Selective expansion and enhanced anti-tumor effect of antigen-specific CD4^(+)T cells by retrovirus-mediated IL-15 expression

Mounting evidence has demonstrated that CD4^(+)T cells play an important role in anti-tumor immune responses.Thus,adoptive transfer of these cells may have great potential for anti-cancer therapy.However,due to the difficulty to generate sufficient tumor-specific CD4^(+)T cells,the use of CD4^(+)T cells in tumor therapy is limited.It has been found that IL-15 transfection enhances the proliferation and anti-tumor activity of tumor-specific CD8+Tcells,but the effect of IL-15 transfection on CD4^(+)T cells remains unknown.Here,the effects of retrovirusmediated IL-15 expression in Ova-specific CD4^(+)T cells from Do11.10 mice were evaluated and it was discovered that IL-15 transfected CD4^(+)T cells expressed both soluble and membrane-bound IL-15.Retrovirusmediated IL-15 expression led to a selective expansion of antigen-specific CD4^(+)T cells by inhibiting their apoptosis.In vivo IL-15 transfected CD4^(+)T cells were more effective in suppressing tumor growth than control retroviral vector transfected ones.To ensure the safety of the method,the employment of thymidine kinase gene made it possible to eliminate these transgenic CD4^(+)T cells following ganciclovir treatment.Together,we show that IL-15 transfection induced a selective expansion of antigen-specific CD4^(+)T cells ex vivo and enhanced their tumor-suppression effects in vivo.This has an important significance for improving the efficacy of adoptive T cell therapy.

The rapid spread of viral zoonoses can cause severe consequences,including huge economic loss,public health problems or even global crisis of society.Clinical detection technology plays a very important role in the prevention and control of such zoonoses.The rapid and accurate detection of the pathogens of the diseases can directly lead to the early report and early successful control of the diseases.With the advantages of being easy to use,fast,portable,multiplexing and cost-effective,semiconductor biosensors are kinds of detection devices that play an important role in preventing epidemics,and thus have become one of the research hotspots.Here,we summarized the advances of semiconductor biosensors in viral zoonoses detection.By discussing the major principles and applications of each method for different pathogens,this review proposed the directions of designing semiconductor biosensors for clinical application and put forward perspectives in diagnostic of viral zoonoses.

Identification of tick-borne pathogens using metagenomic analyses in H.longicornis feeding on humans in downtown Beijing

On August 14th,2018,a Beijing resident living in Xicheng District found a female H.longicornis tick attached to the skin at the front of his upper shin.On examination,the patient was afebrile and appeared well.The species of the tick was identified through morphological characteristics and phylogenetic analysis based on cytochrome C oxidase subunit I.This H.longicornis tick was screened for tick-borne pathogens such as viruses,bacteria and parasites.RNA pathogens were screened by PCR and sequencing,while DNA pathogens were screened by metagenomic analyses.It was found that the tick was positive for the DNA sequences of zoonotic and animal pathogens such as A phagocytophilum,Ehrlichia minasensis and C.burnetii.Considering the good health condition of the patient,we hypothesized that the pathogens originated from the tick specimen itself rather than host blood meal.For the first time,our study reveals the possible risk of transmission of tick-borne pathogens to human beings through tick bit in downtown Beijing.Further research is needed to screen for tick-borne pathogens among unfed ticks collected from central Beijing.

A Cooperative Localization Method for Leader–Follower Multiple UAVs Using Continuous Relative Ranging Information

Cooperative localization(CL)utilizing multiple unmanned aerial vehides(UAVs)has become the current development trend and research hotspot.The tnditional distance-based CL method.typically uses principle of the spherical intersection to achieve UAV high-precision positioning.However,this distance based CL strategy requires several leader UAVs at the known positions.Thus,a cooperative localization method for leader-follower multiple UA Vs using continuous relative ranging information is proposed in this paper.Aiming at the UAV formation consists of a leader UAV node with the known position and at least a follower UAV that needs to be located.Using the Inertial short-time high-precision relative position constraint and the con-tinuous relative distance constraints with the leader UAV nodes,the closed-form solution of follower UAV's three-dimensional position can be obtained using spherical intersection prin-ciple.On this basis,we derive and construct follower UAV's position observation equation.The:developed strategy requires a single known position leader UAV node to realize high-precision CL,which can efctively solve the taditional distance-based collaborative location problem in case the number of leader UAVs is small.The simulation and experimental results demonstrate that the proposed method significantly improves follower UAV's positioning accuracy,and neglects the limitation of the traditional distance-based CL method on the number of leader UAV nodes at known positions.

试验研究了玉米秸秆带状还田对当年土壤养分含量和玉米产量的影响,为黑龙江省黑土区秸秆还田策略提供理论依据。试验在黑龙江省高纬度寒区开展,于2022年春季设置秸秆离田(CK)、整株秸秆带状还田(T1)、粉碎秸秆带状还田(T2)和粉碎秸秆配施有机肥带状还田(T3)4个处理。结果表明:短期内,秸秆带状还田不同模式对土壤养分指标和玉米产量构成指标有不同影响,与CK相比,T3处理土壤有机质和全氮含量分别增加7.62%和9.75%,电导率减少9.21%,玉米穗长增加8%以上,行粒数增加6.98%,单棒重增加23.61%,产量增加24.05%。通过主成分分析和典型相关分析,发现秸秆不同带状还田模式对土壤有机质和全氮影响的权重高于其他养分指标,对玉米产量构成指标影响较大的是单棒重、穗长和百粒重;土壤有机质、全氮含量与玉米单棒重、穗长和百粒重呈正相关性,而土壤电导率与玉米单棒重、穗长和百粒重呈负相关性。

Homologous fault monitoring technology of redundant INS in airborne avionics systems

Redundant technology plays an important role in improving the reliability and fault-tolerance of the airborne avionics systems. A Markov state transition model is introduced to the reliability analysis of the redundant inertial navigation system （RINS） in airborne navigation systems. An information processing mechanism based on difference filtering is put forward to strengthen the consistency between the outputs of the equal-precision inertial navigation system （INS）. On this basis, the homologous fault monitoring algorithm is designed to realize the homologous fault monitoring of RINS. The simulation is carried out based on the above algorithms, and the results verify the effectiveness of the proposed fault monitoring algorithm based on difference filtering. Research results have good reference value for the configuration and design of RINS in airborne integrated avionics systems.

Heavy metal contamination in soils of greenhouse vegetable production systems in a cold region of China

Heavy metal(HM)contamination in soils of greenhouse vegetable production(GVP)systems has drawn increasing attention in terms of food safety.In the present study,64 surface soils were sampled,and the concentrations of select HMs were determined using atomic absorption spectroscopy.The results showed that the concentrations of cadmium(Cd),lead(Pb),zinc(Zn),copper(Cu),nickel(Ni)and chromium(Cr)in the soils were(0.2±0.2)mg/kg,(26.5±8.4)mg/kg,(101.4±43.2)mg/kg,(29.1±8.6)mg/kg,(24.5±3.3)mg/kg,and(56.5±6.3)mg/kg,and the corresponding accumulation index(AI)values were 2.30,1.10,1.43,1.45,1.07,and 0.97,respectively.The spatial distribution of the HMs suggested that Cd pollution displays a fractionation effect,which may be related to the source of Cd and its mobility.The concentration of Zn was significantly correlated with that of other HMs,implying that a comprehensive interactive effect might occur between Zn and other HMs.Furthermore,the values of the potential ecological risk index(RI)ranged from 41.23 to 185.91,meaning that attention should be paid to HM contamination of GVP soils to ensure food quality and safety.

Molecular Identification of D. nuttalli and D.marginatus

This study was to establish a molecular identification method to distinguish Dermacentor nuttalli(D. nuttalli) and Dermacentor marginatus(D. marginatus),and to study their phylogenetic relationship. The ticks were collected from domestic animals in Xinjiang Uygur Autonomous Region for morphological identification,then their genomic DNAs were isolated for amplifying mitochondrial 16 S rRNA and cytochrome oxidase subunit I gene(COI). The phylogenetic tree was constructed based on the sequencing results of PCR products by employing Mega 5. 0 and Mrbayes 3. 2 for homology analysis. Clustering analysis PCR product for 16 S rRNA from both D. marginatus and D. nuttalli was clustered together with their respective 16 S rRNA sequences previously accessed in GenBank,and that for COI gene as well. These are basically identical with morphological identification results. Our results indicate that morphological identification,combined with molecular markers,would be a simple and accurate method for distinguishing D. nuttalli and D. marginatus.

针对园区巡检、物流等固定场景下的任务,无人车的主流导航方法为预先构建场景点云地图,在运行时通过激光雷达(LiDAR)扫描的点云与地图相匹配,进而得到高精度的定位结果。为解决地图构建时位姿估计精度受外参误差影响较大的问题,提出了一种全球导航卫星系统(GNSS)/LiDAR外参在线标定算法,对传感器外参进行在线估计。通过搭建仿真平台对算法进行验证。结果表明:该算法可以有效地估计出GNSS与LiDAR之间的外参,杆臂估计精度达到厘米(cm)级,姿态角估计精度优于0.1°。

N秒平均法与Allan方差法是2种常用的光纤陀螺随机误差性能评价方法,其得到的指标是衡量光纤陀螺性能的重要依据。结合2种性能评价方法的特点,对二者进行了分析与对比,并推导了2种方法所得性能指标的关系表达式:首先从随机信号处理的角度对N秒平均法进行了分析,得到了其幅频响应函数;继而对Allan方差法所辨识的量化噪声、角度随机游走、零偏不稳定性、角速率随机游走、速率斜坡5种随机误差模型进行了分析,并推导了5种随机噪声的N秒平均性能指标。根据俄罗斯产某型号光纤陀螺的随机误差模型进行了仿真,结果表明仿真信号的性能指标与理论推导结论具有较好的一致性。在一定程度上揭示了2种光纤陀螺性能评价方法的内在联系,对2种评价方法的实际应用具有一定指导意义。
