Role of the rhizosphere of a flooding-tolerant herb in promoting mercury methylation in water-level fluctuation zones

The water-level fluctuation zone(WLFZ) has been considered as a hotspot for mercury(Hg) methylation. Flooding-tolerant herbs are gradually acclimated to this water-land ecotone, tending to form substantial root systems for improving erosion resistance. Accompanying rhizosphere microzone plays crucial but unclear roles in methylmercury(Me Hg) formation in the WLFZ. Thus, we conducted this study in the WLFZ of the Three Gorges Reservoir, to explore effects of the rhizosphere of a dominant flooding-tolerant herb(bermudagrass) on Me Hg production. The elevated Hg and Me Hg in rhizosphere soils suggest that the rhizosphere environment provides favorable conditions for Hg accumulation and methylation. The increased bioavailable Hg and microbial activity in the rhizosphere probably serve as important factors driving Me Hg formation in the presence of bermudagrass. Simultaneously, the rhizosphere environments changed the richness, diversity, and distribution of hgc A-containing microorganisms. Here, a typical ironreducing bacterium( Geobacteraceae) has been screened, however, the majority of hgc A genes detected in rhizosphere, near-, and non-rhizosphere soils of the WLFZ were unclassified. Collectively, these results provide new insights into the elevated Me Hg production as related to microbial processes in the rhizosphere of perennial herbs in the WLFZ, with general implications for Hg cycling in other ecosystems with water-level fluctuations.

A novel hierarchical targeting and controllable smart nanoparticles for enhanced in situ nuclear photodynamic therapy

Photodynamic therapy(PDT)is a promising and non-invasive treatment for various cancers.Although nuclear PDT has considerable therapeutic prospects,it is still hindered by the non-specific recognition of tumor tissues or the degradation of nuclear targeting cationic groups by enzymes in the blood.Herein,a hierarchical targeted and controlled release strategy is proposed by using folate-modified poly-β-cyclodextrin(poly-β-CD)as a nano-carrier for loading nuclear localization signals(NLSs)-conjugated photosensitizer PAP(PAP=pyropheophorbide a-PAAKRVKLD).Excitingly,the obtained FA-CD@PAP(FA=folic acid)and nanoparticles(NPs)can specifically recognize tumor cells overexpressing folate receptors(FR)to remarkedly enhance the intracellular accumulation.Furthermore,the encapsulated PAP can be released under acidic conditions to realize precise nuclear localization.The reactive oxygen species(ROS)generated by the intranuclear-accumulated PAP upon irradiation can oxidize and destroy DNA chains or DNA repair enzymes instantaneously,which can directly induce cell death.As a result,FA-CD@PAP NPs exhibit excellent tumor regression and negligible side effects.This work provides an intelligent nuclear-targeted delivery strategy for in situ nuclear PDT with extremely prominent efficacy and high biological safety.

Carbon peaking and carbon neutralization trigger a technical revolution in energy&environment related fields.Development of new technologies for green energy production and storage,industrial energy saving and efficiency reinforcement,carbon capture,and pollutant gas treatment is in highly imperious demand.The emerging porous framework materials such as metal–organic frameworks(MOFs),covalent organic frameworks(COFs)and hydrogen-bonded organic frameworks(HOFs),owing to the permanent porosity,tremendous specific surface area,designable structure and customizable functionality,have shown great potential in major energy-consuming industrial processes,including sustainable energy gas catalytic conversion,energy-efficient industrial gas separation and storage.Herein,this manuscript presents a systematic review of porous framework materials for global and comprehensive energy&environment related applications,from a macroscopic and application perspective.

Nowadays,education and teaching have become a hot topic,and teaching in colleges and universities is facing a brand-new development direction.Principles of Concrete Structure Design,as one of the main courses,transmits professional knowledge for students,enhances the students’professional ability,and further carries out in-depth research on the course to bring a better teaching effect for students.The article mainly focuses on the research of the principles of concrete structure design course,conducts an analysis of the teaching characteristics of the principles of concrete structure design course,and reasonably sets the teaching content from the optimization of the course teaching objectives;innovative course teaching methods can deepen the effect of knowledge understanding;reform of experimental practice teaching can lay down the effect of the internalization of knowledge,etc.The in-depth description and discussion of the relevant aspects of the research aim to provide guidelines for related research.

Mercury bioaccumulation in fish in an artificial lake used to carry out cage culture

As a global toxic pollutant,mercury(Hg)bioaccumulation within food chain could be influenced by human disturbance.Ten typical fish species were collected from Changshou Lake,an artificial lake used to carry out cage fish culture,to investigate the C/N isotopic compositions and Hg bioaccumulation in fish.The results showed that the total Hg(THg)and methylmercury(MeHg)levels in fish muscles((56.03±43.96)and(32.35±29.57)ng/g,wet weight),comparable with those in most studies in China,were significantly lower than the international marketing limit(0.5 mg/kg).Past human input for cage culture in this lake led to abnormal^(15)N enrichment in food chain,as the quantitative trophic levels based onδ^(15)N were different with that classified by feeding behaviors.This phenomenon subsequently demonstrated that it should be considered thoughtfully with respect to the application of the traditional method for understanding Hg bioaccumulation power by the slope of log_(10)[Hg]withδ^(15)N regression in specific water body(i.e.,Changshou Lake).In addition,no significant linear correlation between Hg and body weight or length of some fish species was observed,suggesting that the fish growth in the eutrophic environment was disproportionate with Hg bioaccumulation,and fish length or weight was not the main factor affecting Hg transfer with food web.The occurrence of human disturbance in aquatic system presents a challenge to a better understanding of the Hg bioaccumulation and biomagnification within the food chain.

Erlotinib Analogue-substituted Zinc（Ⅱ） Phthalocyanines for Small Molecular Target-based Photodynamic Cancer Therapy

＂Smart＂ targeted photosensitizer conjugated with small molecule target-based anticancer drug which has a sim- ple chemical structure and high stability, is a new promising targeted therapeutic strategy. We herein extended this strategy and reported a novel series of zinc（II） phthalocyanine-erlotinib analogue conjugates with different periph- eral substituted positions and lengths of the linker. Having erlotinib analogue as the targeting moiety, all conjugates exhibited high specificity and potent affinity to HepG2 cancer cells and kept high photodynamic activity （ICs0 = 3.7 -- 16.7 nmol/L）. Structure-activity relationships of these conjugates were assessed by investigating their photophys- ical/photochemical properties, targeting intracellular uptake and in vitro phototoxicity. The results suggested that a-substituted conjugates showed slightly higher photodynamic activity than ,8-substituted ones. In conclusion, we have developed a series of promising anticancer agents with high tumor selectivity and anticancer activity for targeted photodynamic therapy.

Photocyanine:A novel and effective phthalocyanine-based photosensitizer for cancer treatment

As one of the three key components of photodynamic therapy(PDT),photosensitizers(PSs)greatly influence the photodynamic efficiency in the treatment of tumors.Photosensitizers with tetrapyrrole structure,such as porphyrins,chlorins and phthalocyanines,have been extensively investigated for PDT and some of them have already received clinical approval.However,only a few of porphyrin-based photosensitizers are available for clinical applications,and PDT has not received wide recognition in clinical practice.In this regard,PSs remain a limiting factor.Our research focuses on the rational design of new PSs.Photocyanine,a Zinc(Ⅱ)phthalo-cyanine(ZnPc)type photosensitizer with low dark toxicity and high single oxygen quantum yield,is one of the promising PSs candidates and currently being tested in clinical trials.Here,we present an overview on the development of Photocyanine,including its design,synthesis,purification,characterization and preclinical studies,wishing to contribute to the research of more promising PSs.

A Highly Selective and Turn-on Fluorescent Probe for Fe^(3+) Ion Based on Perylene Tetracarboxylic Diimide

We have demonstrated a turn-on fluorescent sensor 6 for detection of Fe^(3+) based on photo-induced electron transfer(PET)mechanism.The probe comprises a perylene tetracarboxylic diimide(PDI)fluorophore and two bis((1,2,3-triazol-4-yl)methyl)amine(DTA)moieties as the metal ion receptors.It exhibits high selectivity toward Fe^(3+) over various other metal ions in CH_(3)CN/H_(2)O(1∶1,V/V).The binding stoichiometry for 6-Fe^(3+) complexes has been determined to be 1∶2 by a Job plot of fluorescence.The association constant between 6 and Fe^(3+) was estimated to be 1.04×10^(10)(mol/L)^(−2) by Benesi-Hildebrand equation.

新型冠状病毒感染疫情对人类健康和全球经济造成重创。健康相关脆弱人群的免疫功能减退,导致疫苗的防护效果不足,并且,感染新型冠状病毒后重症和死亡风险更高,而目前尚缺乏足够有针对性的新型冠状病毒感染预防和治疗药物。在我国“乙类乙管”的防疫背景下,脆弱人群成为疫情防控的重点人群。因此,针对脆弱人群应该进一步优化个体免疫和防控策略,在疫苗之外,还需要补充其他预防手段,如长效中和抗体。基于此,本文将对脆弱人群的识别、免疫功能特点与预防方法进行综述,为国内健康相关脆弱人群的防治策略提供参考,期待未来可以研发出更适宜脆弱人群的预防药物,降低脆弱人群感染新型冠状病毒的风险。
