AIM： To elucidate the mechanism of liver protection by inhibition of Kupffer cells （KCs） function.METHODS： All the animals were randomly divided into three groups. Blockade group （gadolinium chloride solution （GdCl3） injection plus ischemia/reperfusion （I/R） injury）：GdCl3 solution was injected once every 24 h for 2 d via the tail vein before I/R injury. Non-blockade group （saline solution injection plus I/R injury）： saline instead of GdCl3 as a control was injected as in the blockade group. Sham group： saline was injected without I/R injury. Liver samples were collected 4 h after blood inflow restoration. The blockade of the function of KCs was verified by immunostaining with an anti-CD68 mAb. Toll-like receptor2 （TLR2） was immunostained with a goat antimouse polydonal anti-TLR2 antibody. Membrane proteins were extracted from the liver samples and TLR2 protein was analyzed by Western blot. Portal vein serum and plasma were taken respectively at the same time point for further detection of the levels of tumor necrosis factor-α （TNF-α） and alanine aminotransferase （ALT）, an indicator of liver function.I/R injury via downregulation of theexpression of TLR2.RESULTS： Compared to non-blockade group, CD^68＋ cells significantly reduced in blockade group （OPTDI, optical density integral）： 32.97±10.55 vs 185.65±21.88, P〈0.01） and the liver function impairment was relieved partially （level of ALT： 435.89±178.37 U/L vs 890.21±272.91 U/L,P〈0.01）. The expression of TLR2 protein in blockade group significantly decreased compared to that in non- group（method of immunohistochemistry, OPDTI： 75.74±17.44 vs 170.58±25.14, P〈0.01; method of Western blot, A value： 125.89±15.49 vs 433.91±35.53, P〈0.02）. The latter correlated with the variation of CD68 staining （r = 0.745,P〈0.05）. Also the level of portal vein TNF-α decreased in blockade group compared to that in non-blockade group （84.45±14.73 ng/L vs 112.32±17.56 ng/L, P〈0.05）, butwas still higher than that in sham group （84.45±14.73

Lithium-ion capacitors(LICs)have attracted wide attention due to their potential of achieving merits of high-power output as well as high energy ***-ever,the key issue of kinetics mismatch between anode and cathode hinders the electrochemical performance of ***,a vanadium nitride composite with nanoparti-cles embedded in carbon matrix(VN-C)was prepared as an efficiently pseudocapacitive anode material with high electronic conductivity and fast Li-ion diffusion *** VN-C composites were synthesized through one-step ammonia heating treatment at different temperatures among which the sample annealed at 600℃exhibits high specific capacity(513 mAh·g^(-1)at 0.1 A·g^(-1)),outstanding rate performance(~300 mAh·g^(-1)at 10 A·g^(-1)),and excellent cyclic steadiness(negligible capacity decay over 2000 cycles)in half-cell devices.A high-performance lithium-ion capacitor device was also fabricated by using VN-C-600 as the anode and activated carbon as the cath-ode,delivering a maximum energy density of 112.6 Wh·kg^(-1)and an extreme power density of 10 kW·kg^(-1).

BACKGROUND: Toil-like receptors (TLRs) are a group of evolutionarily conserved pattern recognition receptors involved in the activation of the immune system in response to various pathogens. In this study, we elucidated the relationship between activation of TLR4 and liver injury in partial hepatic ischemia/reperfusion (I/R) injury in mice. METHODS: BALB/c mice were used in a model of partial hepatic I/R injury, and the changes of TLR4 gene expression in ischemic liver lobes were detected with real-time polymerase chain reaction (RT-PCR). The levels of plasma ALT and endotoxin in the portal vein were measured. TLR4-deficient mice (C3H/Hej) and wild type mice (C3H/Heouj) were used in a model of I/R injury; liver function impairment and the level of serum TNF-α were observed. RESULTS: After one hour ischemia, the expression of TLR4 mRNA increased at the 1st, 3rd hour of reperfusion, indicating the value of △Ct(1st hour: 1.21±0.87vs. 5.85±1.07, t=13.72, P<0.01; 3rd hour: 0.85±0.92vs. 6.11±1.24, t=16.33, P<0.01). No endotoxemia developed in every group of mice. At the 3rd hour of reperfusion, the level of serum TNF-α was significantly higher than that of sham group (Hej: 152±43 pg/ml vs. 18±10pg/ml, t=5.26, P<0.01; Heouj: 249±52pg/ml vs. 25±13 pg/ml, t=7.24, P<0.01). At the 1st, 3rd hour reperfusion, the level of plasmid ALT in Hej mice was lower than that in Heouj mice (1st hour 662±106 U/Lvs. 1216±174 U/L, t=4.21, P<0.01; 3rd hour 1145±132U/L vs. 29584±187 U/L, t=13.72, P<0.01). The level of serum TNF-αwas lower than that in Heouj mice (1524.43 U/L vs. 2494±52 U/L, t=3.94, P<0.01) at the 3rd hour reperfusion. CONCLUSION: TLR4 activation causes partial hepatic I/R injury through release of TNF-α.

AIM： Recent laboratory and epidemiological studies suggest that vitamin D is a potential agent for colorectal cancer prevention. Its function is partially mediated by the vitamin D receptor （VDR）. The aim of this study was to investigate whether a novel G （allele ‘U’g〉A （allele ‘u’ polymorphism （Tru9I） in the VDR intron 8 region is associated with risk for colorectal adenoma in a colonoscopy-based case-control study. METHODS： Genotyping for a total of 391 subjects was carried out through PCR and restriction fragment length polymorphism. RESULTS： The frequencies of ‘U’ and ‘u’ alleles were 89.3% and 10.7%, respectively. The ‘Uu’ and ‘uu’ genotypes were associated with decreased risk for adenoma （OR, 0.71; 95%CI, 0.40-1.25）. The inverse association was more pronounced for multiple adenomas and adenomas that were larger had moderate or greater dysplasia, or were sessile： the odds ratios （ORs） were, 0.51 （95%CI, 0.21-1.24）, 0.37 （95%CI, 0.11-1.28）, 0.68 （95%CI, 0.33- 1.41）, and 0.36 （95%CI, 0.13-0.97） respectively. In joint/ combined analyses, inverse associations were more obvious among those who had at least one ‘u’ allele and also were younger （OR, 0.60; 95%CI, 0.26-1.37）, women （OR, 0.38; 95%CI, 0.17-0.88）, did not smoke （OR, 0.39; 95%CI, 0.13-1.23）, or took NSAID （OR, 0.38; 95%CI, 0.12-1.25）, but no evidence existed for interactions with calcium or vitamin D ***： Our findings suggest that the VDR TrugI polymorphism may be associated with lower risk for colorectal adenoma, particularly in interaction with various risk factors, but not with calcium or vitamin D.

AIM: p53-Inducible ribonucleotide reductase small subunit 2 (p53R2) encodes a 351-amino-acid peptide, which catalyzes conversion of ribonucleoside diphosphates to the corresponding deoxyribonucleotides required for DNA replication and repair. A recent study reported that a point mutation (G/T) in the p53 binding sequence in a colon cancer cell line completely impaired p53R2 protein ***: We screened the p53R2 gene coding regions and a regulatory region which contains a p53 binding sequence in 100 patients with colorectal adenoma and 100 control subjects using PCR, cold SSCP, and direct DNA ***: Although we did not identify genetic variation in all nine exons, four regulatory-region variants were found,of which three were single nucleotide polymorphisms (SNPs) (nt 1 789 C/G, nt 1 928 A/G, 1 933 T/C), and one was 20 bp insertion which replaced a ATTTT between nt 1 831 and 1 835. Additionally, we determined the frequency of these p53R2 variants in a recently concluded case-control study of incident sporadic colorectal adenomas (163 cases and 210 controls).CONCLUSION: Although more detailed functional characterizations of these polymorphisms remain to be undertaken, these polymorphic sites may be useful for identifying alleles associated with mis-splicing, additional transcript factors and, more generally, in cancer-susceptibility association studies.

Late Paleozoic to early Mesozoic volcanic-sedimentary history of the southeastern margin of the Central Asian Orogenic Belt: Timing of uplift of the Inner Mongolia paleo-uplift

The Kulun region is located in the eastern Bainaimiao arc belt on the southeastern margin of the Central Asian Orogenic Belt(CAOB), adjacent to the North China Craton(NCC) to the south. This region records the closure of the PaleoAsian Ocean during the Paleozoic to early Mesozoic and is key to study the tectonic evolution of the northern margin of the *** conducted zircon U-Pb geochronology and Hf isotope analysis of volcanic-sedimentary and intrusive rocks in the Kulun region. The volcanic-sedimentary rocks were previously assigned an early Carboniferous age, but include volcanic rocks with ages of 267–266 Ma and sedimentary rocks with youngest detrital zircon age peaks of 265–244 Ma, which were intruded by a granite with an age of 242 Ma. This indicates that these rocks are actually middle Permian to Middle Triassic in age(267–242Ma) and correspond to the Qingfengshan and Liujiagou formations. Moreover, two volcanic rock samples have high εHf(t) values(+1.6 to +16.1) and relatively young two-stage Hf model ages(1095–327 Ma), indicating their sources associated with the CAOB. The age spectra and Hf isotopic characteristics of detrital zircons from the sedimentary rocks suggest that, during the middle Permian to Middle Triassic, the sedimentary provenance in the Kulun region was mainly the CAOB. However, by the Early Jurassic, the provenance changed to the NCC(i.e., the Inner Mongolia Paleo-uplift, IMPU). This result, combined with the development of a series of conglomerates on the northern margin of the NCC in the Late Triassic(i.e., Xingshikou, Xiaoyingzi,and Xiaohekou formations), indicates the Kulun region experienced an abrupt change in paleogeography during the Late Triassic, with the IMPU undergoing significant uplift and thus becoming the main sediment source in the Kulun region. Based on the Late Triassic alkaline igneous belt(235–215 Ma) in the IMPU, we speculate that this uplift was caused by detachment of the southward-subducting Paleo-Asian oceanic

Rituximab Treatment Strategy for Patients with Diffuse Large B-Cell Lymphoma after First-Line Therapy： A Systematic Review and Meta-Analysis

Background： Rituximab in combination with cyclophosphamide, doxorubicin, vincristine, and prednisone （CHOP） significantly prolonged event-free survival in first-line chemotherapy for patients with diffuse large B-cell lymphoma （DLBCL）. But relapse and refractory DLBC L occur frequently. Although rituximab is effective, its role in salvage therapy after autologous transplant remains unclear. Maintenance therapy with rituximab in responding patients after first line chemotherapy may be a useful novel approach capable of eradicating minimal residual disease and to bring survival benefit. This systematic review and meta-analysis evaluated the effects of rituximab maintenance treatment and salvage therapy of patients with DLBCL. Methods： We performed a systematic review and meta-analysis of randomized controlled trials and compared rituximab maintenance or salvage therapy at relapse with observation. We searched the Cochrane Library, PubMed, EMBASE, conference proceedings, databases of ongoing trials, and references of published trials. Two reviewers independently assessed the quality of the trials and extracted data. Hazard ratios for time-to-event data were estimated and pooled. Results： Seven trials including 1470 DLBCL patients were included in this systematic review and recta-analysis. Patients treated with maintenance rituximab have better overall survival （OS） and event-tree survival （EFS） than patients in the observation arm, but there was no statistical significance. Patients who received rituximab salvage therapy for relapse or refractory DLBCL have statistically significantly better OS [HR of death = 0.72, 95% CI （0.55-0.94）, P - 0.02], progression-free survival （PFS） [HR - 0.61,95% CI（0.52-0.72）, P 〈 0.05], odds ratio （OR） [RR = 1.26, 95% CI（1.07-1,47）, P = 0.004] than patients in the observation arm. The term vulnerable patient has been proposed to define subjects susceptible to an acutecoronary syndrome （ACS） or sudden cardiac death based on plaque characteristics, blood abnormalities, or myocardial vulnerability. 1 It will be important in the future to identify both vulnerable patients and vulnerable plaques. Atherosclerotic arteries obtained at autopsy from patients who died suddenly of cardiac causes indicate that pregnancyassociated plasma protein-A （ PAPP-A ） was abundantly expressed in plaque cells and in the extracellular matrix of ruptured and eroded unstable plaques, but not in stable plaques. Here we examined circulating PAPP-A levels in patients with ACS in order to evaluate its potential use in identifying vulnerable patients.
