The reinforcement of calcium phosphate materials with silk fibroin （SF） has been one of the strategies to overcome the brittleness. However, the lack of osteoinductivity may still restrict their further use. This study aimed to investigate the biocompatibility and osteogenesis capacity of a novel Semaphorin 3A-loaded chitosan microspheres/SF/a-tricalcium phosphate composite （Sema3A CMs/SF/a-TCP） in vitro. Sema3A was first incorporated into CMs, and the Sema3A CMs/SF/a-TCP composite was then prepared. The morphology of the CMs was observed using SEM. The in vitro release kinetics, cytotoxicity, and cell compatibility were evaluated, and the real-time quantitative polymerase chain reaction （RT-qPCR） and activity of alkaline phosphatase （ALP） were used to evaluate the osteogenesis capacity of the composite. The in vitro release of Sema3A from the Sema3A CMs/SF/a-TCP composite showed a temporally controlled manner. The extract of the Sema3A CMs/SF/a-TCP composite presented no obvious side effect on the MC3T3-E1 cell proliferation, nor promote cell proliferation. The MC3T3-E1 cells were well-spread and presented an elongated shape on the Sema3A CMs/SF/a-TCP composite surface; the ALP activity and the osteogenic-related gene expression were higher than those seeded on the surface of the CMs/SF/a-TCP and SF/a-TCP composites. In conclusion, Sema3A CMslSF/a-TCP has excellent biocompatibility and contributes to the osteoblastic differentiation of MC3T3-E1 cells.

To search for new bioactive materials which can be used as the substitute of bone repairing and drug carriers, Sema3A-Ioaded chitosan microspheres （SLCM） and silk fibroin （SF） were mixed with calcium sulfate cement （CSC）. SEM, particle size analysis and swelling rate determination were performed to study properties of the microspheres. The drug loading, encapsulation efficiency and drug release rate were determined by EliSA. Microspheres with different SLCM weight contents （0.5%, 1% and 5%） were prepared to determine which one has the strongest mechanical properties and the appropriate setting time. It was revealed that CSC/SF/0.5SLCM has satisfactory mechanical properties, and its in vitro biocompatibility was assessed by MTS. Chitosan microspheres （5-18 μm） were globular, the surface was smooth, and the swelling rate is （77.02 ± 5.57）%. With this formula, the setting time was increased with the addition of SLCM in CSCISF, and the cumulative drug release rate is 44.62% in 28 d. XRD results demonstrate that the main component is calcium sulfate. Also it was found that CSCiSFI 0.5SLCM supports the growth of MC3T3 cells. Thus the preparation of CSCISFIO.5SLCM was reliable, and the products had good structures, physical properties and biocompatibility, appearing to be a promising bone substitute material.

Objective This study investigated the impact of occupational mercury(Hg) exposure on human gene transcription and expression, and its potential biological *** Differentially expressed genes related to Hg exposure were identified and validated using gene expression microarray analysis and extended validation. Hg-exposed cell models and PTEN lowexpression models were established in vitro using 293T cells. PTEN gene expression was assessed using qRT-PCR, and Western blotting was used to measure PTEN, AKT, and pi3K protein levels. IL-6 expression was determined by *** Combined findings from gene expression microarray analysis, bioinformatics, and population expansion validation indicated significant downregulation of the PTEN gene in the high-concentration Hg exposure group. In the Hg-exposed cell model(25 and 10 μmol/L), a significant decrease in PTEN expression was observed, accompanied by a significant increase in pi3K, AKT, and IL-6 ***, a low-expression cell model demonstrated that PTEN gene knockdown led to a significant decrease in PTEN protein expression and a substantial increase in pi3K, AKT, and IL-6 *** This is the first study to report that Hg exposure downregulates the PTEN gene, activates the pi3K/AKT regulatory pathway, and increases the expression of inflammatory factors, ultimately resulting in kidney inflammation.

Amphiphilic diblock copolymers self-assemble into a variety of micellar structures with diverse shapes in selective solvents. Here, we study the concentration and temperature dependence of the packing structure of spherical micelles of a polyisoprene-b-poly(2- vinylpyridine)(pi-b-P2VP) diblock copolymer in toluene using synchrotron radiation small angle X-ray scattering (SR-SAXS) and atomic force microscopy (AFM) techniques. Randomly packed spherical micelles are detected in dilute solutions, while in concentrated solutions, face-centered cubic (FCC), body-centered cubic (BCC) mixed crystal structures, and pure BCC crystal structures are observed with an increase in concentration. In situ SAXS experiments on the FCC/BCC mixed crystal structures reveal a novel FCC/BCC → BCC → Disorder → BCC phase behavior during the thermal annealing process. These results demonstrated that the BCC phase is apparently more stable than the FCC phase in the current sphere-packing system and FCC/BCC is a metastable state. The incompatibility of the pi and P2VP blocks decreases at a higher temperature and renders the variation of domain spacing.

Six new ent-abietane diterpenoids,abientaphlogatones A−F(1−6),along with two undescribed ent-abietane diterpenoid glucosides,abientaphlogasides A−B(7−8)and four known analogs were isolated from the aerial parts of Phlogacanthus curviflorus(***).The structures of these compounds were determined using high-resolution electrospray ionization mass spectrometry(HR-ESI-MS),one-dimensional and two-dimensional nuclear magnetic resonance(NMR)spectroscopy,electronic circular dichroism(ECD)spectra,and quantum chemical ***,compounds 5 and 6 represented the first reported instances of entnorabietane diterpenoids from the genus *** theβ-hematin formation inhibition assay,compounds 2,4,7−10,and 12 displayed antimalarial activity,with IC_(50) values of 12.97−65.01μmol·L−***,compounds 4,5,8,and 10 demonstrated neuroprotective activity in PC12 cell injury models induced by H_(2)O_(2) and MPP+.

With advances in Si-based technology infrastructures and the rapid integration of Si-based optoelectronics, Si-based optoelectronic synaptic devices have the potential to greatly facilitate the large-scale deployment of neuromorphic computing. The incorporation of solution-processable polymer semiconductors into Si-based optoelectronics may enable the cost-effective fabrication of optoelectronic synaptic ***(3-hexylthiophene)(P3HT) is a semiconducting polymer used to manufacture optoelectronic synaptic transistors with P3HT channels and Si gates. The gate dielectric between them consists of a *** inorganic-organic Si/P3HT optoelectronic synaptic transistors can mimic synapses when exposed to optical and electrical stimuli. The Si/P3HT synaptic devices can spatiotemporally integrate optical and electrical stimuli to mimic cross-modal learning.

A novel Fe-10Cr ferritic/martensitic steel called SIMP was chosen to investigate synergistic effects of H and He on the mechanical properties of structural materials for innovative nuclear energy systems. Sequential and separate irradiation experiments on SIMP steel specimens at room temperature using H and He ions with various energy levels were conducted to produce an ion deposition plateau at 300-650 nm. The indentation stress-strain responses were examined using spherical nanoindentation tests after the irradiation experiments. It was found that the sequential irradiation by He and H produced a higher indentation yield stress than separate irradiation, indicating that the hardening was enhanced by the synergy of the H and He irradiation. The micro-mechanism responsible for enhancing the hardening of the SIMP steel through the H and He synergy was investigated using Doppler broadening spectroscopy detection and transmission electron spectroscopy observations.

A analyzed model of gear with wheel hub, web and rim was derived from the Mindlin moderate plate theory. The gear was divided into three annular segments along the locations of the step variations. Traverse displacement, rotation angle, shear force and fiexural moment were equal to ensure the continuity along the interface of the wheel hub, web and rim segments. The governing differential equations for harmonic vibration of annular segments were derived to solve the gear vibration problem. The influence of hole to diameter ratios, segment thickness ratios, segment location ratios, Poisson ratio on the vibration behavior of stepped circular Mindlin disk were calculated, tabletted and plotted. Comparisons were made with the frequencies arising from the presented method, finite elements method, and structure modal experiment. The result correlation among these three ways is very good. The largest error for all frequencies is 5.46%, and less than 5% for most frequencies.
