染色体外环状DNA(extrachromosomal circular DNA,eccDNA)是真核生物细胞核内游离于染色体外的一类特殊环状DNA分子,来源于染色体基因组但独立参与多种细胞生理或病理过程,与肿瘤的发生发展密切相关。eccDNA的发现可以追溯到20世纪70年代,随着分子生物学技术的发展,研究者们逐渐揭示了其在癌症中的重要功能。eccDNA的形成是一个涉及多种机制的复杂过程,包括断裂-融合-桥循环、染色体碎裂、易位-缺失-扩增、游离体模型以及复制叉停滞和模板切换等。eccDNA,尤其是大分子量eccDNA(大于1 Mb,又惯称ecDNA),在促进癌基因扩增与活化、驱动肿瘤异质性和耐药性方面发挥重要作用,并展现出作为肿瘤液体活检分子标志物的广阔应用潜力。本文将综述eccDNA发现历史、生成机制、在肿瘤生物学中的功能作用、在不同类型肿瘤中的特异性以及检测分析方法的最新研究进展,并对其在未来肿瘤基础研究和临床实践中的应用前景进行展望,旨在为eccDNA在肿瘤基础研究及临床实践中的进一步探索提供参考。

为了提高情感分类的性能，本文提出了一种基于加权集成的序贯三支决策情感分类模型。该模型首先对评论数据集的边界域使用不同的分类器获取各自的预测概率，再根据历史分类性能对不同分类器的预测概率进行加权集成，然后根据阈值和代价损失分别进行三支决策，将评论划分为正类、负类和边界域。对于边界域进行序贯的集成概率预测，并根据概率和阈值进一步划分为新的正类、负类和边界域。直至最细粒度上的边界域，最终通过集成二支决策得到最终的分类结果。研究结果表明，该模型在中文计算机评论、酒店评论和服装评论数据集上性能优于现有方法，其中在酒店评论数据集上分类准确率达到86.75%，相比于基于硬投票集成的序贯三支决策情感分类提高了3.6%。

目的分析基于沟通达标理论的护理干预结合多形式健康教育在神经源性膀胱患者间歇导尿中的应用效果。方法选取2022年5月至2023年1月在我院神经外科住院的70例行间歇导尿治疗的神经源性膀胱患者作为研究对象,采用抛硬币法将其分为对照组和观察组,各35例。对照组接受常规护理,观察组接受基于沟通达标理论的护理干预结合多形式健康教育。比较两组的干预效果。结果干预后,两组的最大膀胱容量(MBC)均增加,充盈期逼尿肌压力(***)、残余尿量(RU)均降低,且观察组优于对照组(P<0.05)。干预后,两组的国际下尿路功能症状(LUTS)评分均降低,健康教育知识掌握评分均升高,且观察组优于对照组(P<0.05)。干预后,两组的恐惧心理、自尊心、接受程度评分均降低,且观察组低于对照组(P<0.05)。结论基于沟通达标理论的护理干预结合多形式健康教育在神经源性膀胱患者间歇导尿中的应用效果较好,可提高患者的健康教育知识掌握程度,改善尿动力学指标及膀胱功能,提高治疗接受度。

The mechanical behaviors of filamentous actin (F-actin) bundles play an essential role in filopodial protrusions at the leading edge of crawling cells. These bundles consist of parallel actin filaments that are hexagonally packed and interconnected via cross-linking proteins including α-actinin, filamin, and fascin. As pushing against the plasma membrane and/or external barriers, the actin bundles in filopodial protrusion inevitably encounter a compressive load. The bending stiffness and buckling stability of actin bundles are therefore important in determining the filopodia architecture and subsequent cell morphology. In this work, we employ a coarse-grained molecular dynamics model to investigate the buckling behaviors of cross-linked actin bundles under compression, which explicitly accounts for the properties of constituent filaments and the mechanical descriptions of cross-linkers. The bending stiffness of actin bundles exhibits a generic size effect depending on the number of filaments in the bundles, explicitly depending on the degree of inter-filament coupling. The distinct buckling modes are analyzed for bundles with different coupling states and strengths of cross-linkers. This study could clarify the stability and buckling mechanisms of parallelly packed actin bundles and the structure-function relations of mechanical components in filopodial protrusion.

To gain more understanding of cell-matrix adhesion,we consider an idealized theoretical model of a cluster of integrin-fibronectin bonds at the cell-matrix interface subjected to a dynamic *** distributions of bond traction and interfacial deformation are assumed to obey classical elastic equations,whereas the dissociation/association of individual bonds as well as unfolding/refolding of fibronectin domains are described by stochastic *** stochastic-elasticity coupling,we perform Monte Carlo simulations to investigate how the collective behavior and adhesion performance of the integrin-fibronectin-mediated interface are influenced by two characteristics newly incorporated in the modeling,i.e.,catchlike dissociation between integrin and fibronectin,and unfolding of repeated domains in *** probable unfolding of fibronectin domains is found to have profound effects on the resultant adhesion energy of the integrin-fibronectin-mediated interface,and governs the failure model transiting between uniform decay and catastrophic crack-like rupture.

Newton＇s polynomial interpolation may be the favourite linear interpolation in the sense that it is built up by means of the divided differences which can be calculated recursively and produce useful intermediate results. However Newton interpolation is in fact point based interpolation since a new interpolating polynomial with one more degree is obtained by adding a new support point into the current set of support points once at a time. In this paper we extend the point based interpolation to the block based interpolation. Inspired by the idea of the modern architectural design, we first divide the original set of support points into some subsets （blocks）, then construct each block by using whatever interpolation means, linear or rational and finally assemble these blocks by Newton＇s method to shape the whole interpolation scheme. Clearly our method offers many flexible interpolation schemes for choices which include the classical Newton＇s polynomial interpolation as its special case. A bivariate analogy is also discussed and numerical examples are given to show the effectiveness of our method.

Growing actin networks provide the driving force for the motility of cells and intracellular pathogens. Based on the molecular-level processes of actin polymerization, branching, capping, and depolymerization, we have developed a modeling framework to simulate the stochastic and cooperative behaviors of growing actin networks in propelling obstacles, with an emphasis on the size and shape effects on work capacity and filament orientation in the growing process. Our results show that the characteristic size of obstacles changes the protrusion power per unit length, without influencing the orientation distribution of actin filaments in growing networks. In contrast, the geometry of obstacles has a profound effect on filament patterning, which influences the orientation of filaments differently when the drag coefficient of environment is small, intermediate, or large. We also discuss the role of various parameters, such as the aspect ratio of obstacles, branching rate, and capping rate, in affecting the protrusion power of network growth.

Risk assessment of water security in Haihe River Basin during drought periods based on D-S evidence theory

The weights of the drought risk index （DRI）, which linearly combines the reliability, resiliency, and vulnerability, are difficult to obtain due to complexities in water security during drought periods. Therefore, drought entropy was used to determine the weights of the three critical indices. Conventional simulation results regarding the risk load of water security during drought periods were often regarded as precise. However, neither the simulation process nor the DRI gives any consideration to uncertainties in drought events. Therefore, the Dempster-Shafer （D-S） evidence theory and the evidential reasoning algorithm were introduced, and the DRI values were calculated with consideration of uncertainties of the three indices. The drought entropy and evidential reasoning algorithm were used in a case study of the Haihe River Basin to assess water security risks during drought periods. The results of the new DRI values in two scenarios were compared and analyzed. It is shown that the values of the DRI in the D-S evidence algorithm increase slightly from the original results of Zhang et al. （2005）, and the results of risk assessment of water security during drought periods are reasonable according to the situation in the study area. This study can serve as a reference for further practical application and planning in the Haihe River Basin, and other relevant or similar studies.
