Inhibition Mechanism of Novel Pyrazolo[1,5-a]pyrazin-4(5H)-one Derivatives Against Proliferation of A549 and H322 Cancer Cells

Objective To explore the inhibition mechanism and safety of pyrazolo[1,5-a]pyrazin-4(5H)-one derivatives against proliferation of human lung cancer A549 cells, H322 cells, and human umbilical vein endothelial cell(HUVEC). Methods Cells were treated with 40 μmol/L of the ppo3 a, ppo3 b, ppo3 i, and 0.1% DMSO(control) for 48 hours, respectively. Apoptosis was determined by Hoechst 33258 staining assay in H322 and A549 cells. Cell cycle distribution was determined by flow cytometry analysis in A549 cell. LC3-II, p53, and heat shock protein(HSP) 70 protein levels were detected by Western blotting in A549 cells treated with ppo3 b for 48 hours. The morphology and viability of HUVEC were observed by inverted microscope and sulforhodamine B(SRB) assay. Results Ppo3 a, ppo3 b, and ppo3 i significantly induced apoptosis in H322 and A549 cells. A strong G1-phase arrest was concomitant with the growth inhibitory effect on A549 cells. Ppo3 b effectively elevated the p53 protein level, but significantly reduced the HSP70 protein level. There were no significantly inhibitory effect on the morphology and viability of HUVEC when treated with ppo3 a, ppo3 b, and ppo3 i. Conclusions ppo3 a, ppo3 b, and ppo3 i could inhibit H322 proliferation through apoptosis and inhibit A549 through apoptosis and G1-phase arrest. The protein p53 and HSP70 might involve in the inhibition effects. These derivatives might be a clue to find effective and safe drug for lung cancers.

Late Paleozoic to early Mesozoic volcanic-sedimentary history of the southeastern margin of the Central Asian Orogenic Belt:Timing of uplift of the Inner Mongolia paleo-uplift

The Kulun region is located in the eastern Bainaimiao arc belt on the southeastern margin of the Central Asian Orogenic Belt(CAOB),adjacent to the North China Craton(NCC)to the *** region records the closure of the PaleoAsian Ocean during the Paleozoic to early Mesozoic and is key to study the tectonic evolution of the northern margin of the *** conducted zircon U-Pb geochronology and Hf isotope analysis of volcanic-sedimentary and intrusive rocks in the Kulun *** volcanic-sedimentary rocks were previously assigned an early Carboniferous age,but include volcanic rocks with ages of 267–266 Ma and sedimentary rocks with youngest detrital zircon age peaks of 265–244 Ma,which were intruded by a granite with an age of 242 *** indicates that these rocks are actually middle Permian to Middle Triassic in age(267–242Ma)and correspond to the Qingfengshan and Liujiagou ***,two volcanic rock samples have highεHf(t)values(+1.6 to+16.1)and relatively young two-stage Hf model ages(1095–327 Ma),indicating their sources associated with the *** age spectra and Hf isotopic characteristics of detrital zircons from the sedimentary rocks suggest that,during the middle Permian to Middle Triassic,the sedimentary provenance in the Kulun region was mainly the ***,by the Early Jurassic,the provenance changed to the NCC(i.e.,the Inner Mongolia Paleo-uplift,IMPU).This result,combined with the development of a series of conglomerates on the northern margin of the NCC in the Late Triassic(i.e.,Xingshikou,Xiaoyingzi,and Xiaohekou formations),indicates the Kulun region experienced an abrupt change in paleogeography during the Late Triassic,with the IMPU undergoing significant uplift and thus becoming the main sediment source in the Kulun *** on the Late Triassic alkaline igneous belt(235–215 Ma)in the IMPU,we speculate that this uplift was caused by detachment of the southward-subducting Paleo-Asian oceanic plate,which occurred in a post-o

The maneuverability and stealth of aerial-aquatic vehicles(AAVs)is of significant importance for future integrated air-sea combat *** improve the maneuverability and stealth of AAVs near the water surface,this paper proposed a high-maneuverability skipping motion strategy for the tandem twin-rotor AAV,inspired by the motion behavior of the flying fish to avoid aquatic and aerial predators near the water *** novel tandem twin-rotor AAV was employed as the research subject and a strategybased ADRC control method for validation,comparing it with a strategy-based PID control *** results indicate that both control methods enable the designed AAV to achieve high stealth and maneuverability near the water surface with robust control *** strategy-based ADRC control method exhibits a certain advantage in controlling height,pitch angle,and reducing impact *** motion strategy will offer an inspiring approach for the practical application of AAVs to some extent.

The global spin alignment of vector mesons has been observed by the STAR collaboration at the Relativistic Heavy Ion Collider(RHIC)at Brookhaven National Laboratory(BNL).It provides a unique opportunity to probe the correlation between the po-larized quark and antiquark in the strongly coupled quark-gluon plasma(sQGP)produced in relativistic heavy ion collisions,opening a new window to explore the properties of *** addition,spin alignments of vector mesons have also been observed in other high-energy particle collisions such as e^(+)e^(−)annihilations at high energies where hadron production is dominated by quark fragmentation *** results obtained are quite different from those obtained in heavy ion collisions where quark coalescence/combination mechanism dominates suggesting strong dependence on hadronization *** comprehensive studies in different hadronization processes are *** this article,we present a brief review of theoretical and experimen-tal advances in the study of vector meson’s spin alignments in a variety of high-energy particle collisions,with emphasis on hadronization mechanisms.

Enhancing entanglement and steering in a hybrid atom–optomechanical system via Duffing nonlinearity

We introduce a novel scheme for achieving quantum entanglement and Einstein–Podolsky–Rosen(EPR) steering between an atomic ensemble and a mechanical oscillator within a hybrid atom–optomechanical system. The system comprises an optical cavity, a two-level atomic ensemble and a mechanical resonator that possesses Duffing nonlinearity. The interaction between these components is mediated by the cavity mode, which is driven by an external laser. Our findings indicate that optimizing the coupling strengths between photons and phonons, as well as between atoms and the cavity,leads to maximal entanglement and EPR steering. The amplitude of the driving laser plays a pivotal role in enhancing the coupling between photons and phonons, and the system maintains robust entanglement and EPR steering even under high dissipation, thereby mitigating the constraints on initial conditions and parameter precision. Remarkably, the Duffing nonlinearity enhances the system's resistance to thermal noise, ensuring its stability and entanglement protection. Our analysis of EPR steering conditions reveals that the party with lower dissipation exhibits superior stability and a propensity to steer the party with higher dissipation. These discoveries offer novel perspectives for advancing quantum information processing and communication technologies.

Encapsulating lipase on the surface of magnetic ZIF-8 nanosphers with mesoporous SiO2 nano-membrane for enhancing catalytic performance

The preparation of immobilized enzyme with excellent performance is one of the difficulties that restrict the application of enzyme catalysis technology. Here, Candida rugosa lipase（CRL） was firstly adsorbed on the surface of magnetic zeolitic imidazolate framework-8（ZIF-8） nanospheres, which was further encapsulated with a mesoporous SiO2nano-membrane formed by tetraethyl orthosilicate（TEOS） polycondensation. Consequently, lipase could be firmly immobilized on carrier surface by physical binding rather than chemical binding, which did not damage the active conformation of enzyme. There were mesopores on the silica nano-membrane, which could improve the accessibility of enzyme and its apparent catalytic activity. Moreover, silica membrane encapsulation could also improve the stability of enzyme, suggesting an effective enzyme immobilization strategy. It showed that TEOS amount and the encapsulation time had significant effects on the thickness of silica membrane and the enzyme activity. The analysis in enzyme activity and protein secondary structure showed that lipase encapsulated in silica membrane retained the active conformation to the greatest extent. Compared with the adsorbed lipase, the encapsulated lipase increased its thermostability by 3 times and resistance to chemical denaturants by 7 times. The relative enzyme activity remained around 80% after 8 repetitions, while the adsorbed lipase only remained at7.3%.

CaH2-promoted activity of Ni-carbonate interface for CO_(2) methanation

Transition metal-carbonate interfaces often act as active sites in heterogeneous catalytic *** interface between transition metal and metal carbonate exhibits a dynamic equilibrium during the CO_(2)hydrogenation reaction,involving surface carbonate hydrogenation and CO_(2)***,there have been few reports on engineering the activity of the interface between transition metal and alkaline earth metal carbonate for catalytic CO_(2)*** work demonstrated that the incorporation of CaH_(2)in Ni/CaCO_(3)enhances the CO_(2)methanation activity of the *** CO_(2)conversion for Ni/CaH_(2)-CaCO_(3)reached 68.5%at 400°C,which was much higher than that of the Ni/CaCO_(3)(31.6%) and Ni/CaH_(2)-CaO (42.4%) ***,the Ni/CaH_(2)-CaCO_(3)catalysts remained stable during the stability test for 24 h at 400°C and 8 *** research revealed that CaH_(2)played a crucial role in promoting the activity of the Ni-carbonate interface for CO_(2)***_(2)could modify the electronic structure of Ni and tune the structural properties of CaCO_(3)to generate medium basic sites (OH groups),which are favorable for the activation of H2and CO_(2).In-situ Fourier transform infrared spectroscopy (FTIR) analysis combined with density functional theory calculations demonstrated that CO_(2)activation occurs at the hydroxyl group (OH) on the CaH_(2)-modified Ni-carbonate surface,leading to the formation of CO_(3)H****,our study has confirmed that CO_(2)methanation over the Ni/CaH_(2)-CaCO_(3)catalysts proceeds via the formate pathway.

Specific Heat Capacity and Coordination Number of Nano-Confined Transcritical Water

In the field of nano energy,investigating the specific heat capacity and coordination number of nano-confined water is highly significant for gaining a better understanding of the energy and microstructure of confined *** this work,we employed the method of molecular dynamics（MD） simulation to calculate the specific heat capacity at constant volume and coordination number of water molecules confined in carbon nanotubes（CNTs） under different conditions（T=600-700 K,P=21.776 and 25 MPa,CNT diameter=0.949-5.017nm）.The results showed that near the critical point,the specific heat capacity at constant volume of confined water was lower than that of bulk water,and the energy fluctuation showed a trend of first increasing and then remaining unchanged with the increase of temperature and CNT *** them,the saturation point of temperature is 650 K（reduced pressure Pr=1） and 660 K（Pr=1.15）,and the saturation point of CNT diameter is 2.034 ***,the pseudo-critical temperature of confined water was the same as bulk water,and it increased with the increase of critical ***,with the increase of CNT diameter,the coordination number of confined water increased rapidly,and reaches the saturation state when the CNT diameter is 2.034 *** investigation revealed the mass and energy characteristics of nano-confined water near the critical point,which could provide guidance for the critical phase transition of nano-confined water.

Effect of Combination Regimen of Low-dose Gossypol Acetic Acid with Steroid Hormones on Expression of Protein Kinase C alpha (PKC-α) and Cyclin D1 in Rat Testes

Objective To investigate the contraceptive mechanism of combination regimen of low- dose gossypol acetic acid （GA） with steroid hormones [desogestre/ethinylestradiol/ testosterone undeeanote（DSG/EE/TU）]. Methods Adult male rats were randomly divided into four groups. Group GH： rats were fed orally with gossypol acetic acid （GA, 12.5 mg/kg） and desogestrel （DSG, 0.125 mg/kg）/ ethinylestradiol （EE, 0.025 mg/kg）/testosterone undecanoate （TU, 100 mg/kg per day, qd X4 weeks or 10 weeks）; group G： a single dose of GA （12.5 mg/kg per day, qd X 4 weeks or I0 weeks）; group H： the same dosage of DSG/EE/TUas in group GH; group C： rats were treated with vehicle （1% methyl cellulose） as the control. Expression of protein kinase C alpha （PKC-a） and cyclin D1 in rat testes were tested mainly by immunohistochemistry （IHC） and Western blotting. Results IHC results showed that protein PKC- a was expressed mainly in interstitial tissue of testis among seminiferous tubule. The expression of PKC-oc in groups H and GH at week 10 was decreased greatly compared with that in group C. The protein cyclin D1 was expressed mainly in residual body of seminiferous tubule eavosurface and interstitial tissue among seminiferous tubule of testis. Western blotting results showed that the expression of PKC-a in groups H and GH at week 10 was decreased significantly compared with that in group C （P〈0.05）. The expression of cyclin D1 in groups G, H or GH at week 10 rose significantly compared with that in group C （P 〈0.05）.Conclusion The administration of low-dose gossypol acetic acid with steroid hormones for 10 weeks can decrease the expression of PKC-a greatly.
