Impaired tricarboxylic acid cycle flux and mitochondrial aerobic respiration during isoproterenol induced myocardial ischemia is rescued by bilobalide

Reclaimed water has been widely applied in irrigation and industrial *** the behavior of emerging contaminants in the production process of reclaimed water is the first prerequisite for developing relevant water quality *** study investigated 43 emerging contaminants,including 22 pharmaceuticals and personal care products(PPCPs),11 organophosphorus flame retardants(OPFRs),and 10 antibiotic resistance genes(ARGs)in 3 reclaimed wastewater treatment plants(RWTPs)in *** composition profiles and removal efficiencies of these contaminants in RWTPs were *** results indicated that the distribution characteristics of the different types of contaminants in the three RWTPs were ***,sul2 and tris(1-chloro-2-propyl)phosphate were the dominant substances in the wastewater,and their highest concentrations were 27104 ng/L,1.4×10^(7) copies/mL and 262 ng/L,*** and sul2 were observed to be the dominant substances in the sludge,and their highest concentrations were 5419 ng/g and 3.7×10^(8) copies/g,***/anoxic/oxic system combined with the membrane bioreactor process achieved a relatively high aqueous removal of PPCPs(87%).ARGs and OPFRs were challenging to remove,with average removal rates of 6.5%and 31%,*** meta-analysis indicated that tertiary treatment processes performed better in emerging contaminant removal than secondary *** exhibited the highest mass load discharge,with 33.5 mg/d per 1000 *** and tris(2-ethylhexyl)phosphate posed high risks(risk quotient>1.0)to aquatic *** study provides essential evidence to screen high priority pollutants and develop corresponding standard in RWTPs.

Thymus is the important immune organ,responsible for T cell development and *** lower circulating T counts have been observed in patients who died from COVID-19 compared with ***,also known as FNC,is a thymus-homing anti-SARS-CoV-2 drug in treating COVID-19 *** this study,single-cell transcriptome,proteomics,and parallel reaction monitoring(PRM)were applied to insight into the activation process of FNC in rat and SARS-CoV-2 rhesus monkey *** results indicated that thymic immune cells possess a robust metabolic capacity for cytidine-analogue drugs such as *** enzymes involved in the FNC phosphorylation process,such as Dck,Cmpk1,and Nme2,were highly expressed in CD4+ T cells,CD8+ T cells,and DP(CD4+ CD8+)***,FNC could upregulate multiple phosphorylated kinases in various cell types while downregulating the phosphatases,phosphoribosyl transferases,and deaminases,*** robust phosphorylation capacity of the thymus for cytidine analogue drug FNC,and the activation effect of FNC on the NAs metabolism system potentially contribute to its enrichment in the thymus and immune protection *** suggests that it is crucial to consider the expression level of phosphorylation kinases when evaluating NA drug properties,as an important factor during antiviral drug design.

Pulmonary fibrosis (PF) is a chronic progressive end-stage lung disease. However, the mechanisms underlying the progression of this disease remain elusive. Presently, clinically employed drugs are scarce for the treatment of PF. Hence, there is an urgent need for developing novel drugs to address such diseases. Our study found for the first time that a natural source of Prismatomeris connata Y. Z. Ruan (Huang Gen, HG) ethyl acetate extract (HG-2) had a significant anti-PF effect by inhibiting the expression of the transforming growth factor beta 1/suppressor of mothers against decapentaplegic (TGF-β1/Smad) pathway. Network pharmacological analysis suggested that HG-2 had effects on tyrosine kinase phosphorylation, cellular response to reactive oxygen species, and extracellular matrix (ECM) disassembly. Moreover, mass spectrometry imaging (MSI) was used to visualize the heterogeneous distribution of endogenous metabolites in lung tissue and reveal the anti-PF metabolic mechanism of HG-2, which was related to arginine biosynthesis and alanine, asparate and glutamate metabolism, the downregulation of arachidonic acid metabolism, and the upregulation of glycerophospholipid metabolism. In conclusion, we elaborated on the relationship between metabolite distribution and the progression of PF, constructed the regulatory metabolic network of HG-2, and discovered the multi-target therapeutic effect of HG-2, which might be conducive to the development of new drugs for PF.

Background: The brain bioavailability of novel small molecules developed to address central nervous system disease is classically documented through ex vivo or in vivo analyses conducted in rodent models. Data acquired in rodent models are, however,not easily transferrable to human as the pharmacokinetic and pharmacodynamics profiles of the species are quite ***: Using drugs selected for their differential transport across the blood-brain barrier, we here demonstrate the feasibility of brain microdialysis in normal vigil macaque monkey by measuring brain extracellular fluid bioavailability of carbamazepine, digoxin, oxycodone, and ***: All drugs, but digoxin, were found in dialysate samples. Drugs that are substrate of P-glycoprotein show a difference of bioavailability or brain pharmacokinetic parameters between rodents and ***: Data suggest that brain microdialysis in vigil macaque monkey, the species of choice for classic pharmacokinetic/pharmacodynamics studies could help predicting human brain bioavailability of a small molecule depending on the protein involved in the efflux transport from the brain.

Application of Medical Failure Mode and Effect Analysis in Preoperative Preparation for Coronary Intervention

[Objectives]To explore application effect of healthcare failure mode and effect analysis(HFMEA)in preoperative management of coronary intervention.[Methods]The preoperative management process of 455 patients who underwent coronary intervention surgery from January to March in 2022 was sorted out,and the failure modes and causes in the preoperative management were *** priority risk value was calculated,and the top 6 failure modes were found *** measures were formulated according to the causes of failure *** months after the implementation of the procedure,470 patients who underwent coronary intervention surgery from May to July in 2022 were selected as the intervention group,and the optimized preoperative preparation procedure by HFMEA was *** decrease of risk priority number(RPN)and RPN value of two high-risk failure modes,as well as changes in the rate of the time of taking dual-antiplatelet drugs reaching the standard,the rate of passing the dress code,the rate of timely signing of informed consent,and the rate of filling in the handover form of preoperative preparation were analyzed.[Results]The rate of the time of taking dual-antiplatelet drugs reaching the standard,the rate of passing the dress code,the rate of timely signing of informed consent,and the rate of filling in the handover form of preoperative preparation in experimental groups were higher than those of the control group,and the difference was statistically significant(P<0.05).[Conclusions]Applying HFMEA to the management of preoperative preparation process for coronary intervention can effectively improve the timeliness and qualification rate of preoperative preparation for coronary intervention,and provide guarantee for the safe and smooth implementation of coronary intervention.

Study on the Health Status of Children and Adolescents under the Influence of COVID-19

Background: The COVID-19 epidemic is a public health security event affecting the whole country, which has a certain impact on the development and current situation of the health field. As a special group, children and adolescents need to receive attention and special care. Purpose: To study the health status of children and adolescents under the influence of COVID-19. Method: To collect and analyze the physical, mental and mental health data of 448 children aged 3 - 18 years in the Pediatric Medical Center of the Second Hospital of Jilin University since the COVID-19 outbreak, so as to grasp the physical, mental and mental health status and existing problems of children and adolescents under the impact of COVID-19 outbreak. Results: Food (seafood, mangoes, dairy products, animal protein, soy, peanuts, etc.) is the main allergen in the children and adolescents who participated in the survey, followed by inhalation (pollen, cat and dog hair, etc.). The diet was relatively normal, but there were still unhealthy eating behaviors, the detection rate was 5.36%. High calorie diet, high cholesterol diet, high salt diet, low protein diet, liquid diet in order from high to low, 85.27% of the normal water intake, drinking behavior was greatly affected by the family environment and school environment. The main problem affecting children’s digestion is constipation.
