A"messenger zone hypothesis"based on the visual three-dimensional spatial distribution of motoneurons innervating deep limb muscles

Coordinated contraction of skeletal muscles relies on selective connections between the muscles and multiple classes of the spinal motoneuro *** r,current research on the spatial location of the spinal motoneurons innervating differe nt muscles is *** this study,we investigated the spatial distribution and relative position of different motoneurons that control the deep muscles of the mouse hindlimbs,which were innervated by the obturator nerve,femoral nerve,inferior gluteal nerve,deep pe roneal nerve,and tibial *** were visualized by combining a multiplex retrograde tracking technique compatible with three-dimensional imaging of solvent-cleared o rgans(3DISCO)and 3-D imaging technology based on lightsheet fluorescence microscopy(LSFM).Additionally,we propose the hypothesis that"messenger zones"exist as interlaced areas between the motoneuron pools that dominate the synergistic or antagonist muscle *** hypothesize that these interlaced neurons may participate in muscle coordination as messenger *** revealed the precise mutual positional relationships among the many motoneurons that innervate different deep muscles of the *** only do these findings update and supplement our knowledge regarding the overall spatial layout of spinal motoneurons that control mouse limb muscles,but they also provide insights into the mechanisms through which muscle activity is coordinated and the architecture of motor circuits.

Artificial neural network-based method for discriminating Compton scattering events in high-purity germaniumγ-ray spectrometer

To detect radioactive substances with low activity levels,an anticoincidence detector and a high-purity germanium(HPGe)detector are typically used simultaneously to suppress Compton scattering background,thereby resulting in an extremely low detection limit and improving the measurement ***,the complex and expensive hardware required does not facilitate the application or promotion of this ***,a method is proposed in this study to discriminate the digital waveform of pulse signals output using an HPGe detector,whereby Compton scattering background is suppressed and a low minimum detectable activity(MDA)is achieved without using an expensive and complex anticoincidence detector and *** electric-field-strength and energy-deposition distributions of the detector are simulated to determine the relationship between pulse shape and energy-deposition location,as well as the characteristics of energy-deposition distributions for fulland partial-energy deposition *** relationship is used to develop a pulse-shape-discrimination algorithm based on an artificial neural network for pulse-feature *** accurately determine the relationship between the deposited energy of gamma(γ)rays in the detector and the deposition location,we extract four shape parameters from the pulse signals output by the *** learning is used to input the four shape parameters into the ***,the pulse signals are identified and classified to discriminate between partial-and full-energy deposition *** partial-energy deposition events are removed to suppress Compton *** proposed method effectively decreases the MDA of an HPGeγ-energy dispersive *** results show that the Compton suppression factors for energy spectra obtained from measurements on ^(152)Eu,^(137)Cs,and ^(60)Co radioactive sources are 1.13(344 keV),1.11(662 keV),and 1.08(1332 keV),respectively,and that the corresponding MDAs are 1.4%,5.

Effect of annealing temperature on microstructure and mechanical properties of Mg-Zn-Zr-Nd alloy with large final rolling deformation

In this study,the Mg-3Zn-0.5Zr-χNd(χ=0,0.6)alloys were subjected to final rolling treatment with large deformation of 50%.The impact of annealing temperatures on the microstructure and mechanical properties was *** rolled Mg-3Zn-0.5Zr-0.6Nd alloy exhibited an ultimate tensile strength of 386 MPa,a yield strength of 361 MPa,and an elongation of 7.1%.Annealing at different temperatures resulted in reduced strength and obviously increased elongation for both *** mechanical properties for the Mg-3Zn-0.5Zr-0.6Nd alloy were achieved after annealing at 200℃,with an ultimate tensile strength of 287 MPa,a yield strength of 235 MPa,and an elongation of 26.1%.The numerous deformed microstructures,twins,and precipitated phases in the rolled alloy could impede the deformation at room temperature and increase the work hardening *** annealing,a decrease in the work hardening effect and an increase in the dynamic recovery effect were obtained due to the formation of fine equiaxed grains,and the increased volume fraction of precipitated phases,which significantly improved the elongation of the ***,the addition of Nd element could enhance the annealing recrystallization rate,reduce the Schmid factor difference between basal and prismatic slip systems,facilitate multi-system slip initiation and improve the alloy plasticity.

High speed laser cladding as a new approach to prepare ultra-high temperature ceramic coatings

Ultra-high temperature ceramic(UHTC)coatings are used to protect the hot-end components of hypervelocity aerocrafts from thermal *** study provides a new approach to fabricate UHTC coatings with high speed laser cladding(HSLC)technology,and places more emphasis on investigating the formation mechanism,phase compositions,and mechanical properties of HSLC-UHTC *** show that a well-bonded interface between the coating and the tantalum alloy substrate can be *** coating is mainly composed of(Zr,Ta)C ceramic solid solution phase with a content of higher than 90% by volume and Ta(W)metal solid solution *** a relatively high powder feeding rate,the ZrC ceramic phase appears in the coating while a dense ZrC UHTC top layer with a thickness of up to~50μm is successfully *** for the mechanical properties of the HSLC coatings,the fracture toughness of the coating decreases with the increase of powder feeding *** increase of carbide solid solution phase can significantly improve the high temperature microhardness(552.7±1.8 HV0.5@1000℃).The innovative design of HSLC ZrC-based coatings on refractory alloys accomplishes continuous transitions on microstructure and properties from the substrate to the UHTC top layer,which is a very promising candidate scheme for thermal protection coating.

The interfacial instability of the poly(ethylene oxide)(PEO)-based electrolytes impedes the long-term cycling and further application of all-solid-state lithium metal *** this work,we have shown an effective additive 1-adaman-tanecarbonitrile,which con-tributes to the excellent per-formance of the poly(ethylene oxide)-based *** to the strong interaction of the 1-Adamantanecarboni-trile to the polymer matrix and anions,the coordination of the Li^(+)-EO is weakened,and the binding effect of anions is strengthened,thereby improving the Li^(+)conductivity and the electrochemical *** diamond building block on the surface of the lithium anode can sup-press the growth of lithium ***,the 1-Adamantanecarbonitrile also regulates the formation of LiF in the solid electrolyte interface and cathode electrolyte interface,which contributes to the interfacial stability(especially at high voltages)and protects the electrodes,enabling all-solid-state batteries to cycle at high voltages for long periods of ***,the Li/Li symmetric cell undergoes long-term lithium plating/stripping for more than 2000 h.1-Adamantanecarbonitrile-poly(ethylene oxide)-based LFP/Li and 4.3 V Ni_(0.8)Mn_(0.1)Co_(0.1)O_(2)/Li all-solid-state batteries achieved stable cycles for 1000 times,with capacity retention rates reaching 85%and 80%,respectively.

Internal limiting membrane peeling combined with silicone oil or air tamponade for highly myopic foveoschisis

AIM:To compare the efficacy of pars plana vitrectomy(PPV)combined with internal limiting membrane(ILM)and silicone oil or sterile air tamponade for the treatment of myopic foveoschisis(MF)in highly myopic ***:This retrospective study included 48 myopic eyes of 40 patients with MF and axial lengths(ALs)ranging from 26-32 mm treated between January 2020 and January *** patients were underwent PPV combined with ILM peeling followed by sterile air or silicone oil tamponade and followed up at least *** on the features on spectral-domain optical coherence tomography(SD-OCT),the eyes were divided into the MF-only group(Group A,n=15 eyes),MF with central foveal detachment group(Group B,n=20 eyes),and MF with lamellar macular hole group(Group C,n=13 eyes).According to AL,eyes were further divided into three groups:Group D(26.01-28.00 mm,n=12 eyes),Group E(28.01-30.00 mm,n=26 eyes),and Group F(30.01-32.00 mm,n=10 eyes).The best-corrected visual acuity(BCVA),central foveal thickness(CFT),and complications were ***:The patients included 16 males and 24 females with the mean age of 56±*** BCVA and CFT improved in all groups after surgery(P0.05).The intergroup differences of BCVA and CFT postoperatively were statistically significant in Group D,E,and *** eyes were injected with sterile air,and 28 eyes were injected with silicone oil for tamponade based on the ***,there was no statistically significant difference among Groups D,E,and F in terms of the results of sterile air or silicone oil *** mean recovery time was 5.9mo for MF patients subjected to silicone oil tamponade and 7.7mo for patients subjected to sterile air tamponade,and the difference was not statistically ***:PPV and ILM peeling combined with silicone oil or sterile air tamponade can achieve good results for MF in highly myopic eyes with ALs

Microplastics(MPs)have become a significant concern for their potential ***,the correlation between the size of plastic particles and their toxicity remains ***,we investigate the toxic effects of different sizes(80 nm,800 nm,8μm and 80μm)polystyrene MPs(PS-MPs)on the model organism Nile tilapia(Oreochromis niloticus).The results of bioluminescent imaging indicate that the 80 nm PS-MPs are more likely to invade the body.H&E staining shows severe damage on the intestinal villi and distinct hepatic steatosis in the 80 nm *** U labeling shows that the proliferation activity of intestinal and liver cells reduces significantly in the 80 nm *** gut microbiome analysis shows a severe imbalance of gut microbiota homeostasis in the 80 nm *** analysis of liver transcriptomics and metabolomics shows that the liver lipid metabolism is disordered in the 80 nm *** conclusion,this study confirms that the 80 nm PS-MPs are more likely to induce intestinal and liver *** the above lay the foundation for further study on the pathological damage of MPs to other organisms.

Automated screening of primary cell-based aptamers for targeting and therapy of pancreatic cancer

Although it has been developed for many years, nucleic acid aptamer screening technology still fails to be widely used, a considerable part of it is due to the variability of tumor cell morphology, which leads to the use of immortalized cell lines in the laboratory to screen nucleic acid aptamers for recognition ability of tumor cells in the diseased *** address this, primary cells that can be stably passaged were isolated and extracted from spontaneous tumors of genetically engineered pancreatic ductal adenocarcinoma model mice in this ***, an automated screening instrument for nucleic acid aptamers developed autonomously by our group was used to perform efficient aptamer screening using a limited number of cells, and the obtained nucleic acid aptamers were affinity verified at the cellular ***, to answer the question of the cell growth environment difference on the recognition ability of nucleic acid aptamers, we verified its targeting ability to tumors in vivo on a nude mice xenograft tumor model, and further used a common antitumor drug doxorubicin combined with nucleic acid aptamers to verify the drug loading ability of this aptamer combined with the targeting therapeutic ability.

Effect of Ti_(p) Content on the Work Hardening and Softening Behavior of Mg-Zn-Ca Alloy

The Ti_(p)/ZX60 composites with different Ti_(p) contents were prepared by semi-solid stirring *** extrusion,the microstructure,work hardening and softening behavior of the Ti_(p)/ZX60 composites were analyzed compared with the ZX60(Mg-6Zn-0.2Ca)*** results showed that the addition of Ti_(p) could not only promote the nucleation of dynamic recrystallized(DRXed)grains,but also be propitious to the refinement of DRXed *** increasing Ti_(p) content,the size of DRXed grains decreased accompanied with increasing volume fraction of DRXed *** the Ti_(p) content increased to 15 vol.%,the average size and volume fraction of DRXed grains reached to~0.32μm and 93.2%,***,both the strength and elongation were improved by the addition of Ti_(p).With increasing content of Ti_(p),a substantial increase in the strength was achieved with little change in the ***,the elongation decreased sharply when the Ti_(p) content further increased to 15 vol.%.The addition of Ti_(p) led to an increase in the work hardening rate,which gradually increased with increasing Ti_(p) ***,the softening rate did not demonstrate the same tendency with increasing Ti_(p) *** the conventional ceramic particles,the Ti_(p) can be deformed in coordination with the matrix alloy,which imparted a higher softening rate to the matrix *** though the softening rate improved as the Ti_(p) content increased from 5 to 10 vol.%,it dropped deeply as the Ti_(p) content increased to 15 vol.%owing to the fracture of Ti_(p) during extrusion.
