T cell immune response is correlated with fibrosis and inflammatory activity in hepatitis B cirrhotics

AIM： TO explore the relationship among interferon-γ （IFN-γ） activity, fibrogenesis, T cell immune responses and hepatic inflammatory activity. METHODS： Peripheral blood samples from a total of 43 hepatitis B cirrhotic patients （LC） and 19 healthy controls （NC） were collected to measure their serum levels of IFN-γ, interleukin-2 （IL-2）, soluble interleukin-2 receptor （sIL-2R）, interleukin-10 （IL-10） and three serological markers of fibrosis including hyaluronic acid （HA）, procollagen type III peptide （PIIIP）, and type iV collagen were measured using a double antibody sandwich ELISA. Also, serum total bilirubin （TB） and alanine aminotransferase （ALT） were measured by routine measures. RESULTS： The concentrations of serological markers of fibrosis in patients with active cirrhosis （ALC） were significantly higher than those in stationary liver cirrhosis （SLC） or NC groups. The levels of serological markers in HBeAg-positive patients were significantly higher than those in HBeAg-negative patients. In SLC and ALC patients, a negative linear correlation was found between IFN-γ levels and the serological markers of fibrosis. IFN-γ and IL-2 levels in the ALC group were significantly higher than those in the SLC and NC groups, but the statistical difference was not significant between the latter two. In contrast, IL-10 levels in the SLC group were significantly higher than that in the NC group, but no significant difference was found between SLC and ALC groups. The sIL-2R level was elevated gradually in all these groups, and the differences were significant. Positive linear correlations were seen between IFN-γ activity and ALT levels （r = 0.339, P 〈 0.05）, and IL-2 activity and TB levels （r = 0.517, P 〈 0.05）. sIL-2R expression was positively correlated with both ALT and TB levels （r = 0.324, 0.455, P 〈 0.05）, whereas there was no statistically significant correlation between IL-10 expression and serum ALT and TB levels （r = -0.102, -0.093, P 〉 0.05）. Folate levels in mucosal tissue but not methylenetetrahydrofolate reductase polymorphisms are associated with gastric carcinogenesis

AIM: To evaluate whether folate levels in mucosal tissue and some common methylenetetrahydrofolate reductase (MTHFR) variants are associated with the risk of gastric cancer through DNA methylation. METHODS: Real-time PCR was used to study the expression of tumor related genes in 76 mucosal tissue samples from 38 patients with gastric cancer. Samples from the gastroscopic biopsy tissues of 34 patients with chronic superficial gastritis (CSG) were used as controls. Folate concentrations in these tissues were detected by the FOL ACS: 180 automated chemiluminescence system. MTHFR polymorphisms were analyzed by PCR-RFLP, and the promoter methylation of tumor-related genes was determined by methylation-specific PCR (MSP). RESULTS: Folate concentrations were significantly higher in CSG than in cancerous tissues. Decreased expression and methylation of c-myc accompanied higher folate concentrations. Promoter hypermethylation and loss of p16INK4A in samples with MTHFR 677CC were more frequent than in samples with the 677TT or 677CT genotype. And the promoter hypermethylation and loss of p21WAF1 in samples with MTHFR 677CT were more frequent than when 677CC or 677TT was present. The 677CT genotype showed a non-significant higher risk for gastric cancer as compared with the 677CC genotype. CONCLUSION: Lower folate levels in gastric mucosal tissue may confer a higher risk of gastric carcinogenesisthrough hypomethylation and overexpression of c-myc.

Effects of Cyclic Electron Flow Inhibitor(Antimycin A)on Photosystem Photoinhibition of Sweet Pepper Leaves upon Exposure to Chilling Stress Under Low Irradiance

In chloroplast, there were two pathways involved in the cyclic electron flow around photosystem 1 （PS 1）. One was the NADH dehydrogenase （NDH）-dependent flow and the other was the ferredoxin quinone reductase-dependent flow. It was proposed that the NDH-dependent cyclic electron flow around PSI was related to the xanthophyll cycle-dependent non-photochemical quenching （NPQ） at chilling temperature under low irradiance （CL）. The function of the chloroplastic cyclic electron flow around PS 1 was examined by comparing sweet pepper （Capsicum annuum L.） control with its antimycin A （AA）-fed leaves upon exposure to CL stress. During CL stress, the maximum photochemical efficiency of PS2 （Fv/Fm） decreased markedly in both controls and AA-fed leaves, and P700＋ was also lower in AA-fed leaves than in controls. These results implied that cyclic electron flow around PS 1 functioned to protect the photosynthetic apparatus from CL stress. Under such stress, NPQ and PS2-driven electron transport rate were different between AA-fed leaves and controls. The lower NPQ in AA-fed leaves might be related to an inefficient proton gradient across thylakoid membranes （ApH） because of inhibiting cyclic electron flow around PS 1 under CL stress.

Expression of Endogenous Beta Retroviruses and Hyal-2 mRNA in Immune Organs of Fetuses and Lambs

Endogenous beta retroviruses (enJSRV) are highly homologous with Jaagsiekte sheep retrovirus (exJSRV),this exogenous retrovirus is the aetiological agent of ovine pulmonary adenocarcinoma (OPA). The aim of this study was to clarify the function of enJSRV and the immunological mechanisms of its corresponding antibody, that is undetectable in JSRV-infected ovine serum. The expression of enJSRV envelope protein and Hyal-2 mRNA in immune organs and lungs of ovine fetuses and lambs were analyzed by Real-Time reverse transcription PCR and In Situ Hybridization using specific probes. In Situ Hybridization results indicated that the enJSRV envelope protein and Hyal-2 mRNA were expressed in thymus, spleen, mesenteric lymph nodes and lungs at different times, while no positive signals were detected in the negative controls. On the other hand, results from Real-Time reverse transcription PCR analysis showed that in 130d fetuses and 3d newborn lambs the enJSRV mRNA levels were much higher in organs associated with the immune system than that in lungs, especially in the thymus and spleen, but levels of Hyal-2 mRNA expression was not significantly different in all collected tissue. These results provided evidence from an immunology point of view to understand why the circulating antibodies against exJSRV are undetectable in JSRV-infected ovine, and will help to unravel the pathogenesis of JSRV-infected ovine.

Quantitative Analysis of Seeing with Height and Time at Muztagh-Ata Site Based on ERA5 Database

Seeing is an important index to evaluate the quality of an astronomical *** estimate seeing at the Muztagh-Ata site with height and time quantitatively,the European Centre for Medium-Range Weather Forecasts reanalysis database(ERA5)is *** calculated from ERA5 is compared consistently with the Differential Image Motion Monitor seeing at the height of 12 *** show that seeing decays exponentially with height at the Muztagh-Ata *** decays the fastest in fall in 2021 and most slowly with height in *** seeing condition is better in fall than in *** median value of seeing at 12 m is 0.89 arcsec,the maximum value is1.21 arcsec in August and the minimum is 0.66 arcsec in *** median value of seeing at 12 m is 0.72arcsec in the nighttime and 1.08 arcsec in the *** is a combination of annual and about biannual variations with the same phase as temperature and wind speed indicating that seeing variation with time is influenced by temperature and wind *** Richardson number Ri is used to analyze the atmospheric stability and the variations of seeing are consistent with Ri between *** quantitative results can provide an important reference for a telescopic observation strategy.

NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Comprehensive Evaluation Method for Traffic Flow Data Quality Based on Grey Correlation Analysis and Particle Swarm Optimization

Nowadays,data are more and more used for intelligent modeling and prediction,and the comprehensive evaluation of data quality is getting more and more attention as a necessary means to measure whether the data are usable or ***,the comprehensive evaluation method of data quality mostly contains the subjective factors of the evaluator,so how to comprehensively and objectively evaluate the data has become a bottleneck that needs to be solved in the research of comprehensive evaluation *** order to evaluate the data more comprehensively,objectively and differentially,a novel comprehensive evaluation method based on particle swarm optimization(PSO)and grey correlation analysis(GCA)is presented in this *** first,an improved GCA evaluation model based on the technique for order preference by similarity to an ideal solution(TOPSIS)is ***,an objective function model of maximum difference of the comprehensive evaluation values is built,and the PSO algorithm is used to optimize the weights of the improved GCA evaluation model based on the objective function ***,the performance of the proposed method is investigated through parameter analysis.A performance comparison of traffic flow data is carried out,and the simulation results show that the maximum average difference between the evaluation results and its mean value(MDR)of the proposed comprehensive evaluation method is 33.24%higher than that of TOPSIS-GCA,and 6.86%higher than that of *** proposed method has better differentiation than other methods,which means that it objectively and comprehensively evaluates the data from both the relevance and differentiation of the data,and the results more effectively reflect the differences in data quality,which will provide more effective data support for intelligent modeling,prediction and other applications.

Considering the serious electromagnetic wave(EMW)pollution problems and complex application condition,there is a pressing need to amalgamate multiple functionalities within a single ***,the effective integration of diverse functions into designed EMW absorption materials still faces the huge ***,reduced graphene oxide/carbon foams(RGO/CFs)with two-dimensional/three-dimensional(2D/3D)van der Waals(vdWs)heterostructures were meticulously engineered and synthesized utilizing an efficient methodology involving freeze-drying,immersing absorption,secondary freeze-drying,followed by carbonization *** to their excellent linkage effect of amplified dielectric loss and optimized impedance matching,the designed 2D/3D RGO/CFs vdWs heterostructures demonstrated commendable EMW absorption performances,achieving a broad absorption bandwidth of 6.2 GHz and a reflection loss of-50.58 dB with the low matching ***,the obtained 2D/3D RGO/CFs vdWs heterostructures also displayed the significant radar stealth properties,good corrosion resistance performances as well as outstanding thermal insulation capabilities,displaying the great potential in complex and variable ***,this work not only demonstrated a straightforward method for fabricating 2D/3D vdWs heterostructures,but also outlined a powerful mixeddimensional assembly strategy for engineering multifunctional foams for electromagnetic protection,aerospace and other complex conditions.
