Growth inhibitory effects of Phyllanthus niruri extracts in combination with cisplatin on cancer cell lines

AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in combination with cis- platin on two cancer cell lines. METHoDS： colorectal carcinoma （HT29） and human hepatocellular carcinoma （HepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDEPN） either alone or in combination with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viability, we stained the cells with prop- idium iodide and assessed them by flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences between groups were determined using analysis of variance and Bonferroni＇s test, as indicated. A value of P 〈 0.05 was considered to be statistically significant. RESULTS： SDEPN had significantly different cyto- toxic effects on HT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and HepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDEPN also had significantly different cytotoxic effects on HT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and HepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. Both cell lines were killed by cisplatin in a dose-dependent manner compared to control cells （HepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and HT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In HT29 cells, pretreat- ment with SDEPN and subsequent treatment with cis-platin resulted in a greater number of cells being killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）. Differences in viral kinetics between genotypes 1 and 3 of hepatitis c virus and between cirrhotic and non-cirrhotic patients during antiviral therapy

AIM： To evaluate the impact of hepatitis c virus （HcV） infection with genotype 1 or 3 and the presence or absence of liver cirrhosis （Lc） in the early viral kinetics response to treatment. METHoDS： Naive patients （n = 46） treated with interferon-α （IFN-α） and ribavirin and followed up with frequent early HcV-RNA determinations were analysed. Patients were infected with genotype 1 （n = 28, 7 with Lc） or 3 （n = 18, 5 with Lc）. RESULTS： The first phase decline was larger in genotype 3 patients than in genotype 1 patients （1.72 vs 0.95 log IU/mL, P 〈 0.001）. The second phase slope decline was also larger in genotype 3 patients than in genotype 1 patients （0.87 vs 0.15 log/wk, P 〈 0.001）. Differences were found in both cirrhotic and non-cirrhotic patients. Genotype 1 cirrhotic patients had a slower 2^nd phase slope than non-cirrhotic patients （0.06 vs 0.18 log/wk, P 〈 0,02）. None of genotype 1 cirrhotic patients had a 1^st phase decline larger than 1 log （non-cirrhotic patients： 55%, P 〈 0.02）. A similar trend toward a slower 2nd phase slope was observed in genotype 3 cirrhotic patients but the 1^st phase slope decline was not different. Sustained viral response was higher in genotype 3 patients than in genotype 1 patients ,（72% vs 14%, P 〈 0.001） and in genotype 1 non-cirrhotic patients than in genotype 1 cirrhotic patients （19% vs 0%）. A second phase decline slower than 0.3 log per week was predictive of non-response in all groups. coNcLUSIoN： Genotype 3 has faster early viral decline than genotype 1. cirrhosis correlates with a slower 2nd phase decline and possibly with a lower 1^st phase slope decline in genotype 1 patients.

In this study, coral?like yolk–shell?structured Nio/c composite microspheres(denoted as cYS?Nio/c) were prepared using spray pyrolysis. The unique yolk–shell structure was characterized, and the formation mechanism of the structure was proposed. Both the phase separation of the polyvinylpyrrolidone and polystyrene(PS) colloidal solution and the decompo?sition of the size?controlled PS nanobeads in the droplet played crucial roles in the formation of the unique coral?like yolk–shell structure. The cYS?Nio/c microspheres delivered a reversible discharge capacity of 991 mAh g^(-1) after 500 cycles at the current density of 1.0 A g^(-1). The dis?charge capacity of the cYS?Nio/c microspheres after the 1000 th cycle at the current density of 2.0 A g^(-1) was 635 mAh g^(-1), and the capacity retention measured from the second cycle was 91%. The final discharge capacities of the cYS?Nio/c microspheres at the current densities of 0.5, 1.5, 3.0, 5.0, 7.0, and 10.0 A g^(-1) were 753, 648, 560, 490, 440, and 389 mAh g^(-1), respectively. The synergetic e ect of the coral?like yolk–shell structure with well?defined interconnected mesopores and highly conductive carbon resulted in the excellent Li+?ion storage properties of the cYS?Nio/c microspheres.

Fatigue crack in large-scale tubular joints for offshore structures

Tubular members have been used for constructing offshore structures. Large-scale fatigue tests of welded tubular K-joints under the balanced in-plane bending brace were carried out to investigate the fatigue behavior of API 2W Gr.50 steel produced by PoSco. The experimental results were verified by numerical approaches and compared with the IIW, DnV RP-c203 and API RP 2A-WSD design curves. The test results based on the hot spot stress were in agreement with the design curves. The ScF factor for tubular K-joint was also obtained. The numerical parametric study of the K-joint (using the finite element program) was described and its results were compared with experimental results. The stress effects of various parameters including α, β, γ, τ and θ on the stress in the K-joint were investigated. The stress distribution for each parameter was introduced. The study showed that the maximum stress of the joint varied according to the variation of joint parameters.

Data Utilization-Based Adaptive Data Management Method for Distributed Storage System in WAN Environment

Recently,research on a distributed storage system that efficiently manages a large amount of data has been actively conducted following data production and demand *** expansion limits exist for traditional standalone storage systems,such as I/o and file system ***,the existing distributed storage system does not consider where data is consumed and is more focused on data dissemination and optimizing the lookup cost of data *** this leads to system performance degradation due to low locality occurring in a Wide Area Network(WAN)environment with high network *** problem hinders deploying distributed storage systems to multiple data centers over *** lowers the scalability of distributed storage systems to accommodate data storage *** paper proposes a method for distributing data in a WAN environment considering network latency and data locality to solve this problem and increase overall system *** proposed distributed storage method monitors data utilization and locality to classify data temperature as hot,warm,and *** assigned data temperature,the proposed algorithm adaptively selects the appropriate data center and places data accordingly to overcome the excess latency from the WAN environment,leading to overall system performance *** paper also conducts simulations to evaluate the proposed and existing distributed storage *** result shows that our proposed method reduced latency by 38%compared to the existing ***,the proposed method in this paper can be used in large-scale distributed storage systems over a WAN environment to improve latency and performance compared to existing methods,such as consistent hashing.

Rice genes osDjA2 and osERF104,encoding a chaperone protein and an APETELA2/ethylene-responsive factor,respectively,are strongly induced in a compatible interaction with blast fungus,and also have function in plant susceptibility validated through gene ***,we reported the cRISPR/cas9 knockout of osDjA2 and osERF104 genes resulting in considerable improvement of blast resistance.A total of 15 osDjA2(62.5%)and 17 osERF104(70.8%)T_(0)transformed lines were identified from 24 regenerated plants for each target and used in downstream *** of homozygous T1 mutant lines revealed not only a significant decrease in the number of blast lesions but also a reduction in the percentage of diseased leaf area,compared with the infected control *** results supported cRISPR/cas9-mediated target mutation in rice susceptibility genes as a potential and alternative breeding strategy for building resistance to blast disease.

A plug‐flow reactor coated with carbon‐doped Tio2 (c‐Tio2 ) powder was investigated for the control of vaporous aromatics (benzene, toluene, ethylbenzene, and o‐xylene (BTEX)) under a range of experimental conditions. The characteristics of the as‐prepared c‐Tio2 and a reference Degussa P25 Tio2 powder were examined using X‐ray diffraction, scanning electron microscopy, diffuse‐reflectance ultraviolet‐visible‐near infrared spectroscopy, and Fourier transform infrared spectroscopy. The experimental conditions for the photocatalytic performance of the as‐prepared c‐Tio2 photocatalyst were controlled using three operational parameters, relative humidity, flow rate, and input concentration. Unlike other target compounds, very little benzene was removed by the c‐Tio2 photocatalyst under visible‐light irradiation. In contrast, the c‐Tio2 exhibited higher removal efficiencies for the other three target compounds (toluene, ethylbenzene, and xylene) compared with those achieved using unmodified Tio2 under visible‐light irradiation. The highest removal efficiency was obtained at a relative humidity value of 45%. Specifically, the toluene removal efficiency determined at a relative humidity of 45% was 78%, whereas it was close to 0%, 7.2%, and 5.5% for relative humidity values of 20%, 70%, and 95%, respectively. In addition, the removal efficiencies for the three target compounds decreased as the flow rate or input concentration increased. These findings indicate that the as‐prepared c‐Tio2 photocatalyst could be used for the removal of toxic vaporous aromatics under optimized operating conditions.

PNPLA3 and TM6SF2 polymorphisms in Brazilian patients with nonalcoholic fatty liver disease
