Dissecting the cell microenvironment of ovarian endometrioma through single-cell RNA sequencing

Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

Insufficient MIRO1 contributes to declined oocyte quality during reproductive aging

Mitochondrial Rho-GTPase 1 (MIRO1) is an outer mitochondrial membrane protein which regulates mitochondrial transport and mitophagy in mitosis. In present study, we reported the crucial roles of MIRO1 in mammalian oocyte meiosis and its potential relationship with aging. We found that MIRO1 expressed in mouse and porcine oocytes, and its expression decreased in aged mice. MIRO1 deficiency caused the failure of meiotic resumption and polar body extrusion in both mouse and porcine oocytes, which could be rescued by exogenous MIRO1supplementation. Mass spectrometry data indicated that MIRO1 associated with several cytoskeleton and cell cycle-related proteins, and MIRO1 regulated motor protein Dynein for microtubule-organizing centers (MTOCs) dynamics at germinal vesicle (GV) stage, which determined meiotic resumption. Furthermore, we found that MIRO1 regulated Aurora A and kinesin family member 11 (KIF11) for meiotic spindle assembly in oocytes. Besides, MIRO1 associated with several mitochondria-related proteins dynamic-related protein 1 (DRP1),Parkin and lysosomal-associated membrane protein 2 (LAMP2) for mitochondrial dynamics and mitophagy during oocyte meiosis. Taken together, our results suggested that MIRO1 played pivotal roles in meiotic resumption, spindle assembly and mitochondrial function in mouse and porcine oocytes, and its insufficiency might contribute to the oocyte maturation defects during aging.

Atherosclerosis(AS) is a prevalent condition associated with cardiovascular and cerebrovascular diseases, featuring lipid-laden plaque; however, the insufficient resolution and low sensitivity of traditional medical imaging techniques limit their application in lipid imaging. Currently, the second near-infrared(NIR-II) fluorescence imaging offers promising sensitivity and convenience. However, existing probes have limitations in terms of signal-to-background ratio, penetration depth, and tissue scattering. Herein, we use a molecular regulatory strategy to design a lipid-activatable probe Rh-965 with high specificity and NIR-II emission for improved imaging potential in deep tissues. The potential of Rh-965 for imaging plaque lipids in mice with AS is demonstrated by localizing carotid artery plaques using NIR-II imaging. The development of more stable lipid-activated NIR-II probes can significantly enhance atherosclerosis diagnosis and facilitate in vivo imaging in deeper tissues.

The risk of flammability is an unavoidable issue for gel polymer electrolytes(GPEs).Usually,flameretardant solvents are necessary to be used,but most of them would react with anode/cathode easily and cause serious interfacial instability,which is a big challenge for design and application of nonflammable ***,a nonflammable GPE(SGPE)is developed by in situ polymerizing trifluoroethyl methacrylate(TFMA)monomers with flame-retardant triethyl phosphate(TEP)solvents and LiTFSI–LiDFOB dual lithium *** is strongly anchored to PTFMA matrix via polarity interaction between-P=O and-CH_(2)CF_(3).It reduces free TEP molecules,which obviously mitigates interfacial reactions,and enhances flame-retardant performance of TEP *** TEP molecules are also inhibited in solvation of Li^(+),leading to anion-dominated solvation sheath,which creates inorganic-rich solid electrolyte interface/cathode electrolyte interface *** coordination structure changes Li^(+)transport from sluggish vehicular to fast structural transport,raising ionic conductivity to 1.03 mS cm^(-1) and transfer number to 0.41 at 30℃.The Li|SGPE|Li cell presents highly reversible Li stripping/plating performance for over 1000 h at 0.1 mA cm^(−2),and 4.2 V LiCoO_(2)|SGPE|Li battery delivers high average specific capacity>120 mAh g^(−1) over 200 *** study paves a new way to make nonflammable GPE that is compatible with Li metal anode.

构建含细胞程序性死亡因子配体1(PD-L1)基因的慢病毒载体三质粒体系,并检测其在乳腺癌MDA-MB-231细胞中的表达情况。p MAGic 7.1慢病毒表达载体与质粒△8.91和p VSVG用聚乙烯亚胺(PEI)共转293T包装病毒,感染并筛选获得MDA-MB-231稳转细胞系,检测PD-L1表达情况。(1)MDA-MB-231在3种乳腺癌细胞中PD-L1表达量最高;(2)三质粒转染293T细胞48 h后荧光强度增强,达到90%以上;用病毒感染MDA-MB-231细胞,48 h获得20%左右荧光强度;(3)经筛选的MDA-MB-231稳转细胞系在m RNA水平和蛋白水平低表达PD-L1,其中由p MAGic-sh1包装的病毒干扰能力最强;(4)分别加入病毒后,干扰组细胞增殖能力下降,p MAGic-sh1干扰组细胞在加入病毒7 d后生长速度约为阴性对照的64.77%;(5)混合淋巴实验显示,sh-1干扰组的肿瘤抑制率为35.8%,是正常组的3.06倍,且干扰组的T淋巴细胞活性显著增强。成功通过PEI转染试剂建立了三质粒慢病毒包装体系,该体系获得的慢病毒可有效干扰乳腺癌MDA-MB-231细胞中PD-L1的表达量,所得的干扰细胞株细胞增殖能力下降,可增强T细胞增殖活性及肿瘤细胞杀伤活性。

钠-葡萄糖共转运蛋白(sodium-glucose cotransporters,SGLTs)是一类在小肠黏膜和肾近曲小管中发现的葡萄糖转运蛋白家族,其功能是介导肾脏和肠道中葡萄糖的跨膜转运.近几年研究表明,SGLT2是控制葡萄糖在肾脏重吸收的重要转运子,其抑制剂不仅可以增加尿糖的排泄从而控制糖尿病患者血糖,而且可以降低血压进而起到保护心血管的作用.而SGLT1在SGLT2抑制后,可以代偿肾脏对葡萄糖的重吸收而削弱SGLT2抑制剂的降糖疗效,同时SGLT1也是肠道吸收葡萄糖的主要转运子,因此SGLT2抑制剂对糖尿病也有治疗前景,另外,SGLT1的表达受到胃肠激素如胰高血糖素样肽-1的调控,并与味觉受体相互作用,从而对人的摄食行为和食欲产生影响,参与肥胖的发病机制.本文主要综述了SGLTs的生理功能,及其与味觉受体、肠道激素相互作用,并对其作为糖尿病治疗的新靶点进行展望.

无人机在各行各业中发挥重要作用,多无人机之间的合作已成为研究热点。针对任务分配与路径规划两个核心问题,整理和分析了多无人机协同规划两个关键问题之间的复杂性,以及子问题间的信息耦合因素,并重点探讨了解耦策略;从数学模型方面对多无人机协同规划问题的通用模型进行描述,整理归纳出常见环境建模方法和多目标优化求解的约束条件;综述了基于集中式控制和分布式控制的任务规划方法,以及启发式算法在多无人机协同规划中的应用和研究进展,并重点介绍了多无人机协同规划问题面临实时性要求下的协同规划方法;结合典型研究,讨论了多无人机协同规划问题的未来研究方法与挑战,展望多无人机协同规划的发展。

Dear Editor,This letter studies output consensus problem of heterogeneous linear multiagent systems over directed graphs. A novel adaptive dynamic event-triggered controller is presented based only on the feedback combination of the agent's own state and neighbors' output,which can achieve exponential output consensus through intermittent communication. The controller is obtained by solving two linear matrix equations, and Zeno behavior is excluded.

Al-NaOH-Composited Liquid Metal:A Fast-Response Water-Triggered Material with Thermal and Pneumatic Properties

Water-triggered materials are receiving increasing attentions due to their diverse capabilities such as easy operation,soft actuation,low cost,environmental friendliness,and many more other ***,most of such materials generally have a long reaction time and require strict preservation conditions,which limit their adaptability in *** this study,a novel water-triggered material based on Al-NaOH-composited eutectic gallium–indium(eGaIn)alloys was proposed and demonstrated,which is rather fast-responsive and *** water is applied,the material thus fabricated would achieve a temperature rise of 40C in just several seconds along with gas production,indicating its big potential to be used as a thermal and pneumatic ***,the new material’s reusability and degradation ability were also *** that,a double-layer-structure smart bandage was designed,whose bulk was filled with Al-NaOH-composited eGaIn while BiInSn served as outer supporting *** to the experiments,a sheet structure with a thickness of 2 mm would support a weight of 1.8 kg after it was subjected to a cooling process,which is much better than the weight-bearing capability of *** addition,a prototype of a water-triggered sphere robot was also fabricated using Al-NaOH-eGaIn,which realized rolling and bouncing behaviors under specific external *** findings indicate the potential value of the present material in developing future wearable devices,soft actuators,and soft robotics.
