Regulation of specific abnormal calcium signals in the hippocampal CA1 and primary cortex M1 alleviates the progression of temporal lobe epilepsy

Temporal lobe epilepsy is a multifactorial neurological dysfunction syndrome that is refractory,resistant to antiepileptic drugs,and has a high recurrence rate.The pathogenesis of temporal lobe epilepsy is complex and is not fully understood.Intracellular calcium dynamics have been implicated in temporal lobe epilepsy.However,the effect of fluctuating calcium activity in CA1 pyramidal neurons on temporal lobe epilepsy is unknown,and no longitudinal studies have investigated calcium activity in pyramidal neurons in the hippocampal CA1 and primary motor cortex M1 of freely moving mice.In this study,we used a multichannel fiber photometry system to continuously record calcium signals in CA1 and M1 during the temporal lobe epilepsy process.We found that calcium signals varied according to the grade of temporal lobe epilepsy episodes.In particular,cortical spreading depression,which has recently been frequently used to represent the continuously and substantially increased calcium signals,was found to correspond to complex and severe behavioral characteristics of temporal lobe epilepsy ranging from gradeⅡto gradeⅤ.However,vigorous calcium oscillations and highly synchronized calcium signals in CA1 and M1 were strongly related to convulsive motor seizures.Chemogenetic inhibition of pyramidal neurons in CA1 significantly attenuated the amplitudes of the calcium signals corresponding to gradeⅠepisodes.In addition,the latency of cortical spreading depression was prolonged,and the above-mentioned abnormal calcium signals in CA1 and M1 were also significantly reduced.Intriguingly,it was possible to rescue the altered intracellular calcium dynamics.Via simultaneous analysis of calcium signals and epileptic behaviors,we found that the progression of temporal lobe epilepsy was alleviated when specific calcium signals were reduced,and that the end-point behaviors of temporal lobe epilepsy were improved.Our results indicate that the calcium dynamic between CA1 and M1 may reflect specific epileptic behaviors corresponding to diff

In an effort to clarify the formation mechanism of LPSO structure in Mg-Y-Zn alloy,the chemical environment and structural ordering in liquid Mg-rich Mg-Y-Zn system are investigated with the aid of ab-initio molecular dynamics simulation.In liquid Mg-rich Mg-Y alloys,the strong Mg-Y interaction is determined,which promotes the formation of fivefold symmetric local structure.For Mg-Zn alloys,the weak Mg-Zn interaction results in the fivefold symmetry weakening in the liquid structure.Due to the coexistence of Y and Zn,the strong attractive interaction is introduced in liquid Mg-Y-Zn ternary alloy,and contributes to the clustering of Mg,Y,Zn launched from Zn.What is more,the distribution of local structures becomes closer to that in pure Mg compared with that in binary Mg-Y and Mg-Zn alloys.These results should relate to the origins of the Y/Zn segregation zone and close-packed stacking mode in LPSO structure,which provides a new insight into the formation mechanism of LPSO structure at atomic level.

Lactate is present at a high level in the microenvironment of mammalian preimplantation embryos in vivo and in vitro. However, its role in preimplantation development is unclear. Here, we report that lactate is highly enriched in the nuclei of early embryos when major zygotic genome activation(ZGA) occurs in humans and mice. The inhibition of its production and uptake results in developmental arrest at the 2-cell stage, major ZGA failure, and loss of lactate-derived H3K18lac, which could be rescued by the addition of Lac-Co A and recapitulated by overexpression of H3K18R mutation. By profiling the landscape of H3K18lac during mouse preimplantation development, we show that H3K18lac is enriched on the promoter regions of most major ZGA genes and correlates with their expressions. In humans, H3K18lac is also enriched in ZGA markers and temporally concomitant with their expressions. Taken together, we profile the landscapes of H3K18lac in mouse and human preimplantation embryos, and demonstrate the important role for H3K18lac in major ZGA, showing that a conserved metabolic mechanism underlies preimplantation development of mammalian embryos.

Numerical analysis on the transport properties and residence time distribution of ribbon biomass particles in a riser reactor based on CFD-DEM approach

A bended ribbon biomass particle model was developed to explore the dynamic transport properties inside a riser reactor.Residence time distribution(RTD)of the particles was analyzed by using the Eulerian-Lagrange method.The effects of sampling height,particle density,particle size and gas-to-solid mass ratio on RTD were investigated.The coupled Computational Fluid Dynamics and Discrete Element Method(CFD-DEM)model was verified firstly by experimental data on pressure drop and residence time distribution density function.The simulation results demonstrated that the ribbon biomass particles display a typical annular-core spatial distribution during transportation.The RTD of particles exhibit an approximate single-peaked normal distribution.The mean residence time(MRT)can reach up to 0.7 s when the particle density is 1200 kg/m^(3).Particle with higher density has longer mean residence time.The flow patterns are closer to plug flow if particle length over 12 mm.The particle flow pattern is not sensitive to changes in particle density and size,while the gas-to-material mass ratio has a significant impact on it.

A breathable, designable and flexible leather–heater used in wearable thermotherapy

Flexible heaters with personal thermal management capabilities have great potential in thermal therapy applications due to their excellent flexibility, low power consumption, and portability. However, manufacturing wearable heating devices that are breathable, wear resistant and conformal for long-term use is still challenging. To address these issues, we designed a leather heater using breathable, biocompatible, and tailorable leather as the substrate through a simple in-situ polymerization polypyrrole strategy. This heater exhibits excellent heating and mechanical properties（reaching 64°C at a voltage of 5 V with efficient Joule heat generation of 2286 W/m2and uniform temperature distribution, and functioning properly after 1000 cycles of bendability tests）. In addition, this heater displays better wear resistance and water vapor permeability rate(38.04 g/（m2h）). The cuttable and sewable of leather gives the strategy ability to be flexibly designed to mold the heater to the specific requirements of different body parts, providing a new approach to wearable thermal therapy.

Apparent activation energy of mineral in open pit mine based upon the evolution of active functional groups

This study aimed to investigate the mechanism of mineral spontaneous combustion in an open pit. On the study of coal and mineral mixture in open pit mines, as well as through the specifc surface area and Search Engine Marketing (SEM) experiments, the specifc surface area and aperture characteristics of distribution of open pit coal sample and pit mineral mixture samples were analyzed. Thermal analysis experiments were used to divide the oxidation process was divided into three stages, and the thermal behavior characteristics of experimental samples were characterized. On the basis of the stage division, we explored the transfer law of the key active functional groups of the experimental samples. The apparent activation energy calculation of the key active groups, performed by combining the Achar diferential method with the Coats–Redfern integral method, microstructural and oxidation kinetic properties were revealed. The resulted showed that the mixed sample had high ash, the fxed carbon content was reduced, the specifc surface area was far lower than the raw coal, the large aperture distribution was slightly higher than the medium hole, the micropore was exceptionally low, the gas adsorption capacity was weaker than the raw coal, the pit coal sample had the exceedingly more active functional groups, easy to react with oxygen, more likely to occur naturally, and its harm was relatively large. The mixed sample contained the highest C–O–C functional group absorbance. The functional groups were mainly infuenced by the self-OH content, alkyl side chain, and fatty hydrocarbon in the sample. The main functional groups of the four-like mixture had the highest apparent activation energy, and the two reactions were higher in the low-temperature oxidation phase.

Temperature variation and gas generation at diferent depths and positions in the coal combustion process were studied to determine the propagation and evolution of high temperature regions in the process of coal spontaneous combustion.This study selected coal samples from Mengcun,Shaanxi Province,People’s Republic of China,and developed a semi-enclosed experimental system(furnace)for simulating coal combustion.The thermal mass loss of coal samples under various heating rates(5,10,and 15℃/min)was analyzed through thermogravimetric analysis,and the dynamic characteristics of the coal samples were analyzed;the reliability of the semi-enclosed experimental system was verifed through the equal proportional method of fuzzy response.The results reveal that the high-temperature zone is distributed nonlinearly from the middle to the front end of the furnace,and the temperatures of points in this zone decreased gradually as the layer depth increased.The apparent activation energy of the coal samples during combustion frst increased and then decreased as the conversion degree increased.Furthermore,the proportion of mass loss and the mass loss rate in the coal samples observed in the thermogravimetric experiment is consistent with that observed in the frst and second stages of the experiment conducted using the semi-enclosed system.The research fndings can provide a theoretical basis for the prevention and control of hightemperature zones in coal combustion.

Facile Construction of Carbon Dots Layer and Oxygen Vacancies Simultaneously onto Ti0_(2) to Enhance Photoreduction Activity

We reported a facile and green hydrothermal strategy to tailor the carbon dots layer(CDL)and oxygen vacancies(Vo)modified on Ti0_(2),by using ascorbic acid as CDL precursor and Vo regulator.The amorphous CDL was uniformly coated on the surface of the as prepared CDL@TiO_(2-x) composites by Ti-O-C bond.With the increase of carbon content,the amorphous CDL gradually became thicker,while the Vo concentration first increased and then decreased.1.5 CDL@TiO_(2-x) possessed the highest Vo concentration.With the decoration of CDL and Vo,the photoreduction properties of CDL@TiO_(2-x) composites were significantly improved.Under simulated solar light and visible light,the reaction rates of 1.5 CDL@TiO_(2-x) were 9.6 times and 216 times higher than that of pure Ti0_(2)/respectively.Interestingly,the results discovered that CDL and Vo influenced the properties of CDL@TiO_(2-x) composites in different ways.The primary function of CDL was to improve the utilization of solar light,and the enhancement of the charge transfer ability was decided by Vo and CDL together.Our work provides a new strategy for the modification of Ti0_(2) and sheds new light on the respective role of CDL and Vo when they coexist in the catalyst.

PIP5k1βis crucial to the generation of phosphotidylinosotol(4,5)P2.PIP5k1βparticipates in numerous cellular activities,such as B cell and platelet activation,cell phagocytosis and endocytosis,cell apoptosis,and cytoskeletal organization.In the present work,we aimed to examine the function of PIP5k1βin osteoclastogenesis and osteogenesis to provide promising strategies for osteoporosis prevention and treatment.We discovered that PIP5k1β deletion in mice resulted in obvious bone loss and that PIP5k1β was highly expressed during both osteoclast and osteoblast differentiation.Deletion of the gene was found to enhance the proliferation and migration of bone marrow-derived macrophage-like cells to promote osteoclast differentiation.PIP5k1β-/-osteoclasts exhibited normal cytoskeleton architecture but stronger resorption activity.PIP5kip deficiency also promoted activation of mitogen-activated kinase and Akt signaling,enhanced TRAF6 and c-Fos expression,facilitated the expression and nuclear translocation of NFATC1,and upregulated Grb2 expression,thereby accelerating osteoclast differentiation and function.Finally,PIP5k1β enhanced osteoblast differentiation by upregulating master gene expression through triggering smad1/5/8 signaling.Therefore,PIP5k1βmodulates bone homeostasis and remodeling.

Ore-forming elements diffusion and distribution in the altered host rock surrounding the Koktokay No.3 pegmatite in the Chinese Altay

Petrography and geochemistry of the altered and unaltered host rocks surrounding the Koktokay No.3 pegmatite revealed that the unaltered amphibolite is mainly composed of hornblende, plagioclase, and ilmenite.Beyond these primary components, the altered host rocks contain a few newly formed minerals, including biotite,tourmaline, chlorine, and muscovite. The alteration zone surrounding the Koktokay No.3 pegmatite is limited to 2.0 m, characterized by biotitization, tourmalization, and chloritization. In the altered host rocks, the contents of SiO_2, MgO, MnO, Na_2O, and TiO_2 did not vary greatly.However, Al_2O_3 showed a weak decreasing trend with the increasing distance from the pegmatite contact zone, while Fe_2O_3 and CaO showed an increasing trend. The contents of Li, Rb, and Cs in the altered host rocks were much higher than those in the unaltered host rocks, decreasing with distance from the contact. The chondrite-normalized rare earth element(REE) pattern of the altered and unaltered host rock was right-inclined from La to Lu, but enriched in light REEs over heavy REEs after hydrothermal alteration. An isocon plot shows that some oxides migrated in with an order of P_2O_5>K_2O >TiO_2>Al_2O_3>SiO_2>MnO≥MgO, while others migrated out with an order of Na_2O >CaO >Fe_2O_3. For REEs, the migration ratios are positive values withCs >Rb >Li >Nb >Ta >Be, signifying that all REEs migrated from the exsolved magmatic fluid into the altered host rocks. It was concluded that diffusion was the only mechanism for migration of ore-forming elements in the alteration zone. The effective diffusion coefficients(Deff)of LiF, RbF, and CsF were estimated under a fluid temperature of 500–550 °C. Using a function of concentration(C(x,t)) and distance(x), the order of migration distance was determined to be LiF >CsF >RbF, with diffusion times of (3.39 ± 0.35)× 10~6,(3.19 ± 0.28) × 10~5 and(6.33 ± 0.05) × 10~5 years, respectively.
