Combination of chemopreventive and/or therapeutic agents is the imminent smart approach to cope upwith cancer because it may act on multiple targets through different pathways. In the present study, wehave synthesized multiple chemopreventive and/or therapeutic agents (Curcumin, Quercetin andAspirin) loaded nanoparticles by simple cation-anion interaction among the amine groups of chitosan(CS) and phosphate groups of sodium hexametaphosphate (SHMP). These nanosized bioactive materials(CS-SHMP-CQA-NPs) were well characterized and found most effective in colon cancer cell line (HCT-116)compared to other cancer cell lines. Triplex chemopreventive and/or therapeutic agents-loaded NPs weresynergistically inducing apoptosis in HCT-116 cells compared to two-chemopreventive agents-loadedNPs as evident by an increase in sub-G1 cells (percent), and chromatin condensation along with thedecrease in mitochondrial membrane potential (MMP). Interestingly, ChoueTalalay analysis revealedthat CS-SHMP-CQA-NPs showed strong synergistic effect in its all doses. Thus, our study demonstratesthat nanoparticles based bioactive materials significantly inhibit the growth of HCT-116 cells and thuscould be a promising approach for colon cancer chemoprevention.

Plots under conservation tillage may require higher amount of potassium(K) application for augmenting productivity due to its stratification in upper soil layers, thereby reducing K supplying capacity in a medium or long-term period. To test this hypothesis, a field experiment was performed in 2002-2003 and 2006-2007 to study the effect of K and several crop rotations on yield, water productivity, carbon sequestration, grain quality, soil K status and economic benefits derived in maize(Zea mays L)/cowpea(Vigna sinensis L.) based cropping system under minimum tillage(MT). All crops recorded higher grain yield with a higher dose of K(120 kg K2 O ha-1) than recommended K(40 kg K2 O ha-1). The five years' average yield data showed that higher K application(120 kg K2 O ha-1) produced 16.4%(P<0.05)more maize equivalent yield. Cowpea based rotation yielded 14.2%(P<0.05) higher production than maize based rotation. The maximum enhancement was found in cowpea-mustard rotation. Relationship between yield and sustainable indices revealed that only agronomic efficiency of fertilizer input was significantly correlated with yield. Similarly, higherdoses of K application not only increased the water use efficiency(WUE) of all crops, but also reduced runoff and soil loss by 16.5% and 15.8% under maize and 23.3% and 19.7% under cowpea cover, respectively. This study also revealed that on an average 16.5% of left over carbon input contributed to soil organic carbon(SOC). Here, cowpea based rotation with the higher K application increased carbon sequestration in soil. Potassium fertilization also significantly improved the nutritional value of harvested grain by increasing the protein content for maize(by 9.5%) and cowpea(by 10.6%). The oil content in mustard increased by 5.0% and 6.0% after maize and cowpea, respectively. An Improved Methodology to Overcome Key Issues in Human Fecal Metagenomic DNA Extraction

Microbes are ubiquitously distributed in nature, and recent culture-independent studies have highlighted the significance of gut microbiota in human health and disease. Fecal DNA is the primary source for the majority of human gut microbiome studies. However, further improvement is needed to obtain fecal metagenomic DNA with sufficient amount and good quality but low host genomic DNA contamination. In the current study, we demonstrate a quick, robust, unbiased, and cost-effective method for the isolation of high molecular weight （〉 23 kb） metagenomic DNA （260/280 ratio 〉 1.8） with a good yield （55.8 ± 3.8 ng/mg of feces）. We also confirm that there is very low human genomic DNA contamination （eubacterial： human genomic DNA marker genes = 227.9：1） in the human feces. The newly-developed method robustly performs for fresh as well as stored fecal samples as demonstrated by 16S rRNA gene sequencing using 454 FLX＋. Moreover, 16S rRNA gene analysis indicated that compared to other DNA extraction methods tested, the fecal metagenomic DNA isolated with current methodology retains species richness and does not show microbial diversity biases, which is further confirmed by qPCR with a known quantity of spike-in genomes. Overall, our data highlight a protocol with a balance between quality, amount, user-friendliness, and cost effectiveness for its suitability toward usage for cultureindependent analysis of the human gut microbiome, which provides a robust solution to overcome key issues associated with fecal metagenomic DNA isolation in human gut microbiome studies.

Objective： Fractures of the capitellum and trochlea constitute less than 1% of all elbow fractures and a shear fracture involving the capitellum and extending medially into most of the trochlea is rarely reported. Type Ⅳ capitellum fracture is still controversial in regard to its ra- diographic appearance, surgical approach and osteosynthesis. We report 10 cases of type Ⅳ capitellum fracture with a view to elucidating its clinical features and treatment outcome. Methods： We treated 10 patients of type Ⅳ capitellum fracture with a mean age of 32 years. A uniform surgical approach and postoperative rehabilitation were followed. Results： Nine patients presented to us after a mean of 4 days of injury and one patient was nonunion after 6 months of injury who had been treated conservatively by a bone setter. Double arc sign was absent in 6 cases. Intraopera- tively 6 capitellotrochlear fragments were devoid of soft tissue attachments. By Mayo Elbow Performance Score evaluation, 7 patients got excellent, 2 good and 1 fair results. One patient with associated elbow dislocation developed heterotopic ossification. There was no case of avascular necrosis, osteoarthrosis or fixation failures. Conclusions： Type Ⅳ capitellum fractures are rare and belong to complex articular injuries. A good functional out- come can only be achieved with open reduction and stable internal fixation followed by early mobilization. Preopera- tive radiographic assessment and computed tomography help surgeons in choosing the right surgical approach and implants. Good surgical technique and stable internal fixa- tion are the keys to early mobilization and good functional outcome.

AIM, To study the role of N-acetylcysteine （NAC） as a protective agent in rifampicin （RMP）-induced oxidative hepatic injury of young rats. METHODS： Hepatic injury was produced by giving 50mg/kg body weight/day of RMP for 3 wk. A dose of NAC （100mg/kg body weight/day） was given in combination with RMP intraperitoneally. Analysis of lipid peroxidation, thiol levels, cytochrome P4se, superoxide dismutase （SOD）, catalase, glutathione peroxidase, reductase and transferase were estimated in liver along with the body weight, liver weight and histological observations. RESULTS： RMP exposure resulted in no change in body and liver weight while antioxidative enzymes were altered but the non protein thiol （GSH） status was well preserved. Cytochrome P450 system and peroxidation of lipids were induced by RMP exposure. Partial protection was observed with NAC against RMP-induced changes in liver, which was evidenced from the prevention of increase in lipid peroxidation and the reduction in SOD and catalase enzyme levels. CONCLUSION. NAC protects young rats against RMP- induced oxidative hepatic injury.
