Effect of Berberine on Activation of TLR4-NFκB Signaling Pathway and NLRP3 Inflammasome in Patients with Gout

Objective:To determine the effects of berberine(BBR) on the activation of toll-like receptor 4(TLR4),nuclear factor(NF) κB(NF-κB) signaling and NLRP3 inflammasome in patients with gout.Methods:Peripheral blood mononuclear cells(PBMCs) were obtained from 24 acute(AP) and 41 non-acute(NAP) phases of primary gout patients,respectively,as well as 30 healthy controls(HC).TLR4,NF-κB(p65),NLRP3,apoptosis-associated specklike protein containing a CARD(PYCARD),cysteinyl aspartate specific proteinase-1(CASP1),and interleukin-1 β(IL-1 β) mRNA expression levels in PBMCs were measured by quantitative reverse transcriptase polymerase chain reaction.The protein levels of TLR4,myeloid differentiation factor 88(MyD88),NF-κB(p50/65),inhibitor of kappa B kinase α/β(IKKα/β),NF-κB inhibitor α(IKB α),phospho-IKKα/β(p-IKKα/β),NLRP3,PYCARD,and CASP1were monitored by Western blotting.Serum IL-1 β protein level was measured using enzyme-linked immunosorbent assay(ELISA).In addition,PBMCs from HC and macrophages derived from a spontaneously immortalized monocyte-like cell line(THP-1) were stimulated using monosodium urate(MSU,100 μ g/mL),0.1% dimethyl sulfoxide,25 μmol/L BBR,and 10,25,and 50 μmol/L BBR+100 μg/mL MSU for different time periods.The protein levels of IL-1β and IL-18 in cell culture supernatants was measured by ELISA,and the protein expressions of TLR4,MyD88,NF-κB(p50/p65),IKK α/β,IκBα,p-IKKα/β,NLRP3, PYCARD,and CASP1 in macrophages were analyzed by Western blotting.Results:(1) TLR4,NF-κB(p65),PYCARD,CASP1,and IL-1β mRNA levels in PBMCs were significantly higher in the AP group than in the HC group(P<0.05).The NLRP3 mRNA expression levels in PBMCs were found to be significantly lower in the AP and NAP groups than in the HC group(P<0.05,P<0.01).(2) The protein levels of TLR4,IKKβ,MyD88,NF-κB,p-IKKα/β,PYCARD,and CASP1 in PBMCs were significantly higher,and those of IκBα,IKKα,and NLRP3 were found to be significantly lower in the AP group than in the HC group(P<0.05 or P<0.01).(3) The serum IL-1β protein levels were significantly

Cyclodextrin-functionalized Ordered Porous Block Copolymer Films: Preparation and Property

Ordered porous block copolymer films（PBCFs） have attracted much attention because of their potential applications in several fields. In this paper, we first reported a non-destructive, controllable, and efficient approach for preparation of β-cyclodextrin（β-CD）-functionalized PBCFs（β-CD-PBCFs）. The key point of the approach is to incorporate β-CD units into the hydrophilic segment of amphiphilic block copolymers before the preparation of films. β-CD-PBCFs with structural integrity and controllable pore parameters were fabricated through combining of self-assembly and breath figure methods. And then, the effective adsorption capacity of β-CD-PBCFs toward Congo red was confirmed through UV-Vis spectroscopy and was found to be affected by β-CD content and solution p H values. Adsorption kinetic results showed that the adsorption behavior of β-CD-PBCFs was consistent with the pseudo-second-order kinetic model and the chemisorption mechanism.

A preliminary study of the effects of plastic film-mulched raised beds on soil temperature and crop performance of early-sown short-season spring maize(Zea mays L.) in the North China Plain

To identify a strategy for earlier sowing and harvesting of spring maize(Zea mays L.) in an alternative maize–maize double cropping system, a 2-year field experiment was performed at Quzhou experimental station of China Agricultural University in 2014 and 2015. A short-season cultivar, Demeiya number 1(KX7349), was used in the experiment. Soil temperature to 5 cm depth in the early crop growth stage, crop growth, crop yield, and water use of different treatments(plastic film-mulched raised bed(RF) and flat field without plastic film mulching(CK) in 2014; RF, plastic film-mulched flat field(FF), and CK in 2015)were measured or calculated and compared. Soil temperature in the film-mulched treatments was consistently higher than that in CK(1.6–3.5 °C in average) during the early growth stage. Crops in plastic film-mulched treatments used 214 fewer growing-degree days(GDDs) in 2014 and 262 fewer GDDs in 2015. In 2014, the RF treatment yielded 32.7%higher biomass than CK, although its 9.4% higher grain yield was not statistically significant. Also, RF used 17.9% less water and showed 33.1% higher water use efficiency(WUE) than CK. In 2015, RF and FF showed 56.2% and 49.5% higher yield, 15.0% and 4.5%lower water use(ET), and 63.4% and 75.7% higher WUE, respectively, than CK. RF markedly increased soil temperature in the early crop season, accelerated crop growth, reduced ET,and greatly increased crop yield and WUE. Compared with FF, RF had no obvious effect on crop growth rate, although soil temperature during the period between sowing and stem elongation was slightly increased. However, RF resulted in lower ET and higher WUE than FF. Effects of RF on soil water dynamics as well as its cost-effectiveness remain topics for further study.

PerfMon: Measuring Application-Level Performance in a Large-Scale Campus Wireless Network

WiFi has become one of the most popular ways to access the Internet.However,in large-scale campus wireless networks,it is challenging for network administrators to provide optimized access quality without knowledge on fine-grained traffic characteristics and real network performance.In this paper,we implement PerfMon,a network performance measurement and diagnosis system,which integrates collected multi-source datasets and analysis methods.Based on PerfMon,we first conduct a comprehensive measurement on application-level traffic patterns and behaviors from multiple dimensions in the wireless network of T university(TWLAN),which is one of the largest campus wireless networks.Then we systematically study the application-level network performance.We observe that the application-level traffic behaviors and performance vary greatly across different locations and device types.The performance is far from satisfactory in some cases.To diagnose these problems,we distinguish locations and device types,and further locate the most crucial factors that affect the performance.The results of case studies show that the influential factors can effectively characterize performance changes and explain for performance degradation.

Expression of PYCARD Gene Transcript Variant mRNA in Peripheral Blood Mononuclear Cells of Primary Gout Patients with Different Chinese Medicine Syndromes

Objective： To study the expression level and role of apoptosis-associated speck-like protein containing a caspase recruitment domain （PYCARD） gene transcript variant mRNA in peripheral blood mononuclear cells （PBMCs） of primary gout （PG） patients with different Chinese medicine （CM） syndromes. Methods： The expressions of PYCARD gene transcript variant mRNA and interleukin-1β （IL-1β） mRNA in PBMCs were investigated in 96 PG patients with acute phase （APPG, 44 cases） and non-acute phase （NAPPG, 52 cases） and 30 healthy controls （HCs） by reverse transcription-polymerase chain reaction （PCR） and/or real-time quantitative PCR. PYCARD and nuclear factor-κB （p50） [NF-κB （p50）] protein was detected by Western blot in PBMCs respectively. IL-1β, IL-4 and IL-10 protein levels in plasma of HCs and PG patients were measured by enzyme-linked immuno sorbent assay. Results： The main CM syndromes in APPG patients were obstruction of dampness and heat syndrome （ODHS, 36.36%） and intermingled phlegm-blood stasis syndrome （IPBSS, 27.27%）, while in NAPPG patients were Pi （Spleen）-deficiency induced dampness syndrome （PDIDS, 40.38%） and qi-blood deficiency syndrome （QBDS, 26.92%）. It showed statistical significances of the expressions of PYCARD gene and its transcript variant mRNA, the protein of PYCARD and NF-κB （p50） and the plasma IL-1β, IL-4 and IL-10 in APPG, NAPPG, ODHS, IPBSS, PDIDS and QBDS groups, compared with the HC group respectively （P〈0.05 or P〈0.01）. There were also significant differences of mRNA expressions of PYCARD-1 and PYCARD-2 as well as protein expressions of IL-1β, IL-4 and IL-10 among the 4 CM syndromes groups （P〈0.05 or P〈0.01）. Correlation analysis showed positive correlation between the mRNA expressions of PYCARD-1 gene transcript variant and IL-1β in APPG patients （r=0.3088, P=0.0183）. Conclusion： PYCARD gene and its transcript variant may play a critical and regulative role in the inflammatory response of PG patients with different phases and CM syndromes.

ATP13A2 protects dopaminergic neurons in Parkinson's disease:from biology to pathology

As a late endosomal/lysosomal transport protein of the P5-type, ATP13A2 is capable of removing the abnormal accumulation of α-synuclein, which maintains the homeostasis of metal ions and polyamines in the central nervous system. Furthermore, ATP13A2 regulates the normal function of several organelles such as lysosomes, endoplasmic reticulum (ER) and mitochondria, and maintains the normal physiological activity of neural cells. Especially, ATP13A2 protects dopaminergic (DA) neurons against environmental or genetically induced Parkinson's disease (PD). As we all know, PD is a neurodegenerative disease characterized by the loss of DA neurons in the substantia nigra pars compacta. An increasing number of studies have reported that the loss-of- function of ATP13A2 affects normal physiological processes of various organelles, leading to abnormalities and the death of DA neurons. Previous studies in our laboratory have also shown that ATP13A2 deletion intensifies the neuroinflammatory response induced by astrocytes, thus inducing DA neuronal injury. In addition to elucidating the normal structure and function of ATP13A2, this review summarized the pathological mechanisms of ATP13A2 mutations leading to PD in existing literature studies, deepening the understanding of ATP13A2 in the pathological process of PD and other related neurodegenerative diseases. This review provides inspiration for investigators to explore the essential regulatory role of ATP13A2 in PD in the future.

Influence of varied drought types on soil conservation service within the framework of climate change:insights from the jinghe River Basin,China

Severe soil erosion and drought are the two main factors affecting the ecological security of the Loess Plateau,China.Investigating the influence of drought on soil conservation service is of great importance to regional environmental protection and sustainable development.However,there is little research on the coupling relationship between them.In this study,focusing on the jinghe River Basin,China as a case study,we conducted a quantitative evaluation on meteorological,hydrological,and agricultural droughts(represented by the Standardized Precipitation Index(SPI),Standardized Runoff Index(SRI),and Standardized Soil Moisture Index(SSMI),respectively)using the Variable Infiltration Capacity(VIC)model,and quantified the soil conservation service using the Revised Universal Soil Loss Equation(RUSLE)in the historical period(2000-2019)and future period(2026-2060)under two Representative Concentration Pathways(RCPs)(RCP4.5 and RCP8.5).We further examined the influence of the three types of drought on soil conservation service at annual and seasonal scales.The NASA Earth Exchange Global Daily Downscaled Projections(NEX-GDDP)dataset was used to predict and model the hydrometeorological elements in the future period under the RCP4.5 and RCP8.5 scenarios.The results showed that in the historical period,annual-scale meteorological drought exhibited the highest intensity,while seasonal-scale drought was generally weakest in autumn and most severe in summer.Drought intensity of all three types of drought will increase over the next 40 years,with a greater increase under the RCP4.5 scenario than under the RCP8.5 scenario.Furthermore,the intra-annual variation in the drought intensity of the three types of drought becomes smaller under the two future scenarios relative to the historical period(2000-2019).Soil conservation service exhibits a distribution pattern characterized by high levels in the southwest and southeast and lower levels in the north,and this pattern has remained consistent both in the historical and future pe

Sentinel Surveillance of Schistosomiasis—China,2021

Introduction:This report analyzes the national surveillance data for schistosomiasis in 2021 to understand the current status and provide evidence for further policy actions to promote elimination.This analysis is in line with the National Surveillance Plan of Schistosomiasis,which was revised in 2020 to adapt to the new stage of moving towards elimination.Methods:Data from the 2021 national surveillance of schistosomiasis in humans,livestock,and snails were collected from 13 provincial-level administrative divisions(PLADs)and analyzed using descriptive epidemiological methodology.The antibody-positive rate and area of newly discovered and re-emergent snail habitats were calculated.Results:In 2021,a total of 31,661 local residents and 101,558 transient population were screened for antibodies using indirect hemagglutination assay(IHA).Of those who tested positive,745 local residents and 438 transient population underwent further parasitological examination,with only one stool-positive result in the transient population.Additionally,12,966 livestock were examined using the miracidia hatching test,with no positives detected.The total area of newly discovered and re-emergent snail habitats was 957,702 m^(2) and 4,381,617 m^(2),respectively.No infected snails were found using the microscopic dissection method,but six pooled snail samples were reported as positive using the loopmediated isothermal amplification method for detecting specific sequences of Schistosoma.japonicum,in Anhui and Jiangxi Provinces.Conclusions:The prevalence of schistosomiasis among humans and livestock was found to be low,however,a potential transmission risk was identified in certain areas.To reduce the risk of transmission,a comprehensive control strategy should be continued and new techniques should be implemented in the surveillance and early warning system.

Cardiomyocyte-targeted anti-inflammatory nanotherapeutics against myocardial ischemia reperfusion (IR) injury

Myocardial ischemia reperfusion(IR)injury is closely related to the overwhelming inflammation in the myocardium.Herein,cardiomyocyte-targeted nanotherapeutics were developed for the reactive oxygen species(ROS)-ultrasensitive co-delivery of dexamethasone(Dex)and RAGE small interfering RNA(siRAGE)to attenuate myocardial inflammation.PPTP,a ROSdegradable polycation based on PGE2-modified,PEGylated,ditellurium-crosslinked polyethylenimine(PEI)was developed to surface-decorate the Dex-encapsulated mesoporous silica nanoparticles(MSNs),which simultaneously condensed siRAGE and gated the MSNs to prevent the Dex pre-leakage.Upon intravenous injection to IR-injured rats,the nanotherapeutics could be efficiently transported into the inflamed cardiomyocytes via PGE2-assisted recognition of over-expressed E-series of prostaglandin(EP)receptors on the cell membranes.Intracellularly,the over-produced ROS degraded PPTP into small segments,promoting the release of siRAGE and Dex to mediate effective RAGE silencing(72%)and cooperative antiinflammatory effect.As a consequence,the nanotherapeutics notably suppressed the myocardial fibrosis and apoptosis,ultimately recovering the systolic function.Therefore,the current nanotherapeutics represent an effective example for the codelivery and on-demand release of nucleic acid and chemodrug payloads,and might find promising utilities toward the synergistic management of myocardial inflammation.

Shenzhu Guanxin Recipe Granules (参术冠心方颗粒) for Improving Exercise Tolerance in Patients with Stable Angina (SERIES Trial):A Protocol of Multicenter,Randomized,Double-Blind,Placebo Parallel Controlled Clinical Trial

Background: Many patients with chronic angina experience anginal episodes despite successful recanalization, antianginal and antiischemic medications. Empirical observations suggested that Shenzhu Guanxin Recipe Granules(参术冠心方颗粒, SGR), a Chinese herbal compound, exerted potential impacts on increased treadmill exercise performance and angina relieve. However, there has been no systematic study to clarify the impact of SGR on exercise tolerance in patients with stable angina. The SERIES(ShEnzhu guanxin Recipe for Improving Exercise tolerance in patients with Stable angina) trial is designed to determine the effects of SGR on exercise duration, electrocardiographic(ECG) evidence of myocardial ischemia, and incidence of major adverse cardiac events(MACE) in stable anginal patients. Methods: A total of 184 eligible patients with stable angina will be randomly assigned to receive placebo or SGR(10 g/day for 12 weeks) in a 1:1 ratio. The primary outcome will be the change from baseline in total exercise tolerance duration, time to onset of angina and ECG ischemia during exercise treadmill testing performed over a 12-week study period. The secondary outcome will include ECG measures, the occurrence and composite of MACE and the Seattle Angina Questionnaire score. Moreover, the coronary microcirculation will be evaluated to explore the possible effects in response to treatment of SGR. After the procedure, all participants will be followed up by interview at 3 and 6 months, enquiring about any cardiac events, hospitalizations, cardiac functional level and medication usage. Additionally, the occurrence of adverse events will be evaluated at each follow-up. Discussion: This study may provide novel evidence on the efficacy of SGR in improving exercise tolerance and potentially reducing clinical adverse events.
