Highly pathogenic avian influenza(HPAI)subtype H5Nx viruses have spread globally and are a major concern for poultry,wild birds,mammals,and even humans(de Vries et al.2015;Zeng et al.2022).The hemagglutinin(HA)genes of H5 subtype viruses have evolved into multiple clades and some of these clades have been further divided into subclades(Cui et al.2022).Clade 2.***.4.4H5N8 HPAI viruses(HPAIVs)have caused several waves of disease outbreaks in wild birds and domestic poultry(Wang et al.2022).

普通PCR引物对目的基因扩增的单一特异性,限制了其在临床疾病检测中的通用性范围,而多重PCR需要避免若干引物对之间二级结构的干扰,应用同样受到一定的限制。简并PCR技术中简并性引物是基于基因保守区域的分析并结合简并碱基而设计,可以检测出高度相似的靶基因,具有高效性、灵敏性、特异性和低成本的特点,目前已被广泛应用于多个领域。综述了简并PCR技术在临床、食品安全、动植物疫病、寄生虫、水产养殖等领域疾病检测中的应用,并对其优缺点进行了讨论,以期为简并PCR的进一步发展以及在其他领域的应用提供参考和指导。

This study assessed and compared the immunogenicity of various immunization strategies in mice using combinations of recombinant DNA(pCCMp24) and recombinant attenuated vaccinia virus Tian Tan(rddVTT-CCMp24).Intramuscular immunization was performed on days 0(prime) and 21(boost).The immunogenicity of the vaccine schedules was determined by measuring human immunodeficiency virus(HIV)-specific binding antibody levels and cytokine(interleukin-2 and interleukin-4) concentrations in peripheral blood,analyzing lymphocyte proliferation capacity against HIV epitopes and CD4 + /CD8 + cell ratio,and monitoring interferon-gamma levels at different times *** results showed that pCCMp24,rddVTT-CCMp24 and their prime-boost immunization induced humoral and cellular immune *** pCCMp24/rddVTT-CCMp24 immunization strategy increased CD8 + T cells and induced more IFN-γ-secreting cells compared with single-shot *** prime-boost immunization strategy also induced the generation of cellular immunological memory to HIV epitope *** results demonstrated that prime-boost immunization with rDNA and rddVTT-CCMp24 had a tendency to induce greater cellular immune response than single-shot vaccinations,especially IFN-γ response,providing a basis for further studies.

In this paper, a type of general nonholonomic systems is proposed, which contains both the Brockett’s two example systems, and their extended n-dimensional chained forms, as special cases. For the stabilization of such systems, a stabilizing controller is proposed based on the fully actuated system(FAS)approach, which is discontinuous at the origin but time-invariant when the open-loop system is time-invariant,and drives the feasible trajectories of the system to the origin exponentially. Furthermore, the proposed FAS approach is also extended to the sub-normal system case and the time-delay system case.

利用巨细胞病毒(cytomegalovirus,CMV)启动子,以pUC57质粒为骨架,设计了一种多启动子的三独立表达框载体pCMV-3MCS。为验证所构建的质粒系统,在3个表达框中依次插入绿色荧光蛋白(enhanced green fluorescent protein,EGFP)、红色荧光蛋白(mCherry)、蓝色荧光蛋白(blue fluorescent protein,BFP),转染293细胞后,在荧光显微镜下可观察到明显的绿、红、蓝三色荧光。为更广泛的应用所设计的表达系统,将犬瘟热病毒(canine distemper virus,CDV)的N、P和L基因依次插入表达框中,与携带EGFP的病毒mini基因组(EGFP反向插入mini基因组)共转染后,可观察到明显的荧光。结果表明,利用pCMV-3MCS载体可以使N、P、L蛋白通过单质粒转染实现各自蛋白表达,可以容纳近10 kb的外源基因插入,且功能不受影响。这为多质粒共转染转变为单质粒转染提供参考,也为进一步提高副黏病毒的拯救效率提供新的思路。

In this paper, continuous time-varying stabilizing controllers for the type of general nonholonomic systems proposed and treated in part 1 are designed using the fully actuated system(FAS) approach. The key step is to differentiate the first scalar equation, and by control of the obtained second-order scalar system, a proportional plus integral feedback form for the first control variable is obtained. With the solution to this designed second-order scalar system, the rest equations in the nonholonomic system form an independent time-varying subsystem which is then handled by the FAS approach. The overall designed controller contains an almost arbitrarily chosen design parameter, and is proven to guarantee the uniformly and globally exponential stability of the closed-loop system. The proposed approach is simple and effective,and is demonstrated with a practical example of ship control.

This paper studies a robust fault compensation and vibration suppression problem of flexible hypersonic *** controlled plant is represented by a cascade system composed of a nonlinear Ordinary Differential Equation(ODE)and an Euler-Bernoulli Beam Equation(EBBE),in which the vibration dynamics is coupled with the rigid dynamics and suffers from distributed faults.A state differential transformation is introduced to transfer distributed faults to an EBBE boundary and a longitudinal dynamics is refined by utilizing T-S fuzzy IF-THEN rules.A novel T-S fuzzy based fault-tolerant control algorithm is developed and related stability conditions are *** robust exponential stability and well-posedness are proved by using the modified l_(0)-semigroup based Lyapunov direct approach.A simulation study on the longitudinal dynamics of flexible hypersonic vehicles effectively verifies the validity of the developed theoretical results.
