Objective:The aim of this study was to explore the effects of Babao dan(BBD),a traditional Chinese medicine,on gastric cancer(GC)progression in *** and Methods:A subcutaneous xenograft mouse model of GC was established using MGC80-3 *** terminal deoxynucleotidyl transferase-mediated dUTP:2'-deoxyuridine 5'-triphosphate-biotin nick-end labeling method was adopted to detect cell apoptosis in *** expression levels of proteins associated with proliferation,apoptosis,and angiogenesis were measured by immunohistochemical staining or western blotting(WB).The activation and protein levels of p-c-Jun N-terminal kinase(JNK),p-p38,p-extracellular-regulated kinase 1/2,p-nuclear factor-κB(NF-κB),and p-STAT3 were examined by Bio-plex and ***:BBD significantly inhibited tumor growth in GC mouse models with no adverse effect on body weight or organ *** was also found that BBD significantly suppressed the proliferation of GC tumor cells,induced the apoptosis of tumor cells,and inhibited angiogenesis through inactivating with mitogen-activated protein kinase,NF-κB,and STAT3 ***:BBD exerts suppressive effects on GC tumor growth by regulating multiple pathways in vivo,which may provide a novel treatment option for GC therapy.

Qingjie Fuzheng granules inhibit colorectal cancer cell growth by the PI3K/AKT and ERK pathways

BACKGROUND Qingjie Fuzheng granules(QFGs) are part of a traditional Chinese medicine formula, which has been widely used and found to be clinically effective with few side effects in various cancer treatments, including colorectal cancer(CRC).However, the precise mechanisms and molecular signaling pathways involved in the activity of QFGs' anticancer effect have not been reported in the literature. In this study, we hypothesized that QFGs can inhibit the growth of colorectal cancer cells, and that its mechanism is closely related to one or more intracellular signal transduction *** To better evaluate the mechanism underlying the anti-cancer effect of QFGs on the CRC cell lines HCT-116 and *** First, we measured cell viability and cytotoxicity by performing MTT and lactate dehydrogenase(LDH) assays. We evaluated the role of QFGs in cell proliferation and apoptosis by assessing colony formation and analyzing Hoechst 33258 staining. Second, cell cycle and apoptosis rates were measured by fluorescence activated cell sorting, and the expression levels of survivin, cyclin D1, CDK4, p21,Bax, Bcl-2, Fas, FasL, and cleaved-caspase-3/-8/-9 were measured by performing western blots and caspase activity assays. Furthermore, inhibitors of caspase-3/-8/-9 were used to elucidate the specific apoptosis pathway induced by QFGs in cancer cells. Finally, activation of the PI3 K/AKT and ERK signaling pathwayswas examined using the western blot assay to investigate the possible *** MTT and LDH assays revealed that after 0.5-2.0 mg/mL of QFGs treatment, cell viability was reduced by(6.90% ± 1.03%)–(59.70% ± 1.51%)(HCT-116; P < 0.05)and(5.56% ± 4.52%)–(49.44% ± 2.47%)(HCT-8; P < 0.05), and cytotoxicity was increased from 0.52 ± 0.023 to 0.77 ± 0.002(HCT-116; P < 0.01) and from 0.56 ±0.054 to 0.81 ± 0.044(HCT-8; P < 0.01) compared with the non-QFGs treatment groups. The lifetime of Si bond coatings in environmental barrier coatings is constrained by phase-transition-induced cracking of the SiO_(2)*** this study,Si-HfO_(2)dual-state duplex composite materials are proposed to address this issue by partially forming HfSiO_(4)and minimizing the SiO_(2)*** as-prepared composite exhibited a structure comprising discrete HfO_(2)“bricks”embedded in a continuous Si“mortar”,while the oxidized state transformed into discrete HfSiO_(4)“bricks”within continuous thin SiO_(2)“mortars”.The results indicate that continuous thin SiO_(2)contributes to reducing the oxidation rate to a level comparable to that of pure Si,and discrete HfSiO_(4)particles aid in relieving phase transition-induced stress and inhibiting crack propagation,thereby enhancing oxidation and cracking resistance ***,the composite with 20 mol%HfO_(2)and a mean particle size of~500 nm at 1370℃exhibited a service lifetime 10 times greater than that of pure *** research provides valuable insights for designing Si-based bond coatings with improved service lifetime.

Heat stress induced hepatocyte apoptosis in largemouth bass Micropterus salmoides via IRE1α/TRAF2/ASK1/JNK pathway

Heat stress(HS)has been shown to adversely affect fish livers and can lead to extensive *** investigate the relationship between endoplasmic reticulum(ER)stress and HS-induced apoptosis in fish livers,we isolated and cultured primary hepatocytes of largemouth bass,Micropterus salmoides by trypsin method,then established an in-vitro model of liver cells under HS(35℃).The contents of lactic dehydrogenase(LDH)and hydrogen peroxide(H2O2)were determined to evaluate the effects of HS on hepatocyte injury and oxidative ***-qPCR was performed to discover the key genes in unfolded protein response(UPR)pathways involved at different HS *** inhibitor 4-PBA and IRE1αinhibitor 4μ8C were used to further investigate the effects of HS on IRE1αapoptosis pathway in *** show that HS led to significant increases in the release of LDH,the content of H2O2,and the expressions of oxidative protein folding genes(ero1αand pdi)under HS,suggesting severe hepatocyte injury and oxidative stress happened in heat-stressed largemouth bass *** continuous activation of IRE1αpathway genes(grp78,grp94,atf6,perk,eif2a,atf4,chop,ire1α,traf2,ask1,jnk1,and jnk2)indicated that HS led significantly to ER *** particular,the mRNA expression levels of ER stress-related genes(grp78,grp94,atf6,perk,ire1α,chop,jnk1,and jnk2)in the high temperature(HT)+4-PBA group and the HT+4μ8C group were significantly down-regulated under *** 4μ8C treatment,the expression levels of apoptosis-related genes(caspase-2,caspase-3,caspase-6,caspase-7,caspase-8,caspase-9,and caspase-10)and LDH content were significantly decreased,whereas the cell survival rate was significantly increased when given 4-PBA or 4μ8C *** findings demonstrate that HS could induce liver apoptosis of largemouth bass through the IRE1αpathway,which may act as a key switch mediating liver apoptosis of largemouth bass under HS.

Most viruses and transposons serve as effective carriers for the introduction of foreign DNA up to 11 kb into vertebrate ***,their activity markedly diminishes with payloads exceeding 11 *** the payload capacity of transposons could facilitate more sophisticated cargo designs,improving the regulation of expression and minimizing mutagenic risks associated with molecular therapeutics,metabolic engineering,and transgenic animal *** this study,we improved the Tol2 transposon by increasing protein expression levels using a translational enhancer(QBI SP163,ST)and enhanced the nuclear targeting ability using the nuclear localization protein H2B(SHT).The modified Tol2 and ST transposon efficiently integrated large DNA cargos into human cell cultures(H1299),comparable to the well-established super PiggyBac ***,mRNA from ST and SHT showed a significant increase in transgene delivery efficiency of large DNA payloads(8 kb,14 kb,and 24 kb)into zebrafish(Danio rerio).This study presents a modified Tol2 transposon as an enhanced nonviral vector for the delivery of large DNA payloads in transgenic applications.

The innate immune pathway is receiving increasing attention in cancer *** pathway is ubiquitous across various cell types,not only in innate immune cells but also in adaptive immune cells,tumor cells,and stromal *** targeting the innate immune pathway have shown profound changes in the tumor microenvironment(TME)and improved tumor prognosis in preclinical ***,to date,the clinical success of drugs targeting the innate immune pathway remains ***,recent studies have shown that activation of the innate immune pathway can paradoxically promote tumor *** uncertainty surrounding the therapeutic effectiveness of targeted drugs for the innate immune pathway is a critical issue that needs immediate *** this review,we observe that the role of the innate immune pathway demonstrates heterogeneity,linked to the tumor development stage,pathway status,and specific cell *** propose that within the TME,the innate immune pathway exhibits multidimensional *** diversity is fundamentally rooted in cellular heterogeneity and is manifested as a variety of signaling *** pro-tumor effect of innate immune pathway activation essentially reflects the suppression of classical pathways and the activation of potential pro-tumor alternative *** our understanding of the tumor's innate immune pathway network and employing appropriate targeting strategies can enhance our ability to harness the anti-tumor potential of the innate immune pathway and ultimately bridge the gap from preclinical to clinicalapplication.

Adaptively Enhancing Facial Expression Crucial Regions via a Local Non-local Joint Network

Facial expression recognition(FER)is still challenging due to the small interclass discrepancy in facial expression *** view of the significance of facial crucial regions for FER,many existing studies utilize the prior information from some annotated crucial points to improve the performance of ***,it is complicated and time-consuming to manually annotate facial crucial points,especially for vast wild expression *** on this,a local non-local joint network is proposed to adaptively enhance the facial crucial regions in feature learning of FER in this *** the proposed method,two parts are constructed based on facial local and non-local information,where an ensemble of multiple local networks is proposed to extract local features corresponding to multiple facial local regions and a non-local attention network is addressed to explore the significance of each local *** particular,the attention weights obtained by the non-local network are fed into the local part to achieve interactive feedback between the facial global and local ***,the non-local weights corresponding to local regions are gradually updated and higher weights are given to more crucial ***,U-Net is employed to extract the integrated features of deep semantic information and low hierarchical detail information of expression ***,experimental results illustrate that the proposed method achieves more competitive performance than several state-of-the-art methods on five benchmark datasets.

Responses of arbuscular mycorrhizal fungi to straw return and nitrogen fertilizer reduction in a rainfed maize field

Straw return can be used to reduce fertilizer input and improve agricultural sustainability and soil ***,how straw return and reduced fertilizer application affect beneficial soil microbes,particularly arbuscular mycorrhizal fungi(AMF),remains poorly ***,we conducted a five-year field experiment in a rainfed maize field on the Loess Plateau of northwestern *** tested four treatments with straw return combined with four nitrogen(N)application rates,i.e.,100%,80%,60%,and 0%of the common N application rate(225 kg N ha^(-1)year^(-1))in this region,and two reference treatments(full or no N application),with three replicates for each *** colonization was quantified and AMF communities colonizing maize roots were characterized using Illumina *** virtual taxa(VTs)of AMF were identified in root samples,among which VT113(related to Rhizophagus fasciculatus)and VT156(related to Dominikia gansuensis)were the predominant *** root length colonization and AMF VT richness were sensitive to N fertilization,but not to straw return;furthermore,both gradually increased with decreasing N application *** VT composition of the AMF community was also affected by N fertilization,but not by straw return,and the community variation could be well explained by soil available N and phosphorus ***,60%,80%,and full N fertilization produced similar maize ***,our study revealed the response patterns of AMF to straw return and N fertilizer reduction and showed that straw return combined with N fertilizer reduction may be a promising practice to maintain mycorrhizal symbiosis concomitantly with crop productivity.
