Dicer Deletion in Astrocytes Inhibits Oligodendroglial Differentiation and Myelination

Increasing evidence has shown that astrocytes are implicated in regulating oligodendrocyte myelination,but the underlying mechanisms remain largely *** understand whether microRNAs in astrocytes function in regulating oligodendroglial differentiation and myelination in the developing and adult CNS,we generated inducible astrocyte-specific Dicer conditional knockout mice(hGFAP-CreERT;Dicer fl/fl).By using a reporter mouse line(mT/mG),we confirmed that hGFAP-CreERT drives an efficient and astrocyte-specific recombination in the developing CNS,upon tamoxifen treatment from postnatal day 3(P3)to *** Dicer deletion in astrocytes resulted in inhibited oligodendroglial differentiation and myelination in the developing CNS of Dicer cKO mice at P10 and P14,and did not alter the densities of neurons or axons,indicating that Dicer in astrocytes is required for oligodendrocyte ***,the Dicer deletion in astrocytes at P3 resulted in impaired spatial memory and motor coordination at the age of 9 *** understand whether Dicer in astrocytes is also required for remyelination,we induced Dicer deletion in 3-month-old mice and then injected lysolecithin into the corpus callosum to induce *** Dicer deletion in astrocytes blocked remyelination in the corpus callosum 14 days after induced ***,our results indicate that Dicer in astrocytes is required for oligodendroglia myelination in both the developing and adult CNS.

Exposure to chronic hypoxia is considered to be a risk factor for deficits in brain function in adults,but the underlying mechanisms remain largely *** active myelinogenesis persists in the adult central nervous system,here we aimed to investigate the impact of chronic hypoxia on myelination and the related functional consequences in adult *** a transgenic approach to label newly-generated myelin sheaths(NG2-CreER^(TM);Tau-mGFP),we found that myelinogenesis was highly active in most brain regions,such as the motor cortex and corpus *** exposure to hypoxia(10%oxygen)12 h per day for 4 weeks,myelinogenesis was largely inhibited in the 4-month old brain and the mice displayed motor coordination deficits revealed by the beam-walking *** determine the relationship between the inhibited myelination and functional impairment,we induced oligoden-droglia-specific deletion of the transcription factor 01ig2 by tamoxifen(NG2-CreER^(TM);Tau-mGFP;Olig2 fl/fl)in adult mice to mimic the decreased myelinogenesis caused by *** deletion of OHg2 inhibited myelinogenesis and consequently impaired motor coordination,suggesting that myelinogenesis is required for motor function in adult *** understand whether enhancing myelination could protect brain functions against hypoxia,we treated hypoxic mice with the myelination-enhancing drug-clemastine,which resulted in enhanced myelogenesis and improved motor *** together,our data indicate that chronic hypoxia inhibits myelinogenesis and causes functional deficits in the brain and that enhancing myelinogenesis protects brain functions against hypoxia-related deficits.

Co-production of hydrogen, oxygen, and electricity via an integrated solar-driven system with decoupled water electrolyzer and Na-Zn ion battery

Combining water electrolysis and rechargeable battery technologies into a single system holds great promise for the co-production of hydrogen (H_(2)) and ***,the design and development of such systems is still in its ***,an integrated hydrogen-oxygen (O_(2))-electricity co-production system featuring a bipolar membrane-assisted decoupled electrolyzer and a Na-Zn ion battery was established with sodium nickelhexacyanoferrate (NaNiHCF) and Zn^(2+)/Zn as dual redox *** decoupled electrolyzer enables to produce H_(2)and O_(2)in different time and space with almost 100%Faradaic efficiency at 100 mA cm^(-2).Then,the charged NaNiHCF and Zn electrodes after the electrolysis processes formed a Na-Zn ion battery,which can generate electricity with an average cell voltage of 1.75 V at 10 m A cm^(-2).By connecting Si photovoltaics with the modular electrochemical device,a well-matched solar driven system was built to convert the intermittent solar energy into hydrogen and electric energy with a solar to hydrogen-electricity efficiency of 16.7%,demonstrating the flexible storage and conversion of renewables.

An efficient quantum proactive incremental learning algorithm

In scenarios where a large amount of data needs to be learned,incremental learning can make full use of old knowledge,signif-icantly reduce the computational cost of the overall learning process,and maintain high *** this paper,taking the MaxCut problem as our example,we introduce the idea of incremental learning into quantum computing,and propose a Quantum Proactive Incremental Learning algorithm(QPIL).Instead of a one-off training of quantum circuit,QPIL contains a multi-phase training on gradually-increased subgraphs of all vertices,proactively reducing large-scale problems to smaller ones to solve in steps,providing an efficient solution for ***,some vertices and corresponding edges are randomly selected for training to obtain optimized parameters of the quantum circuit at ***,in each incremental phase,the remaining vertices and corresponding edges are gradually added and the parameters obtained from the previous phase are reused in the parameter initialization of the current *** perform experiments on 120 different small-scale graphs,and it shows that QPIL performs superior to prevalent quantum and classical baselines in terms of approximation ratio(AR),time cost,anti-forgetting,and solv-ing *** particular,QPIL’s AR surpasses 20%of mainstream quantum baselines,and the time cost is less than 1/5 of *** idea of QPIL is expected to inspire efficient and high-quality solutions in large-scale MaxCut and other combinatorial optimization problems.

Global interest in lithium-sulfur batteries as one of the most promising energy storage technologies has been sparked by their low sulfur cathode cost,high gravimetric,volumetric energy densities,abundant resources,and environmental ***,their practical application is significantly impeded by several serious issues that arise at the cathode-electrolyte interface,such as interface structure degradation including the uneven deposition of Li_(2)S,unstable cathode-electrolyte interphase(CEI)layer and intermediate polysulfide shuttle ***,an optimized cathode-electrolyte interface along with optimized electrodes is required for overall ***,we comprehensively outline the challenges and corresponding strategies,including electrolyte optimization to create a dense CEI layer,regulating the Li_(2)S deposition pattern,and inhibiting the shuttle effect with regard to the solid-liquid-solid pathway,the transformation from solid-liquid-solid to solid-solid pathway,and solid-solid pathway at the cathode-electrolyte *** order to spur more perceptive research and hasten the widespread use of lithium-sulfur batteries,viewpoints on designing a stable interface with a deep comprehension are also put forth.

Highly conductive and stable iodine doped argyrodite electrolyte for all-solid-state lithium batteries

Lithium argyrodites with high ionic conductivity and low cost are considered as one of the most prospective solid electrolytes for all-solid-state lithium ***,the poor chemical stability and compatibility with lithium metal limit their ***,Li_(5.4)PS_(4.4)Cl1.4I0.2solid electrolyte with high ionic conductivity of 11.49 m S ccm^(-1)and improved chemical stability is synthesized by iodine *** ultra-thin Li_(5.4)PS_(4.4)Cl_(1.4)I_(0.2)membrane with thickness of 10μm can be obtained by wet coating process,exhibiting a high ionic conductivity of 2.09 mS ccm^(-1)and low areal resistance of 1.17Ωcm^(-2).Moreover,iodine doping could in-situ form LiI at the lithium/electrolyte interface and improve the critical current density of Li_(5.4)PS_(4.4)Cl_(1.6)from 0.8 to 1.35 mA cm^(-2).The resultant LiCoO_(2)/Li_(5.4)PS_(4.4)Cl_(1.4)I_(0.2)/Li battery shows excellent cycling stability at 1 C,with a reversible specific capacity of 110.1 mA h g^(-1)and a retention of 80.5% after 1000 *** addition,the assembled LiCoO_(2)/Li_(5.4)PS_(4.4)Cl_(1.4)I_(0.2)membrane/Li pouch cell delivers an initial discharge capacity of 110.4 mA h g^(-1)and 80.5% capacity retention after 100 cycles.

Calcium(Ca^(2+))is an essential nutrient and a cellular signal in *** level of free Ca^(2+)in the apoplast is~10,000-fold higher than that in the cytoplasm at the resting *** large Ca^(2+)level gradient across the membrane between the interior and exterior can cause Ca^(2+)overload and *** have evolved mechanisms to export Ca^(2+)out of the cytoplasm or store it in intracellular organelles,such as endoplasmic reticulum(ER),mitochondria,and ***,Ca^(2+)-binding proteins,including those containing the most common Ca^(2+)-binding EF-hand domains,maintain the cytosolic Ca^(2+)level by directly binding them,thus participating in Ca^(2+)***^(2+)/H+exchangers(CAXs,antiporter)and Ca^(2+)-ATPases(ACAs)are responsible for transferring Ca^(2+)from the cytosol to either the external space or intracellular stores(Bose et al.,2011).However,it is still unknown how these Ca^(2+)-scavenging transporters are activated when the cytosolic Ca^(2+)level is elevated to threshold ***“auto-regulatory”mechanism to scavenge excess cytosolic Ca^(2+),providing a guarding system to maintain cytosolic Ca^(2+)homeostasis in plants.

Quantum instability and Ehrenfest time for an inverted harmonic oscillator

We use out-of-time order correlators(OTOCs)to investigate the quantum instability and Ehrenfest time for an inverted harmonic oscillator(IHO).For initial states located in the stable manifolds of the IHO we find that the corresponding OTOC exhibits identical evolutionary characteristics to the saddle point before the Ehrenfest *** initial states located in the unstable manifolds,the OTOCs still grow exponentially but the time to maintain exponential growth is related to the center position of its wave packet in phase ***,we use the Husimi Q function to visualize the quantum wave packets during exponential growth of the *** results show that quantum instability exists at arbitrary orbits in the IHO system,and the Ehrenfest time in the IHO system depends not only on the photon number of the initial system but also on the central positions of the initial states in phase space.

Precise gradient separation of nanoparticles using reduced graphene oxide-polyoxomolybdate membranes

The purification and size sieving of nanoparticles(NPs)represents a significant scientific ***,the rGO-polyoxometalates(POMs=PMo_(12),Mo_(132)and Mo_(368))membranes were fabricated successfully via a simple and scalable POMsassisted in-situ photoreduction *** the mass ratio of rGO to PMo_(12)was 1:30,the resultant rGO-PMo_(12)membranes with highly ordered and uniform nanochannels of 1.0 nm can be obtained,which showed an accurate carbon quantum dots(CQDs)separation with a cut-off size approximately 1.0 *** increase of the size of the interlayered POMs from PMo_(12)(1.0 nm),{Mo132}(2.9 nm)to{Mo_(368)}(3.8 nm),the resultant rGO-PMo_(12),rGO-Mo_(132)and rGO-Mo_(368)exhibited precise gradient separation of CQDs and Au nanoparticles by size exclusion *** results pave a promising way to construct new twodimensional(2D)membranes for highly efficient size-selective separation.
