Maintaining metabolic homeostasis is essential for cellular and organismal health throughout life.Multiple signaling pathways that regulate metabolism also play critical roles in aging,such as PI3K/AKT,mTOR,AMPK,and sirtuins(SIRTs).Among them,sirtuins are known as a protein family with versatile functions,such as metabolic control,epigenetic modification and lifespan extension.Therefore,by understanding how sirtuins regulate metabolic processes,we can start to understand how they slow down or accelerate biological aging from the perspectives of metabolic regulation.Here,we review the biology of SIRT3,SIRT4,and SIRT5,known as the mitochondrial sirtuins due to their localization in the mitochondrial matrix.First,we will discuss canonical pathways that regulate metabolism more broadly and how these are integrated with aging regulation.Then,we will summarize the current knowledge about functional differences between SIRT3,SIRT4,and SIRT5 in metabolic control and integration in signaling networks.Finally,we will discuss how mitochondrial sirtuins regulate processes associated with aging and aging-related diseases.

Stable NiPt-Mo_(2)C active site pairs enable boosted water splitting and direct methanol fuel cell

Sluggish kinetics of methanol oxidation reaction(MOR)and alkaline hydrogen evolution reaction(HER)even on precious Pt catalyst impede the large-scale commercialization of direct methanol fuel cell(DMFC)and water electrolysis technologies.Since both of MOR and alkaline HER are related to water dissociation reaction(WDR),it is reasonable to invite secondary active sites toward WDR to pair with Pt for boosted MOR and alkaline HER activity on Pt.Mo_(2)C and Ni species are therefore employed to engineer NiPt-Mo_(2)C active site pairs,which can be encapsulated in carbon cages,via an in-situ self-confinement strategy.Mass activity of Pt in NiPt-Mo_(2)C@C toward HER is boosted to11.3 A mg_(pt)^(-1),33 times higher than that of Pt/C.Similarly,MOR catalytic activity of Pt in NiPt-Mo_(2)C@C is also improved by 10.5 times and the DMFC maximum power density is hence improved by 9-fold.By considering the great stability,NiPt-Mo_(2)C@C exhibits great practical application potential in DMFCs and water electrolysers.

Phototheranostic Nanoagents for Imaging-Guided Treatment of Oral Cancer

Phototherapy showed almost a lack of drug resistance and,depending on the therapeutic effects of non-invasive light-stimulating photosensors.The side effects of phototherapy were greatly reduced compared to their traditional equivalents.Phototheranostic nanoagents had new properties in drug delivery,biocompatibility,targeting and response,in which traditional phototheranostic drugs can not possess.Recently,a large number of relevant studies have demonstrated that photodynamic therapy(PDT)in combination with other agents and image-guided multifunctional photothermal therapy(PTT)were well suited for the treatment of oral cancer.Through the design of the nanoagents,researchers have discovered various applications for phototherapy,such as targeted release of co-packaged drugs,multifunctional imaging for diagnosis and treatment combination,accurate targeting caused by nanocarriers,and synergistic chemotherapy with phototherapy.In this paper,we first reviewed the research related to phototheranostic nanoagents for image-guided treatment of oral cancer.We tried to introduce the design concept and the treatment effect by three parts of components of phototheranostic nanoagents,categories of phototheranostic nanoagents and application of phototheranostic nanoagents.It also provided a reference for nanomaterial development and clinical applications in research of oral cancer treatment.

Recent advances in BiOBr-based photocatalysts for environmental remediation

The efficient utilization of solar energy through photocatalysis is ideal for solving environmental issues and the development sustainable future.BiOBr-based semiconductors possess unique narrowed bandgaps and layered structures,thereby widely studied as photocatalysts for environmental remediation.However,a little has been focused on the comprehensive reviewing of BiOBr despite its extensive and promising applications.In this review,the state-of-the-art developments of BiOBr-based photocatalysts for environmental remediation are summarized.Particular focus is paid to the synthetic strategies for the control of the resulting morphologies,as well as efficient modification strategies for improving the photocatalytic activities.These include boosting the bulk phase by charge separation,enhancing the spatial charge separation,and engineering the surface states.The environmental uses of BiOBr-based photocatalysts are also reviewed in terms of purification of pollutants and CO_(2) reduction.Finally,future challenges and opportunities of BiOBr-based materials in photocatalysis are discussed.Overall,this review provides a good basis for future exploration of high-efficiency solar-driven photocatalysts for environmental sustainability.

Identification and Evaluation of Insect and Disease Resistance in Transgenic Cry1Ab13-1 and NPR1 Maize

PCR detection,quantitative real-time PCR(q-RTPCR),outdoor insect resistance,and disease resistance identification were carried out for the detection of genetic stability and disease resistance through generations(T2,T3,and T4)in transgenic maize germplasms(S3002 and 349)containing the bivalent genes(insect resistance gene Cry1Ab13-1 and disease resistance gene NPR1)and their corresponding wild type.Results indicated that the target genes Cry1Ab13-1 and NPR1 were successfully transferred into both germplasms through tested generations;q-PCR confirmed the expression of Cry1Ab13-1 and NPR1 genes in roots,stems,and leaves of tested maize plants.In addition,S3002 and 349 bivalent gene-transformed lines exhibited resistance to large leaf spots and corn borer in the field evaluation compared to the wild type.Our study confirmed that Cry1Ab13-1 and NPR1 bivalent genes enhanced the resistance against maize borer and large leaf spot disease and can stably inherit.These findings could be exploited for improving other cultivated maize varieties.

Early expression of PTX3 in Aspergillus fumigatus infected rat cornea

AIM： To investigate the expression of pentraxin 3（PTX3） in rat corneal epithelium at the early stage of Aspergillus fumigatus（A. fumigatus） infection. METHODS： A total of 50 Wistar rats were randomly divided into control group, Sham group and experimental group（fungal keratitis group, FK group）. The right eye was chosen as the experiment one and infected by A. fumigatus. Rats were executed at 8, 16 and 24 h after the experimental models being established. Corneal epithelia were collected to assess the expression of PTX3 by quantitative reverse transcription polymerase chain reaction（q RT-PCR） and Western blot analysis. RESULTS： Corneal inflammation scores increased as infection prolonged（P〈0.05, P〈0.001）. PTX3 m RNA expression was low in normal and Sham group rats＇ corneas. Level of PTX3 m RNA in infected rat cornea was elevated at 8 h and peaked at 16 h. The difference was significant compared with control group（P〈0.001）. Western blot analysis also showed a significant increase of PTX3 protein in experimental group at 8 h and peaked at 16 h（P〈0.001）. The synchronous expression of control group and experimental group were also in significant difference（P〈0.001）. CONCLUSION： PTX3 exists in cornea epithelium and is significantly increased after A. fumigatus infection. PTX3 plays an important role in the early stage of cornea innate immunity against A. fumigatus.

Aging research has shifted from studying phenotypes to studying in-depth mechanisms in recent decades.However,extrapolating cellular and molecular bases of aging from studying traditional model systems to humans has been challenging.The advent of organoids holds promise for overcoming the limitations of monolayer cell culture and bridging the gap between animal models and humans.Here,we mainly discuss recent paradigms for using organoid models in studying organ aging.Pluripotent stem cells-derived organoid provides a promising platform for simulating the pathophysiology of several aging-related diseases,especially neurodegenerative diseases,and adult stem cells organoids derived from different age groups have been applied to detect aging-related functional changes.We also assess the value of organoid model systems in understanding human aging and aging-related diseases,and identify challenges to be addressed in the future,such as the immaturity of organoids,and effective methods of inducing aging.
