BACKGROUnD Portal shunt and immune status related to the spleen are related to the occurrence of hepatic encephalopathy(HE).It is unknown whether spleen volume before transjugular intrahepatic portosystemic shunt(TIPS)is related to postoperative *** To investigate the relationship between spleen volume and the occurrence of *** This study included 135 patients with liver cirrhosis who underwent TIPS,and liver and spleen volumes were elevated upon computed tomography *** Kaplan-Meier curve was used to compare the difference in the incidence rate of HE among patients with different spleen *** and multivariate Cox regression analyses were performed to identify the factors affecting overt HE(OHE).Restricted cubic spline was used to examine the shapes of the dose-response association between spleen volumes and OHE *** The results showed that 37(27.2%)of 135 patients experienced OHE during a 1-year follow-up *** with preoperative spleen volume(901.30±471.90 cm3),there was a significant decrease in spleen volume after TIPS(697.60±281.0 cm^(3))in OHE *** the severity of OHE increased,the spleen volume significantly decreased(Pnts with a spleen volume≥782.4 cm^(3),those with a spleen volume2.4 cm^(3) had a higher incidence of HE(Pn analysis showed that spleen volume was an independent risk factor for post-TIPS OHE(hazard ratio=0.494,Pne model showed that with an increasing spleen volume,OHE risk showed an initial increase and then decrease(PnCLUSIOn Spleen volume is related to the occurrence of OHE after *** spleen volume is an independent risk factor for post-TIPS OHE.

SHA: QoS-Aware Software and Hardware Auto-Tuning for Database Systems

While databases are widely-used in commercial user-facing services that have stringent quality-of-service(QoS)requirement,it is crucial to ensure their good performance and minimize the hardware usage at the same *** investigation shows that the optimal DBMS(database management system)software configuration varies for different user request patterns(i.e.,workloads)and hardware *** is challenging to identify the optimal software and hardware configurations for a database workload,because DBMSs have hundreds of tunable knobs,the effect of tuning a knob depends on other knobs,and the dependency relationship changes under different hardware *** this paper,we propose SHA,a software and hardware auto-tuning system for *** is comprised of a scaling-based performance predictor,a reinforcement learning(RL)based software tuner,and a QoS-aware resource *** performance predictor predicts its optimal performance with different hardware configurations and identifies the minimum amount of resources for satisfying its performance *** software tuner fine-tunes the DBMS software knobs to optimize the performance of the *** resource reallocator assigns the saved resources to other applications to improve resource utilization without incurring QoS violation of the database *** results show that SHA improves the performance of database workloads by 9.9%on average compared with a state-of-the-art solution when the hardware configuration is fixed,and improves 43.2%of resource utilization while ensuring the QoS.

5S-Heudelotinone alleviates experimental colitis by shaping the immune system and enhancing the intestinal barrier in a gut microbiota-dependent manner

Aberrant changes in the gut microbiota are implicated in many diseases,including inflammatory bowel disease(IBD).Gut microbes produce diverse metabolites that can shape the immune system and impact the intestinal barrier integrity,indicating that microbe-mediated modulation may be a promising strategy for preventing and treating *** fecal microbiota transplantation and probiotic supplementation are well-established IBD therapies,novel chemical agents that are safe and exert strong effects on the gut microbiota are urgently ***,we report the total synthesis of heudelotinone and the discovery of 5S-heudelotinone(an enantiomer)as a potent agent against experimental colitis that acts by modulating the gut microbiota.5S-Heudelotinone alters the diversity and composition of the gut microbiota and increases the concentration of short-chain fatty acids(SCFAs);thus,it regulates the intestinal immune system by reducing proinflammatory immune cell numbers,and maintains intestinal mucosal integrity by modulating tight junctions(TJs).Moreover,5S-heudelotinone(2)ameliorates colitis-associated colorectal cancer(CAC)in an azoxymethane(AOM)/dextran sulfate sodium(DSS)-induced in situ carcinoma ***,these findings reveal the potential of a novel natural product,namely,5S-heudelotinone,to control intestinal inflammation and highlight that this product is a safe and effective candidate for the treatment of IBD and CAC.

Semaphorin 7A impairs barrier function in cultured human corneal epithelial cells in a manner dependent on nuclear factor-kappa B

●AIM:To evaluate the role of semaphorin 7A(Sema7A)and its associated regulatory mechanisms in modulating the barrier function of cultured human corneal epithelial cells(HCEs).●METHODS:Barrier models of HCEs were treated with recombinant human Sema7A at concentrations of 0,125,250,or 500 ng/mL for 24,48,or 72h in *** electrical resistance(TEER)as well as Dextran-fluorescein isothiocyanate(FITC)permeability assays were conducted to assess barrier *** quantify tight junctions(TJs)such as occludin and zonula occludens-1(ZO-1)at the mRnA level,reverse transcriptionpolymerase chain reaction(RT-PCR)analysis was *** was used to examine the activity of the nuclear factor-kappa B(nF-κB)signaling pathway and the production of TJs *** analyses were employed to localize the ***-linked immunosorbent assay(ELISA)and RT-PCR were utilized to observe changes in interleukin(IL)-1β*** investigate the role of nF-κB signaling activation and IL^(-1)βin Sema7A’s anti-barrier mechanism,we employed 0.1μmol/L IκB kinase 2(IKK2)inhibitor IV or 500 ng/mL IL^(-1)receptor(IL-1R)antagonist.●RESULTS:Treatment with Sema7A resulted in decreased TEER and increased permeability of Dextran-FITC in HCEs through down-regulating mRnA and protein levels of TJs in a time-and dose-dependent manner,as well as altering the localization of ***,Sema7A stimulated the activation of inhibitor of kappa B alpha(IκBα)and expression of IL-1β.The anti-barrier function of Sema7A was significantly suppressed by treatment with IKK2 inhibitor IV or IL-1R antagonists.●COnCLUSIOn:Sema7A disrupts barrier function through its influence on nF-κB-mediated expression of TJ proteins,as well as the expression of IL-1β.These findings suggest that Sema7A could be a potential therapeutic target for the diseases in corneal epithelium.

A General Simulation Method for Complex Deformation of Irregular-Shaped Origami Configurations

Most existing treatments for origami-folding simulations have focused on regular-shaped *** article aims to introduce a general strategy for simulating and analyzing the deformation process of irregular shapes by means of computational capabilities *** better simulate origami deformation with folding orders,the concept of plane follow-up is introduced to achieve automated computer simulation of complex folding patterns,thereby avoiding intersection and penetration between *** on the evaluation criteria such as the lowest storage energy with tightening and the fastest pace from tightening to unfolding,the optimal crease distribution patterns for four irregular(‘n’-,‘T’-,‘O’-,and‘P’-shaped)origami configurations are then presented under five *** the dimensions of the origami are fixed,it is discovered that simpler folding patterns lead to faster deformation of the origami *** the folding complexity is fixed,higher strain energy results in more rapid origami expansion.

Suitable region of dynamic optimal interpolation for efficiently altimetry sea surface height mapping

The dynamic optimal interpolation(DOI)method is a technique based on quasi-geostrophic dynamics for merging multi-satellite altimeter along-track observations to generate gridded absolute dynamic topography(ADT).Compared with the linear optimal interpolation(LOI)method,the DOI method can improve the accuracy of gridded ADT locally but with low computational ***,considering both computational efficiency and accuracy,the DOI method is more suitable to be used only for regional *** this study,we propose to evaluate the suitable region for applying the DOI method based on the correlation between the absolute value of the Jacobian operator of the geostrophic stream function and the improvement achieved by the DOI *** verifying the LOI and DOI methods,the suitable region was investigated in three typical areas:the Gulf Stream(25°n-50°n,55°W-80°W),the Japanese Kuroshio(25°n-45°n,135°E-155°E),and the South China Sea(5°n-25°n,100°E-125°E).We propose to use the DOI method only in regions outside the equatorial region and where the absolute value of the Jacobian operator of the geostrophic stream function is higher than1×10^(-11).
