Pregnancy outcomes in papillary thyroid cancer survivors:A retrospective cohort study

Objective:The purpose of this study was to evaluate the pregnancy outcomes among papillary thyroid cancer survivors.Methods:65 cases singleton gravidas of papillary thyroid cancer survivors treated in the Department of Obstetrics,Peking University People's Hospital between January 2011 and January 2020 were selected retrospectively as the study group,and 130 cases of singleton pregnancy during the same time were randomly selected as the control group.The differences in general conditions,maternal and fetal complications between the two groups were analyzed.Risk factors of maternal complications and fetal malformation in patients with papillary thyroid cancer survivors were analyzed.Data was analyzed with t-test,χ^(2)test and logistic regression for statistical differences.Results:Totally 65 cases of papillary thyroid cancer with 70 pregnancies were analyzed,which including 65 cases achieved live birth,5 cases underwent induced labor because of fetal malformation.The study group showed higher incidence in gestational diabetes(20%vs 10%),cesarean section(39.1%vs 24.6%),postpartum hemorrhage(350193 ml vs 290181 ml)and fetal malformation(5.7%vs 1.5%),and lower live birth rate(91.4%vs 99.2%)(P<0.05).Multivariate analysis by logistic regression found that the patient's age,thyroid function,operation mode and postoperative I-131 treatment were not the risk factors which may affect the pregnancy outcomes or fetal malformation.Conclusion:Elevated maternal and fetal complications were observed among pregnancies in papillary thyroid cancer survivors.Management of these survivors may require a careful gestation monitoring,and further investigation is needed.

Archetype identification and urban building energy modeling for city-scale buildings based on GIS datasets

Urban building energy modeling has become an efficient way to understand urban building energy use and explore energy conservation and emission reduction potential.This paper introduced a method to identify archetype buildings and generate urban building energy models for city-scale buildings where public building information was unavailable.A case study was conducted for 68,966 buildings in Changsha city,China.First,clustering and random forest methods were used to determine the building type of each building footprint based on different GIS datasets.Then,the convolutional neural network was employed to infer the year built of commercial buildings based on historical satellite images from multiple years.The year built of residential buildings was collected from the housing website.Moreover,twenty-two building types and three vintages were selected as archetype buildings to represent 59,332 buildings,covering 87.4%of the total floor area.Ruby scripts leveraging on OpenStudio-Standards were developed to generate building energy models for the archetype buildings.Finally,monthly and annual electricity and natural gas energy use were simulated for the blocks and the entire city by EnergyPlus.The total electricity and natural gas use for the 59,332 buildings was 13,864 GWh and 23.6×10^(6) GJ.Three energy conservation measures were evaluated to demonstrate urban energy saving potential.The proposed methods can be easily applied to other cities in China.

HSV-2-encoded miRNA-H4 Regulates Cell Cycle Progression and Act-Dinduced Apoptosis in HeLa Cells by Targeting CDKL2 and CDKN2A

MicroRNAs(miRNAs)encoded by latency-associated transcript are associated with both latent and acute stages of herpes simplex virus 2(HSV-2)infection.In this study,miRNA-H4-5p and miRNA-H4-3p were ectopically expressed in HeLa cells to explore potential cellular targets of viral miRNAs and demonstrate their potential biological functions.The results showed that miRNA-H4-5p could reverse apoptosis induced by actinomycin D(Act-D)and promote cell cycle progression,but miRNA-H4-3p had no such obvious functions.Bioinformatics analysis,luciferase report assay,quantitative reverse transcription polymerase chain reaction(qRT-PCR),and Western blotting demonstrated that miRNA-H4-5p could bind to the 3-′untranslated region(UTR)of cyclin-dependent kinase inhibitor 2A(CDKN2A)and cyclin-dependent kinase-like 2(CDKL2)to negatively regulate their expression.We verified that these two targeted genes were associated with cell apoptosis and cell cycle.Furthermore,in HeLa cells infected with HSV-2,we detected significantly reduced expression of CDKN2A and CDKL2 and demonstrated the negative regulation effect of miRNA-H4-5p on these two target genes.Our findings show that viral miRNAs play a vital role in regulating the expression of the host's cellular genes that participate in cell apoptosis and progression to reshape the cellular environment in response to HSV-2 infection,providing further information on the roles of encoded herpesvirus miRNAs in pathogen-host interaction.

To provide preferential protection for users while keeping good service utility, a preferential private recommendation framework(named as PrefER) is proposed. In this framework, a preferential budget allocation scheme is designed and implemented at the system side to provide multilevel protection.And users’ preference is utilized at the user side to improve recommendation performance without increasing users’ burden. This framework is generic enough to be employed with other schemes. Recommendation accuracy based on the MovieLens dataset by the collaborative filtering schemes and PrefER are compared and analyzed.The experimental results show that PrefER can provide preferential privacy protection with the improvement of recommendation accuracy.

With the recent increase in routing App usage, it is urgent to analyze the impact of inevitable information delay in routing Apps on traffic.To analyze the negative impact of such delayed routing Apps on traffic, this paper investigates the impact of ideal and delayed routing Apps on traffic. Firstly, the effect of ideal routing Apps has been explored with theoretical analysis and derivation based on a macroscopic network model. Then, an extended network model is built to characterize the delayed routing.The impact of such delayed routing Apps is investigated by simulation experiments while considering routing usage proportion and amount of delay. The results demonstrate that ideal navigation does improve traffic efficiency, but in some cases, delayed navigation is even worse than no navigation.

N-heterocyclic phosphines(NHPs) have recently emerged as a new group of promising catalysts for metal-free reductions, owing to their unique hydridic reactivity. The excellent hydricity of NHPs, which rivals or even exceeds those of many metal-based hydrides, is the result of hyperconjugative interactions between the lone-pair electrons on N atoms and the adjacent σ*(P–H) orbital. Compared with the conventional protic reactivity of phosphines, this umpolung P–H reactivity leads to hydridic selectivity in NHP-mediated reductions. This reactivity has therefore found many applications in the catalytic reduction of polar unsaturated bonds and in the hydroboration of pyridines. This review summarizes recent progress in studies of the reactivity and synthetic applications of these phosphorus-based hydrides, with the aim of providing practical information to enable exploitation of their synthetically useful chemistry.

Cancer is a major threat to human health.Among various treatment methods,precision therapy has received significant attention since the inception,due to its ability to efficiently inhibit tumor growth,while curtailing common shortcomings from conventional cancer treatment,leading towards enhanced survival rates.Particularly,organelle-targeted strategies enable precise accumulation of therapeutic agents in organelles,locally triggering organelle-mediated cell death signals which can greatly reduce the therapeutic threshold dosage and minimize side-effects.In this review,we comprehensively discuss history and recent advances in targeted therapies on organelles,specifically including nucleus,mitochondria,lysosomes and endoplasmic reticulum,while focusing on organelle structures,organelle-mediated cell death signal pathways,and design guidelines of organelle-targeted nanomedicines based on intervention mechanisms.Furthermore,a perspective on future research and clinical opportunities and potential challenges in precision oncology is presented.Through demonstrating recent developments in organelle-targeted therapies,we believe this article can further stimulate broader interests in multidisciplinary research and technology development for enabling advanced organelle-targeted nanomedicines and their corresponding clinic translations.

Comparison of DUS testing and SNP fingerprinting for variety identification in cucumber

Variety identification plays an important role in protecting the intellectual property of varieties,ensuring seed quality,and encouraging breeding innovation.Currently,morphological evaluation in the field,such as distinctness,uniformity,and stability(DUS)testing,and DNA fingerprinting in the laboratory using molecular markers are two dominant methods used for variety identification.Few studies have compared the results of these approaches,and the relationship between the two methods is obscure.In this study,134 dominant cucumber varieties were evaluated using 50 DUS testing traits and genotyped by 40 single nucleotide polymorphisms(SNPs).The 40 SNPs were developed in our previous study and arewell suited for variety identification.In the DUS testing,significant positive or negative correlations among 50 DUS traits were observed,and 20 core traits,including 15 fruit traits,were further selected to increase field inspection efficiency.This suggested that fruit shape plays an important role in variety identification.The ratio of fruit length/diameter was themost important trait,explaining 9.2%of the phenotypic variation.In the DNA fingerprinting test,the 40 SNPs were highly polymorphic and could distinguish all of the 134 cucumber varieties,and 14 core SNPs were selected to improve the identification rate.Interestingly,the population structure analysis of 134 cucumber varieties by phenotypic data in the DUS test was in accordance with the genotypic data from the DNA fingerprinting,indicating that all varieties could be divided into the same four subgroups:European type,North China type,South China type,and hybrids of the North China and South China types.Moreover,linear correlativity of distinguishment for each pair of varieties was observed between the DUS test and the DNA fingerprinting.These results indicated that these two methods have good application in future research,especially for the scaled-up analysis of hundreds of varieties.
