Proteomics analysis of coronary atherosclerotic heart disease with different Traditional Chinese Medicine syndrome types before and after percutaneous coronary intervention

OBJECTIVE: To investigate the underlying protein molecular mechanisms of "Qi stagnation and blood stasis syndrome"(QS) and "Qi deficiency and blood stasis syndrome"(QD), as two subtypes of coronary artery disease(CAD) in Traditional Chinese Medicine(TCM),following percutaneous coronary intervention(PCI).METHODS: In this study, a total of 227 CAD patients with QS and 211 CAD patients with QD were enrolled; all participants underwent PCI. Label-free quantification proteomics were employed to analyze the changes in serum in two subtypes of CAD patients before and 6 months after PCI, aiming to elucidate the intervention mechanism of PCI in treating CAD characterized by two different TCM syndromes.RESULTS: Biochemical analysis revealed significant changes in tumor necrosis factor-α, high density lipoprotein cholesterol, blood stasis clinical symptoms observation, and Gensini levels in both patient groups post-PCI; Proteomic analysis identified 79 and 95 differentially expressed proteins in the QS and QD patient groups, respectively, compared to their control groups.complement C8 alpha chain, complement factor H,apolipoprotein H, apolipoprotein B, plasminogen,carbonic anhydrase 2, and complement factor Ⅰ were altered in both comparison groups. Furthermore,enrichment analysis demonstrated that cell adhesion and connectivity-related processes underwent changes in QS patients post-PCI, whereas lipid metabolism-related pathways, including the peroxisome proliferator-activated receptor signaling pathway and extracellular matrix receptor interaction, underwent changes in the QD group.The protein-protein interaction network analysis further enriched 52 node proteins, including apolipoprotein B,lipoprotein(a), complement C5, apolipoprotein A4,complement C8 alpha chain, complement C8 beta chain,complement C8 gamma chain, apolipoprotein H,apolipoprotein A-Ⅱ, albumin, complement C4-B,apolipoprotein C3, among others. Rickettsia felis is an exclusively cytozoic Gram-negative prokaryote with cat fleas as the major vectors.[1]As early as 1918,Rickettsia felis was detected in cat fleas in Europe and named Rickettsia ctenocephali.[2]Symptoms of fever,malaise,headache,maculopapular rash and eschar are observed in patients with Rickettsia felis infection.

Objective: Despite cardiotoxicity overlap, the trastuzumab/pertuzumab and anthracycline combination remains crucial due to significant benefits. Pegylated liposomal doxorubicin(PLD), a less cardiotoxic anthracycline, was evaluated for efficacy and cardiac safety when combined with cyclophosphamide and followed by taxanes with trastuzumab/pertuzumab in human epidermal growth factor receptor-2(HER2)-positive early breast cancer(BC).Methods: In this multicenter, phase II study, patients with confirmed HER2-positive early BC received four cycles of PLD(30-35 mg/m^(2)) and cyclophosphamide(600 mg/m^(2)), followed by four cycles of taxanes(docetaxel,90-100 mg/m^(2) or nab-paclitaxel, 260 mg/m^(2)), concomitant with eight cycles of trastuzumab(8 mg/kg loading dose,then 6 mg/kg) and pertuzumab(840 mg loading dose, then 420 mg) every 3 weeks. The primary endpoint was total pathological complete response(tp CR, yp T0/is yp N0). Secondary endpoints included breast p CR(bp CR),objective response rate(ORR), disease control rate, rate of breast-conserving surgery(BCS), and safety(with a focus on cardiotoxicity).Results: Between May 27, 2020 and May 11, 2022, 78 patients were treated with surgery, 42(53.8%) of whom had BCS. After neoadjuvant therapy, 47 [60.3%, 95% confidence interval(95% CI), 48.5%-71.2%] patients achieved tp CR, and 49(62.8%) achieved bp CR. ORRs were 76.9%(95% CI, 66.0%-85.7%) and 93.6%(95% CI,85.7%-97.9%) after 4-cycle and 8-cycle neoadjuvant therapy, respectively. Nine(11.5%) patients experienced asymptomatic left ventricular ejection fraction(LVEF) reductions of ≥10% from baseline, all with a minimum value of >55%. No treatment-related abnormal cardiac function changes were observed in mean N-terminal pro-BNP(NT-pro BNP), troponin I, or high-sensitivity troponin.Conclusions: This dual HER2-blockade with sequential polychemotherapy showed promising activity with rapid tumor regression in HER2-positive BC. Importantly, this regimen showed an acceptable safety profile,especially a low risk of cardiac events,

The inconsistency of documentation:a study of online C standard library documents

The C standard libraries are basic function libraries standardized by the C language.Programmers usually refer to their APl documentation provided by third-party websites.Unfortunately,these documents are not necessarily complete or accurate,especially for constraint sentences of APl usage,which are called Security Specifications(SSs).SS issues can prevent programmers from following obligatory constraints,which results in APl misuse vulnerabilities.Previous work studying SS issues could only find certain types of inaccurate SSs through checking the compliance between APl usage and existing SSs.Therefore,we propose a novel approach SSeeker for quickly discovering missing and inaccurate SSs through the inconsistency of semantically similar SSs.More specifically,SSeeker first completes broken sentences and discovers SSs from them by judging their constraint sentiment.Then SSeeker puts semantically similar SSs from different sources into a group,which can be used to discover missing or inaccurate SSs.With the help of SSeeker,we investigated 4 popular online third-party C standard library documents,studied their conformity with the C99 standard,analyzed their APls and SSs,and discovered 92 prototype issues,15 web page issues,and 96 SS issues.

Photon-lattice(phonon)coupling is fundamental to light-matter interaction,particularly when it reaches the quantum limit of the phonon-coupled single-photon emission,which holds great potential for quantum manipulation and quantum information transduction.Here,we report single defect state-phonon coupling in hexagonal boron nitride(h BN)at room temperature.An ultrabroad spectrum of single-photon emissions can be achieved by selecting the excitation energies.Using photoluminescence excitation spectroscopy,we observe single-phonon-assisted resonance-enhanced single-photon emission,along with multiple phonon replicas that herald the creation of phonon Fock state.We also develop a transition model to gain insight into the physical process behind the single defect state-phonon coupling.Our work sets the stage for manipulating electron-phonon coupling state with single quantum-level precision at room temperature.

High temperature vulcanised silicone rubber(SR)has excellent hydrophobicity and thus is widely applied both in indoor and outdoor insulation.According to the analysis of different experimental ageing tests and field investigation of SR,temperature and humidity are considered to be the fundamental factors during degradation.Hygrothermal ageing was proposed in this study to simulate the chalking state of the SR,finally leading to the irreversible degradation far below the thermal decomposition temperature.The variation of morphology,hardness and polydimethylsiloxane(PDMS)content was quantified during the yearlong hygrothermal ageing test.It is implied that the gradual process of whitening and hardening are accompanied by the decline of the PDMS content.Finally,ageing delamination in SR and its degradation mechanism were proposed.

文明作为一种历史文化单位,其基本性质是“复杂的社会组织”。对文明及文明之间关系的研究,可从本体、比较、互动、互鉴等多个维度进行。文明自身具有一整套的核心价值观念和行为体系,可称之为文明的本体,它是文明的灵魂和认同标志。正因为文明各具特色,文明模式和发展路径多元,才应对文明进行比较,包括整体性的宏观比较和专题性的微观比较。文明间互动是各种主客观条件下的自发行为。共时性文明双方介入互动的力度以及互动对双方的影响力强弱,均有程度差异;而先在文明对后来文明的影响是单向度的。互动多是正向促进社会发展,但也有强制性互动造成破坏或缺乏互动造成发展迟滞的反例。文明互鉴则是有意识的、主动性强的高级互动。互鉴行为的发出是精神层面认识的产物,互鉴内容多在物质和制度层面,不顾自身条件的盲目模仿则难以取得成功。

Jeu de mots paronomasia:a StackOverflow‑driven bug discovery approach

Locating bug code snippets(short for BugCode)has been a complex problem throughout the history of software security,mainly because the constraints that define BugCode are obscure and hard to summarize.Previously,security analysts attempted to define such constraints manually(e.g.,limiting buffer size to detect overflow),but were limited to the types of BugCode.Recent researchers address this problem by extracting constraints from program documentation,which shows the potential for API misuse.But for bugs beyond the scope of API misuse,such an approach becomes less effective since the corresponding constraints are not defined in documents,not to mention the programs without documentation In this paper,inspired by the fact that expert programmers often correct the BugCode on open forums such as StackOverflow,we design an approach to automatically extract knowledge from StackOverflow and leverage it to detect BugCode.As we all know,the contexts in StackOverflow come from ordinary developers.Their writing tends to be loosely organized and in various styles,which are more challenging to analyze than program documentation.To address the challenges,we design a custom tokenization approach to segment sentences and employ sentiment analysis to find the Controversial Sentences(CSs)that typically contain the constraints we need for code analysis.Then we use constituency parsing to extract knowledge from CSs,which helps locate Bug-Code.We evaluated our system on 41,144 comments from the questions tagged with Java and Android.The results show that our approach achieves 95.5%precision in discovering CSs.We have discovered 276 pieces of BugCode proved to be true through manual validation including an assigned CVE.89.3%of the discovered bugs remained in the current version of answers,which are unknown to users.

以热空气吹掠型太阳能蒸馏器为研究对象,利用数值模拟结果和均匀设计法分析蒸馏器内流动与传热场协同角的变化规律和影响因素。结果表明,影响场协同角的主要因素是热空气进口速度,热空气进口温度和水温的影响不显著。在蒸馏水温30~70℃、进口空气温度40~90℃,进口空气速度为8 m/s时平均传热场协同角最小,为73.14°,传热系数最大,可达到同等条件下自然对流传热系数的4倍。

Targeting FAPα-positive lymph node metastatic tumor cells suppresses colorectal cancer metastasis

Lymphatic metastasis is the main metastatic route for colorectal cancer, which increases the risk of cancer recurrence and distant metastasis. The properties of the lymph node metastatic colorectal cancer(LNM-CRC) cells are poorly understood, and effective therapies are still lacking. Here, we found that hypoxia-induced fibroblast activation protein alpha(FAPα) expression in LNM-CRC cells. Gain-or loss-function experiments demonstrated that FAPα enhanced tumor cell migration, invasion, epithelial-mesenchymal transition, stemness, and lymphangiogenesis via activation of the STAT3 pathway. In addition, FAPα in tumor cells induced extracellular matrix remodeling and established an immunosuppressive environment via recruiting regulatory T cells, to promote colorectal cancer lymph node metastasis(CRCLNM). Z-GP-DAVLBH, a FAPα-activated prodrug, inhibited CRCLNM by targeting FAPα-positive LNM-CRC cells. Our study highlights the role of FAPα in tumor cells in CRCLNM and provides a potential therapeutic target and promising strategy for CRCLNM.
