AIM:To investigate the pathophysiological role of C/EBP homologous protein(CHOP)in severe acute pancreatitis and associated lung ***:A severe acute pancreatitis model was induced with 6 injections of cerulein(Cn,50μg/kg)at 1-h intervals,then intraperitoneal injection of lipopolysaccharide(LPS,7.5 mg/kg)in CHOP-deficient(Chop-/-)mice and wild-type(WT)*** were sacrificed under anesthesia,3 h or 18 h after LPS *** amylase,lipase,and cytokines[interleukin(IL)-6 and tumor necrosis factor(TNF)-α],pathological changes,acute lung injury,and apoptosis in the pancreas were *** amylase and lipase activities were detected using a medical automatic chemical ***-linked immunosorbent assay kits were used to evaluate TNF-αand IL-6 levels in mouse serum and lung tissue *** cells in sections of pancreatic tissues were determined by terminal deoxynucleotidyl transferase-mediated dUTPbiotin nick-end labeling(TUNEL)*** mouse carotid arteries were cannulated and arterial blood samples were collected for *** oxygenation index was expressed as PaO2/***:Administration of Cn and LPS for 9 and 24 h induced severe acute pancreatitis in Chop-/-and WT *** comparing Chop-/-mice and WT mice,we observed that CHOP-deficient mice had greater increases in serum TNF-α(214.40±19.52 pg/mL vs 150.40±16.70 pg/mL;P=0.037),amylase(4236.40±646.32U/L vs 2535.30±81.83 U/L;P=0.041),lipase(1678.20±170.57 U/L vs 1046.21±35.37 U/L;P=0.008),and IL-6(2054.44±293.81 pg/mL vs 1316.10±108.74pg/mL;P=0.046)than WT *** histopathological changes in the pancreases and lungs,decreased PaO2/FiO2ratio,and increased TNF-αand IL-6 levels in the lungs were greater in Chop-/-mice than in WT mice(pancreas:Chop-/-vs WT mice,hemorrhage,P=0.005;edema,P=0.005;inflammatory cells infiltration,P=0.005;total scores,P=0.006;lung:hemorrhage,P=0.017;edema,P=0.017;congestion,P=0.017;neutrophil infiltration,P=0.005,total scores,P=0

A Network Signal Timing Design Bilevel Optimization Model with Traveler Trip-Chain Route Choice Behavior Consideration

This study proposed a bilevel optimization model for the network signal timing design problem by considering the linkflows reflected by the trip-chain route choice behaviors of road users. The bilevel programming model is formulated based on theinteractions between signal timing control and trip-chain behavior, and a solution algorithm by combining variational inequalitysensitivity analysis, the generalized inverse matrix method, and a gradient projection approach revised for trip-chain user equilibriumis developed for the transportation design problem. The performance of the developed model framework was verified throughnumerical analysis under different test scenarios.

电热熔渣焊(Electro-slag Welding,ESW)为钢骨箱形柱制作柱内横隔板最有效率的焊接方法之一,因此台湾钢骨建筑结构的箱形柱构件广泛采用ESW作为柱内横隔板与柱板连接的焊接方法。由过去实尺寸梁柱接合试验研究成果发现,箱形柱内横隔板采ESW与柱板连接的梁柱接合抗震性能可靠度不佳,在未发展充分非弹性变形前发生破坏的几率较高。进一步针对ESW焊道制作与质量检测的相关研究也显示,传统ESW焊道的制作与检测质量仍有改善空间,并提出喇叭形扩大孔的ESW熔池改良细部。该研究共进行4组实尺寸梁翼盖板式增强型梁柱接合试验,研究参数包括梁翼板与箱形柱内横隔板的偏心、ESW焊接熔池细部与荷载历时。试验结果显示,若梁翼板未偏心且ESW焊道制作良好的试件,可提供5%rad层间位移角的变形容量。当梁翼板与柱内横隔板高程偏心1倍梁翼板厚度时,采用传统矩形ESW熔池细部的梁柱接合试件,其层间位移角约降至3%rad;箱形柱板与内横隔板连接的ESW采用改良喇叭形熔池细部的梁柱接合试件,具有5%rad以上层间位移角的抗震性能,甚至可高达8%rad而不会发生断裂。

目的:了解基质细胞衍生因子1α(SDF-1α)及其受体CXCR4在人根尖牙乳头干细胞(SCAP)的表达,研究SDF-1α对SCAP的趋化作用。方法:收集牙根未发育完全的人第三磨牙,体外分离培养SCAP;逆转录PCR检测SCAP中SDF-1α的表达水平、流式细胞术检测CXCR4在SCAP中的表达;设置阴性对照组与SDF-1α处理组(100 ng/mL),将Transwell小室与胶原凝胶结合建立三维迁移模型,共孵育7 d后通过激光共聚焦显微镜(40×)重建胶原图像,分析各组细胞在胶原支架内部纵向迁移的距离。结果:逆转录PCR结果显示SCAP表达SDF-1α条带,流式细胞术结果显示CXCR4主要存在于SCAP胞浆内(表达率57.49%-98.48%,n=4),在细胞膜表面鲜有表达(表达率0.3%-2.34%,n=4);对照组细胞纵向迁移距离较浅,在胶原表面下方0-100μm水平细胞量(38.3±4.2)显著高于SDF-1α组(9.7±2.5),差异具有统计学意义(t=6.72,P=0.004<0.05),而SDF-1α组细胞纵向迁移距离则较深,在100-200μm水平细胞量(37.0±2.7)显著高于对照组(8.7±1.5),差异具有统计学意义(t=-16.063,P<0.001)。结论:SCAP表达趋化因子SDF-1α及其受体CXCR4;SDF-1α与CXCR4发生特异性结合形成SDF-1α/CXCR4轴后,可以趋化SCAP发生体外定向迁移。
