Emerging role of liquid biopsy in rat sarcoma virus mutated metastatic colorectal cancer:A case report

BACKGRoUND In patients with metastatic colorectal cancer(mCRC),the treatment options are limited and have been proved to be affected by rat sarcoma virus(RAS)mutational *** RAS wild-type(wt)patients,the combination of antiepidermal growth factor receptor(EGFR)monoclonal antibodies with chemotherapy(CT)is more effective than CT *** the other hand,RAS-mutated patients are not eligible for treatment with anti-EGFR *** SUMMARY Eleven patients with initially RAS-mutated mCRC were followed from diagnosis to May *** the time of cell-free DNA determination,five patients had undergone one CT line,five patients had undergone two CT lines,and one patient had undergone three CT lines(all in combination with bevacizumab).At the second and third treatment lines[second line(2L),third line(3L)],patients with neo-RAS wt received a combination of CT and *** neo-RAS wt patients treated with anti-EGFR,our findings indicated an increase in progression-free survival for both 2L and 3L(14.5 mo,P=0.119 and 3.9 mo,P=0.882,respectively).Regarding 2L overall survival,we registered a slight increase in neo-RAS wt patients treated with anti-EGFR(33.6 mo vs 32.4 mo,P=0.385).At data cut-off,two patients were still alive:A RAS-mutated patient undergoing 3L treatment and a neo-RAS wt patient who received 2L treatment with anti-EGFR(ongoing).CoNCLUSIoN our case series demonstrated that monitoring RAS mutations in mCRC by liquid biopsy may provide an additional treatment line for neo-RAS wt patients.

Twisted and Absolute Degrees of Maps into the Projective Plane

For each based map f : X → RP2from a closed surface into the real projective plane, we compute its absolute and twisted degrees and describe the action of the fundamental group of RP2over the based homotopy class of f. We emphasize the finding that for any nonorientable closed surface,there exists only one based homotopy class of maps from it into RP2whose maps have twisted degree zero and absolute degree nonzero – which shows that, unlike the absolute degree, the twisted degree is not able to detect the strong surjectivity in this setting. In all the other scenarios, the absolute degree of each map is either equal to the twisted degree or its absolute value – and so the twisted degree detects strong surjectivity.

This work describes the use of Tio_(2)nanotubes-based electrodes(TNT)modified with Cu_(2)o nanostructures and gold nanoparticles for the photoelectroreduction of Co_(2)to produce value-added compounds.A thin layer of polydopamine was used as both an adherent agent and an electron transfer mediator,due to itsπ-conjugated electron *** highest production yield was achieved using a TNT@PDA/Nc/Au40%electrode,with Faradaic efficiencies of 47.4%(110.5μM cm^(-2))and 27.8%(50.4μM cm^(-2))for methanol and methane,*** performance of the photoelectrodes was shown to be Cu_(2)o facet-dependent,with cubic structures leading to greater conversion of Co_(2)to methanol(43%)and methane(27%),compared to the octahedral morphology,while a higher percentage of metallic gold on the nanostructured Cu_(2)o surface was mainly important for *** functional theory(DFT)calculations supported these findings,attributing the superior photoelectrocatalytic performance of the TNT@PDA/Nc/Au40%electrode for CH4generation to the formation of an oCH3intermediate bonded to Au *** using isotope-labeling and analysis by gas chromatograph-mass(GC-MS)demonstrated that13Co_(2)was the source for photoelectrocatalytic generation of13CH3oH and13CH313CH2oH.

The broke of a tailings dam from Vale S.A. destroyed a gigantic area killing 270 persons in Brumadinho/Brazil. organizing activities after a mass disaster is a complex process that requires the involvement of many people and resources in the laboratory. To DNA identification of the victims, a daunting work had to be made by DNA laboratory staff in a collaborative effort with many partners. Efforts to implement good practice guidelines in Disaster Victim Identification (DVI) have revealed several important aspects that need to change in the forensic DNA laboratory. This article highlights the challenges of implementing DVI best practice guidelines in resource-poor settings, but with professionals from different sectors engaged in the same goal of briefly identifying victims and helping families and society.

Evaluation of the Efficiency of the Magicramp^® Device for Reducing Cramps Resulting from oncological Treatments: “Double-Blind, Randomized Clinical Trial”

We identified that oncological treatments in general (chemotherapies, immunotherapies and radiotherapies) frequently cause peripheral neuropathy, including cramps, characterized by excess protons due to metabolic and neuronal factors, such as sudden changes in pH, uremia and aspects that affect neuromotor functions. Such situations and others like them are often neglected in treatment, which naturally concerns itself with the main problem: Cancer. Sometimes toxic solutions are implemented that have comorbid side effects, such as duloxetine (standard treatment). Based on monitoring of cancer patients who used the non-toxic product, called “Magicramp® Electrostatic Charge Reduction Cushion” (MECRC), approved in Europe more than 10 years ago, we carried out a controlled test in Brazil. In this clinical trial, we hypothesized that reducing excessive ionic charges (electrostatic charge), which is one of the side effects often described in the literature as “Chemotherapy-Induced Peripheral Neuropathy” (CIPN), would decrease or eliminate cramping, under the hypothesis that such elimination would prevent or attenuate muscular vulnerability to action impulses, and increase the power of relaxation through the same mechanism. In this double-blind and randomized clinical trial, 40 (forty) adult patients with muscle cramps caused by oncological treatments were tested, evaluating the degree of efficiency of the product that aims to reduce muscle cramps, by eliminating and/or reducing excess loads electrostatic ionic. A significant portion of Landslide Early Warning Systems (LEWS) relies on the definition of operational thresholds and the monitoring of cumulative rainfall for alert issuance. These thresholds can be obtained in various ways, but most often they are based on previous landslide data. This approach introduces several limitations. For instance, there is a requirement for the location to have been previously monitored in some way to have this type of information recorded. Another significant limitation is the need for information regarding the location and timing of incidents. Despite the current ease of obtaining location information (GPS, drone images, etc.), the timing of the event remains challenging to ascertain for a considerable portion of landslide data. Concerning rainfall monitoring, there are multiple ways to consider it, for instance, examining accumulations over various intervals (1 h, 6 h, 24 h, 72 h), as well as in the calculation of effective rainfall, which represents the precipitation that actually infiltrates the soil. However, in the vast majority of cases, both the thresholds and the rain monitoring approach are defined manually and subjectively, relying on the operators’ experience. This makes the process labor-intensive and time-consuming, hindering the establishment of a truly standardized and rapidly scalable methodology on a large scale. In this work, we propose a Landslides Early Warning System (LEWS) based on the concept of rainfall half-life and the determination of thresholds using Cluster Analysis and data inversion. The system is designed to be applied in extensive monitoring networks, such as the one utilized by Cemaden, Brazil’s National Center for Monitoring and Early Warning of Natural Disasters.

Validation of the Portuguese version of the social isolation scale with a sample of community-dwelling older adults

NDRank: optimised parallel search for weather analogues

Global meteorology data are now widely used in various areas, but one of its applications, weather analogues, still require exhaustive searches on the whole historical data. We present two optimisations for the state-of-the-art weather analogue search algorithms: a parallelization and a heuristic search. The heuristic search (NDRank) limits of the final number of results and does initial searches on a lower resolution dataset to find candidates that, in the second phase, are locally validated. These optimisations were deployed in the Cloud and evaluated with ERA5 data from ECMWF. The proposed parallelization attained speedups close to optimal, and NDRank attains speedups higher than 4. NDRank can be applied to any parallel search, adding similar speedups. A substantial number of executions returned a set of analogues similar to the existing exhaustive search and most of the remaining results presented a numerical value difference lower than 0.1%. The results demonstrate that it is now possible to search for weather analogues in a faster way (even compared with parallel searches) with results with little to no error. Furthermore, NDRank can be applied to existing exhaustive searches, providing faster results with small reduction of the precision of the results.
