost,efficient,stable,and photoactive material for light-driven water *** such,this manuscript aims to review the most recent publications regarding the strategies to improve the phoelectroactivity of Wo_(3)films for water *** addition,this review aims to graphically highlight and discuss the general trendings of the photocurrent density response and stability test of the recent outstanding studies in the literature for photoelectrochemical water splitting *** strategies covered in this review will not only concern the Wo_(3)morphology and crystal plane growth,but also the many arrangements possibilities to improve the Wo_(3)efficiency for water photoelectrooxidation,such as defect engineering based on oxygen vacancies,doping,decorations,and homo and *** these strategies are compared by the photocurrent density results and by the stability of these *** best results in this sense were observed in cases where the use of heterojunction was applied together with a desired morphology and crystal plane of the Wo_(3)***,the modifications that caused a decrease in the photocurrent density reaching values that are even lower than the pure Wo_(3)were also *** this way,this review intends to improve the knowledge about the synthesis and design of Wo_(3)photoanodes to further obtain an efficient photocatalyst to minimize the recombination losses or losses across the interfaces and improve the photoelectroactivity for water splitting in the large-scale application.

This study investigated the action of proteolytic enzymes in the production of goat viscera hydrolysates,and their flavoring *** study focus the production of flavor precursor molecules and the formation of volatile compounds resulting from four hydrolysis treatments:HA-Alcalase®,HF-Flavourzyme®,HN-Neutrase®,and HFN-Flavourzyme® and Neutrase®.The highest degree of hydrolysis (26.74%),number (23 volatiles) and concentration of volatile compounds (274.0 μg/L) were obtained with the action of *** main classes of volatile compounds identified in hydrolysates included acids,aldehydes,pyrazines,terpenes and Streacker *** acids,sugars and fatty acids were identified with higher concentrations in HF and HFN hydrolysates due to the low participation of these precursors in the formation of volatile ***,the use of Alcalase in the proteolysis of goat viscera stood out compared to other enzymes,providing impacting aroma compounds,such as pyrazines,for meat flavor.

As the most pervasive epigenetic marker present on mRNAs and long non-coding RNAs(lncRNAs),N6-methyladenosine(m^(6)A)RNA methylation has been shown to participate in essential biological *** studies have revealed the distinct patterns of m^(6)A methylome across human tissues,and a major challenge remains in elucidating the tissue-specific presence and circuitry of m^(6)A *** present here a comprehensive online platform,m^(6)A-TSHub,for unveiling the context-specific m^(6)A methylation and genetic mutations that potentially regulate m^(6)A epigenetic mark.m^(6)A-TSHub consists of four core components,including(1)m^(6)A-TSDB,a comprehensive database of 184,554 functionally annotated m^(6)A sites derived from 23 human tissues and 499,369 m^(6)A sites from 25 tumor conditions,respectively;(2)m^(6)A-TSFinder,a web server for high-accuracy prediction of m^(6)A methylation sites within a specific tissue from RNA sequences,which was constructed using multi-instance deep neural networks with gated attention;(3)m^(6)ATSVar,a web server for assessing the impact of genetic variants on tissue-specific m^(6)A RNA modifications;and(4)m^(6)A-CAVar,a database of 587,983 The Cancer Genome Atlas(TCGA)cancer mutations(derived from 27 cancer types)that were predicted to affect m^(6)A modifications in the primary tissue of *** database should make a useful resource for studying the m^(6)A methylome and the genetic factors of epitranscriptome disturbance in a specific tissue(or cancer type).m^(6)A-TSHub is accessible at ***/biologicalsciences/m^(6)ats.

Femoral positioning influences ipsi-and contralateral anterior cruciate ligament rupture following its reconstruction:Systematic review and meta-analysis

AIM To systematically review the incidence of ipsilateral graft re-rupture and contralateral anterior cruciate ligament(ACL) rupture following its reconstruction, with special attention to the femoral drilling technique. METHoDS Systematic review and meta-analysis of high-level prospective studies searched in MEDLINE database following PRISMA statement. The rate of ipsilateral graft re-rupture and contralateral rupture in patients submitted to either transtibial(TT) technique(isometric) or anteromedial(AM) technique(anatomic) was compared. RESULTS Eleven studies met the criteria and were included in final analysis. Reconstructions using the AM technique had a similar chance of contralateral ACL rupture when compared to the chance of ipsilateral graft failure(oR = 1.08, P = 0.746). In reconstructions using TT technique, the chance of contralateral ACL rupture was approximately 1.5 times higher than ipsilateral graft failure(oR = 1.49, P = 0.048). Incidence of contralateral lesions were similar among the techniques TT(7.4%) and AM(7.0%)(P = 0.963), but a trend could be noticed with a lower incidence of lesion in the ipsilateral limb when using the TT technique(4.9%) compared to the AM technique(6.5%)(P = 0.081).CoNCLUSIoN ACL reconstruction by TT technique leads to lower incidence of graft re-injury than contralateral ACL lesion. There is no difference between the chance of re-injury after AM technique and the chance of contralateral ACL lesion(native ligament) with either technique.

Extramedullary plasmocytoma associated with a massive deposit of amyloid in the duodenum

We report a rare case of extramedullary plasmocytoma associated with a massive deposit of amyloid in the duodenum. A 72-year-old Japanese man was admitted to our hospital presenting with a 3-mo history of epigastric pain, vomiting and weight loss. on computed tomography （CT） a wall thickening of the fourth part of the duodenum was observed. Multiple biopsies obtained from the lesion showed infiltration of plasma cells and lymphocytes, but they were not conclusive. The patient underwent resection of the lesion and, on histopathological examination, the lesion consisted of a dense and diffuse infiltrate of plasma cells and a few admixed lymphocytes with reactive follicles extending to the muscular propria. An extensive deposition of amyloid was also observed. Immunohistochemical stains revealed that a few plasmacytoid cells showed λ light chain staining, though most were κ： light chain positive. These cells also were positive for CD138 and CD56 but negative for CD20 and CD79. The findings were consistent with extramedullary plasmocytoma associated with a massive deposit of amyloid in duodenum. A subsequent workup for multiple myeloma was completely negative. The patient showed no signs of local recurrence or dissemination of the disease after 12 mo follow-up. Because of the association of plasmocytoma and amyloidosis, the patient must be followed up because of the possible systemic involvement of the neoplasm and amyloidosis in future.
