The role of axon guidance molecules in the pathogenesis of epilepsy

Current treatments for epilepsy can only manage the symptoms of the condition but cannot alter the initial onset or halt the progression of the disease. Consequently, it is crucial to identify drugs that can target novel cellular and molecular mechanisms and mechanisms of action. Increasing evidence suggests that axon guidance molecules play a role in the structural and functional modifications of neural networks and that the dysregulation of these molecules is associated with epilepsy susceptibility. In this review, we discuss the essential role of axon guidance molecules in neuronal activity in patients with epilepsy as well as the impact of these molecules on synaptic plasticity and brain tissue remodeling. Furthermore, we examine the relationship between axon guidance molecules and neuroinflammation, as well as the structural changes in specific brain regions that contribute to the development of epilepsy. Ample evidence indicates that axon guidance molecules, including semaphorins and ephrins, play a fundamental role in guiding axon growth and the establishment of synaptic connections. Deviations in their expression or function can disrupt neuronal connections, ultimately leading to epileptic seizures. The remodeling of neural networks is a significant characteristic of epilepsy, with axon guidance molecules playing a role in the dynamic reorganization of neural circuits. This, in turn, affects synapse formation and elimination. Dysregulation of these molecules can upset the delicate balance between excitation and inhibition within a neural network, thereby increasing the risk of overexcitation and the development of epilepsy. Inflammatory signals can regulate the expression and function of axon guidance molecules, thus influencing axonal growth, axon orientation, and synaptic plasticity. The dysregulation of neuroinflammation can intensify neuronal dysfunction and contribute to the occurrence of epilepsy. This review delves into the mechanisms associated with

Toward Analytical Homogenized Relaxation Modulus for Fibrous Composite Material with Reduced Order Homogenization Method

In this manuscript,we propose an analytical equivalent linear viscoelastic constitutive model for fiber-reinforced composites,bypassing general computational *** method is based on the reduced-order homogenization(ROH)*** ROH method typically involves solving multiple finite element problems under periodic conditions to evaluate elastic strain and eigenstrain influence functions in an‘off-line’stage,which offers substantial cost savings compared to direct computational homogenization *** to the unique structure of the fibrous unit cell,“off-line”stage calculation can be eliminated by influence functions obtained *** the standard solid model to the ROH method enables the creation of a comprehensive analytical homogeneous viscoelastic constitutive *** method treats fibrous composite materials as homogeneous,anisotropic viscoelastic materials,significantly reducing computational time due to its analytical *** approach also enables precise determination of a homogenized anisotropic relaxation modulus and accurate capture of various viscoelastic responses under different loading *** sets of numerical examples,including unit cell tests,three-point beam bending tests,and torsion tests,are given to demonstrate the predictive performance of the homogenized viscoelastic ***,the model is validated against experimental measurements,confirming its accuracy and reliability.

The study of the hydrodynamic limit of the Boltzmann equation with physical boundary is a challenging problem due to the appearance of the viscous and Knudsen boundary *** this paper,the hydrodynamic limit from the Boltzmann equation with the specular reflection boundary condition to the incompressible Euler equations in a channel is *** on the multi-scaled Hilbert expansion,the equations with boundary conditions and compatibility conditions for interior solutions,and viscous and Knudsen boundary layers are derived under different scaling,***,some uniform estimates for the interior solutions,and viscous and Knudsen boundary layers are *** the help of the L2-L∞framework and the uniform estimates obtained above,the solutions to the Boltzmann equation are constructed by the truncated Hilbert expansion with multiscales,and hence the hydrodynamic limit in the incompressible Euler level is justified.

Cobalamin(Cbl)-dependent radical S-adenosyl-L-methionine(SAM) proteins constitute the largest collection of the radical SAM superfamily that has hundreds of thousands of individual members. Many of these proteins are involved in the biosynthesis of pharmaceutically important natural products to catalyze chemically demanding reactions. In the biosynthetic pathway of chuangxinmycin(CXM), a unique indole alkaloid antibiotic with potent anti-infective activity, functionalization of the characteristic thiopyrano[4,3,2-cd]indole scaffold by regio-and stereoselective C3-methylation is believed to rely on a Cbl-dependent radical process, which, however, remained to be reconstituted biochemically. We here report the dissection of this enzymatic process, which requires the incorporation of Cxm8, a Cbl-dependent radical SAM protein, with Cxm9, a DUF5825family protein that shares no homology to any proteins of known functions. Cxm8 and Cxm9 function together by forming an unexpected heterodimeric complex that selectively catalyzes C3-methylation of the tricyclic indole-S-hetero ring system in a successive manner, achieving CXM and a recently identified, C3-dimethylated congener. Detailed biochemical characterization,isotope labeling, structural simulation and bioinformatics analysis rationalized the catalysis of the Cxm8/Cxm9 complex and particularly the necessity of the DUF5825 protein for C3-methylase activity. This is the first example that a Cbl-dependent protein acts with a partner to exhibit radical SAM activity.

GIS-based logistic regression method for landslide susceptibility mapping in regional scale

Landslide susceptibility map is one of the study fields portraying the spatial distribution of future slope failure sus- ceptibility. This paper deals with past methods for producing landslide susceptibility map and divides these methods into 3 types. The logistic linear regression approach is further elaborated on by crosstabs method, which is used to analyze the relationship between the categorical or binary response variable and one or more continuous or categorical or binary explanatory variables derived from samples. It is an objective assignment of coefficients serving as weights of various factors under considerations while expert opinions make great difference in heuristic approaches. Different from deterministic approach, it is very applicable to regional scale. In this study, double logistic regression is applied in the study area. The entire study area is first analyzed. The logistic regression equation showed that elevation, proximity to road, river and residential area are main factors triggering land- slide occurrence in this area. The prediction accuracy of the first landslide susceptibility map was showed to be 80%. Along the road and residential area, almost all areas are in high landslide susceptibility zone. Some non-landslide areas are incorrectly divided into high and medium landslide susceptibility zone. In order to improve the status, a second logistic regression was done in high landslide susceptibility zone using landslide cells and non-landslide sample cells in this area. In the second logistic regression analysis, only engineering and geological conditions are important in these areas and are entered in the new logistic regression equation indicating that only areas with unstable engineering and geological conditions are prone to landslide during large scale engineering activity. Merging bound states in the continuum in an open acoustic resonator

Bound states in the continuum （BICs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIC is very sensitive to the structure ***,we report merging acoustic BICs in a single open resonator,robust against the structure *** find that both symmetry-protected BIC and Friedrich-Wintgen BIC are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BICs move toward each other and merge into a single one at a critical *** to an individual BIC,the merged BIC is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BICs in a coupled 3D waveguide-resonator,which are evidenced by the vanishing of the linewidth of Fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.
