Interstitial cystitis(IC)is a severely debilitating and chronic disorder with unclear etiology and pathophysiology,which makes the diagnosis difficult and treatment *** investigate the role of immunity in IC bladders,we sequenced 135,091 CD45^(+) immune cells from 15 female patients with IC and 9 controls with stress urinary incontinence using single-cell RNA sequencing(scRNA-seq).22 immune subpopulations were identified in the constructed *** them,M2-like macrophages,inflammatory CD14^(+) macrophages,and conventional dendritic cells had the most communications with other immune ***,a significant increase of central memory CD4^(+)T cells,regulatory T cells,GZMK^(+)CD8^(+)T cells,activated B cells,un-switched memory B cells,and neutrophils,and a significant decrease of CD8+effector T cells,Th17 cells,follicular helper T cells,switched memory B cells,transitional B cells,and macrophages were noted in IC *** enrichment analysis identified a virus-related response during the dynamic change of cell proportion,furthermore,the human polyomavirus-2 was detected with a positive rate of 95%in urine of patients with *** integrating the results of scRNA-seq with spatial transcriptomics,we found nearly all immune subpopulations were enriched in the urothelial region or located close to fibroblasts in IC bladders,but they were discovered around urothelium and smooth muscle cells in control *** findings depict the immune landscape for IC and might provide valuable insights into the pathophysiology of IC.

The gene AtCSR encodes peptidyl-prolyl cis/trans isomerases (PPIases) that accelerate energetically unfavorable cis/trans isomerization of the peptide bond preceding proline *** our studies,we found that AtCSR was associated with cadmium (Cd)-sensitive response in *** results show that AtCSR expression was triggered by Cd-stress in wild type *** expression of some genes responsible for Cd2+ transportation into vacuoles was induced,and the expression of the iron-regulated transporter 1 (IRT1) related to Cd2+ absorption from the environment was not induced in wild type with Cd2+ *** expression of Cd-transportation related genes was not in response to Cd-stress,whereas IRT expression increased dramatically in atcsr-2 with Cd2+ *** expression of glutathione 1 (GSH1) was consistent with GSH being much lower in atcsr-2 in comparison with the wild type with Cd2+ ***,malondialdehyde (MDA),hydrogen peroxide,and Cd2+ contents,and activities of some antioxidative enzymes,differed between the wild type and *** sulfide (H2S) has been confirmed as the third gas-transmitter over recent *** findings revealed that the expression pattern of H2 S-releasing related genes and that of Cd-induced chelation and transportation genes matched well in the wild type and atcsr-2,and H2S could regulate the expression of the Cd-induced genes and alleviate Cd-triggered ***,one possible suggestion was given:down-regulation of atcsr-2,depending on H2S gas-transmitter not only weakened Cd2+ chelation,but also reduced Cd2+ transportation into vacuoles,as well as enhancing the Cd2+ assimilation,thus rendering atcsr-2 mutant sensitive to Cd-stress.

Cereal is an essential source of calories and protein for the global *** predicting cereal quality before harvest is highly desirable in order to optimise management for farmers,grading harvest and categorised storage for enterprises,future trading prices,and policy *** use of remote sensing data with extensive spatial coverage demonstrates some potential in predicting crop quality *** studies have also proposed models and methods for predicting such traits based on multiplatform remote sensing *** this paper,the key quality traits that are of interest to producers and consumers are *** literature related to grain quality prediction was analyzed in detail,and a review was conducted on remote sensing platforms,commonly used methods,potential gaps,and future trends in crop quality *** review recommends new research directions that go beyond the traditional methods and discusses grain quality retrieval and the associated challenges from the perspective of remote sensing data.

Investigation on short-term burst pressure of plastic pipes reinforced by cross helically wound steel wires

Plastic pipes reinforced by cross helically wound steel wires (PSP), which have exhibited excellent mechanical performance, consist of inner polyethylene (PE) layer, winding layer and outer PE layer. The winding layer is composed of two monolayers where steel wires are cross helically wound. An analytical procedure is developed to predict the short-term burst pressure of PSP as the monolayer is assumed to be elastic and orthotropic. The 3D anisotropic elasticity and Maximum Stress Failure Criterion are employed in the formulation of the elasticity problem. Good agreement between the theoretical results and the experimental data shows that the proposed approach can well predict the short-term burst pressure of PSP.

The cyclic plastic straining electrode technique has been used to investigate the transient electrochemical behaviour of Fe-26Cr1Mo stainless steel in 1M H2SO4 solution at a passive *** influence of plastic strain amplitude and plastic strain rate on the dissolution current response was analysed. The experimental results showed that the transient current was dependent on the competitive process of the surface film rupture and repassivation of the new surface. Adverse experiences in early life have long-lasting negative impacts on behavior and the brain in adulthood,one of which is sleep *** the corticotropin-releasing hormone(CRH)–corticotropin-releasing hormone receptor 1(CRHR1)system and nucleus accumbens(NAc)play important roles in both stress responses and sleep-wake regulation,in this study we investigated whether the NAc CRH–CRHR1 system mediates early-life stress-induced abnormalities in sleep-wake behavior in adult *** the limited nesting and bedding material paradigm from postnatal days 2 to 9,we found that early-life stress disrupted sleep-wake behaviors during adulthood,including increased wakefulness and decreased non-rapid eye movement(NREM)sleep time during the dark period and increased rapid eye movement(REM)sleep time during the light *** stress-induced sleep disturbances were accompanied by dendritic atrophy in the NAc and both were largely reversed by daily systemic administration of the CRHR1 antagonist antalarmin during stress ***,Crh overexpression in the NAc reproduced the effects of early-life stress on sleep-wake behavior and NAc morphology,whereas NAc Crhr1 knockdown reversed these effects(including increased wakefulness and reduced NREM sleep in the dark period and NAc dendritic atrophy).Together,our findings demonstrate the negative influence of early-life stress on sleep architecture and the structural plasticity of the NAc,and highlight the critical role of the NAc CRH–CRHR1 system in modulating these negative outcomes evoked by early-life stress.
