Poly-L-ornithine blocks the inhibitory effects of fibronectin on oligodendrocyte differentiation and promotes myelin repair

The extracellular matrix surrounding oligodendrocytes plays an important role during myelination and remyelination in the brain.In many cases,the microenvironment surrounding demyelination lesions contains inhibitory molecules,which lead to repair failure.Accordingly,blocking the activity of these inhibitory factors in the extracellular matrix should lead to more successful remyelination.In the central nervous system,oligodendrocytes form the myelin sheath.We performed primary cell culture and found that a natural increase in fibronectin promoted the proliferation of oligodendrocyte progenitors during the initial stage of remyelination while inhibiting oligodendrocyte differentiation.Poly-L-ornithine blocked these inhibitory effects without compromising fibronectin’s pro-proliferation function.Experiments showed that poly-L-ornithine activated the Erk1/2 signaling pathway that is necessary in the early stages of differentiation,as well as PI3K signaling pathways that are needed in the mid-late stages.When poly-L-ornithine was tested in a lysolecithin-induced animal model of focal demyelination,it enhanced myelin regeneration and promoted motor function recovery.These findings suggest that poly-L-ornithine has the potential to be a treatment option for clinical myelin sheath injury.

The addition and substitution reactions across a variety of alkenes and alkynes have been playing an important role in organic synthesis.In particular,catalyses enabled by homogeneous complexes as well as heterogenous nanomaterials have been much received attentions over decades.Along with these blooming progresses in these fields,single-atom site catalysts(SACs)exhibit outstanding performances in terms of reactivity and selectivity,thus providing a new powerful strategy to upgrade these bulk chemicals.Herein,we summarize the reactions of alkenes and alkynes enabled by SACs,in which catalytic hydrogenation,hydrosilylation,hydroboration,hydroformylation,and cross coupling have been included.Moreover,preparations and fine structures of the corresponding SACs have been described,therefore providing a better understanding and overview to address the origin of catalytic activity in the reported examples.

Before the implementation of offshore oil and gas exploitation,it is essential to understand the various factors that influence the stability of submarine sediments surrounding the project.Considering the factors such as cost and operability,it is not feasible to assess the physical-mechanical properties of sediments covering the entire region by borehole sampling.In this study,the correlation between near seafloor seismic amplitude and the mean shear strength of shallow sediments was explored using seismic and core testing data from the northern continental slope area of the South China Sea.Results showed that the mean water content of sediments in the layer up to 12 m below the seafloor(mbsf)gradually increased with increasing water depth,and the mean shear strength tended to decrease rapidly near the 1000 m depth contour.The near seafloor seismic amplitude could reflect the mean shear strength of sediments in the 12 mbsf layer under seismic frequency of 65 Hz and wave velocity of 1600 m/s.When the mean shear strength was greater than 10 kPa or the water depth was less than 1000 m,there was a significant linear positive correlation between mean shear strength and near seafloor seismic amplitude.Otherwise,there was a significant linear negative correlation between mean shear strength and near seafloor seismic amplitude.On the basis of these correlations,the pattern of shear strength was estimated from near seafloor seismic amplitude and mapped.The mean shear strength of sediments above 12 mbsf gradually decreased with increasing water depth in the continental slope area,whereas little change occurred in the continental shelf and the end of the canyon.Within the canyon area,the mean shear strength of sediments was characterized by larger values in both sides of the canyon walls and smaller values in the canyon bottom,which was consistent with the infinite slope stability theory.The study provides a method for using near seafloor seismic amplitude data to guide sediment sampling design,and presents a continuous da

The selective synthesis of N^(2)-sulfonyl and N^(2)-H 1,2,3-triazoles via organocatalytic annulation of enaminone/enaminoester with sulfonyl azide has been realized. The unconventional selectivity providing N^(2)-sulfoyl 1,2,3-triazoles takes place in pure water, wherein the hydrogen bond effect between water and the intermediate resulting from enamine-azide corporation accounts for the novel reaction selectivity. On the other hand, the reactions conducted in DMSO specifically afford N^(2)-H 1,2,3-triazoles in the absence of such hydrogen bond effect.

Simiao Wan(SMW)is a traditional Chinese formula,including Atractylodis Rhizoma,Achyranthis Bidentatae Radix,Phellodendri Chinensis Cortex and Coicis Semen at the ratio of 1:1:2:2.It can be used to the treatment of diabetes.However,its bioactive compounds and underlying mechanism are unclear.This study was designed to screen the antilipolytic fraction from SMW and investigate its therapeutic mechanisms on hepatic insulin resistance.Different fractions of SMW were prepared by membrane separation combined with macroporous resin and their antilipolytic activity was screened in fasted mice.The effects of 60%ethanol elution(ESMW)on lipolysis were investigated in 3T3-L1 adipocytes stimulated by palmitic acid(PA)and high fat diet(HFD)-fed mice.In our study,ESMW was the bioactive fraction responsible for the antilipolytic activity of SMW and 13 compounds were characterized from ESMW by UHPLC-QTOF-MS/MS.ESMW suppressed protein kinase A(PKA)-hormone-sensitive lipase(HSL)related lipolysis and increased AMP-activated protein kinase(AMPK)phosphorylation in PA challenged 3T3-L1 adipocytes.AMPKa knockdown abolished the inhibitory effects of ESMW on IL-6 and HSL pSer-660,revealing that the antilipolytic and anti-inflammatory activities of ESMW are AMPK dependent.Furthermore,ESMW ameliorated insulin resistance and suppressed lipolysis in HFD-fed mice.It inhibited diacylglycerol accumulation in the liver and inhibited hepatic gluconeogenesis.Conditional medium collected from ESMW-treated 3T3-L1 cells ameliorated insulin action on hepatic gluconeogenesis in liver cells,demonstrating that the antilipolytic activity contributed to ESMW beneficial effects on hepatic glucose production.In conclusion,ESMW,as the antilipolytic fraction of SMW,inhibited PKA-HSL related lipolysis by activating AMPK,inhibiting diacylglycerol(DAG)accumulation in the liver and thereby improving insulin resistance and hepatic gluconeogenesis.

Glioma is a common tumor originating in the brain that has a high mortality rate.Temozolomide(TMZ)is the first-line treatment for high-grade gliomas.However,a large pro-portion of gliomas are resistant to TMZ,posing a great challenge to their treatment.In the study,the specific functions and mechanism(s)by which cortistatin(CORT)regulates TMZ resis-tance and glioma progression were evaluated.The decreased expression of CoRT was detected in glioma tissues,and highly expressed CORT was associated with a better survival rate in pa-tients with glioma.CORT overexpression notably decreased the capacity of glioma cells to pro-liferate and migrate in vitro and to form tumors in vivo.CORT overexpression also markedly suppressed the viability and enhanced the apoptosis of TMZ-resistant U251 cells by regulating MGMT,p21,and Puma expression.Importantly,CORT overexpression reduced the resistance of gliomas to TMZ in vivo.CORT expression Was negatively correlated with MGMT expression in both glioma tissues and cells,and it was found that CORT inhibited NF-kB pathway activation in glioma cells,thereby inhibiting MGMT expression.In conclusion,CORT regulates glioma cell growth,migration,apoptosis,and TMZ resistance by weakening the activity of NF-kB/p65 and thereby regulating MGMT expression.The CORT/NF-kB/MGMT axis might be regarded as a molecular mechanism contributing to the resistance of glioma to TMZ.Our data also suggest that CORT regulates the viability and metastatic potential of glioma cells,independent of its effects on TMZ resistance,providing evidence of novel therapeutic targets for glioma that should be evaluated infurther studies.

Trifluoromethyl group is one of the most frequently available and promising functional groups in pharmaceutical,agrochemical as well as material molecules.Developing synthetic methods for the synthesis of trifluoromethyl functionalized compounds by trifluoromethylation reactions occupies significant position in modern organic chemistry.On the other hand,as a typical tool of sustainable synthesis,transition metal-free synthesis possesses unique advantages in atom economy,easy product purification as well as excellent eco-tolerance.Consequently,transition metal-free trifluoromethylation reactions constitute the most important approaches for accessing trifluoromethylated molecules.Herein,the advances in the transition metal-free C—H trifluoromethylation reactions are reviewed for the first time.According to the updated state-of-art,both the catalytic reactions by conventional heating chemistry and the more sustainable photocatalysis are included,covering the trifluoromethylation reactions of C(sp^(3))–H bond,aromatic C(sp^(2))–H bond as well as the non-aromatic C(sp^(2))–H bond in different substrates over the last five years(2017—2021).

Modeling effect of cooling conditions on solidification process during thermal cycle of rollers in twin-roll strip casting

In the twin-roll strip casting process,molten steel solidifies by losing heat through its interface with the casting rollers.The heat extraction along this interface has an effect on the quality of the strips and should be affected by coating,rolls’material,and cooling water flow rate.It is necessary to understand the effect of these casting parameters on the solidification structure of twin-roll strip casting.A three-dimensional computational domain is set up to simulate the solidification process of molten steel and heat exchange between steel strip/air,coating,rolls,and cooling water in the channel of roll sleeves.The effect of the cooling water intensity and flow intensity of molten steel in the pool on the solidification structures is studied during the thermal cycle of rolls in the twin-roll strip casting.These predicted results are helpful to optimize casting parameters and improve the strip quality in the twin-roll strip casting process.

AIM: To evaluate the efficacy and safety of gemcitabine-oxaliplatin (GEMOX) combined with huachansu (cinobufagin) injection treatment in patients with locally advanced or metastatic gallbladder carcinoma (GBC), and to assess the quality of life (QOL) of such patients. METHODS: Twenty-fi ve patients with locally advanced or metastatic GBC were treated with intravenous gemcitabine (1000 mg/m2) over 30 min on days 1 and 8, 2 h infusion of oxaliplatin (120 mg/m2) on day 1, and 2-3 h infusion of huachansu (20 mL/m2) on days -3-11, every 3-4 wk. Treatment was continued until occurrence of unacceptable toxicity or disease progression. QOL of patients was assessed by the EORTC QLQ-C30 at baseline, at the end of the fi rst, third and sixth chemotherapy cycles, and 1 mo after the treatment. RESULTS: Among the 25 patients with a median age of 64 years (range 42-78 years), 23 were evaluable in the study. A total of 137 cycles of therapy were performed and the median cycle was 5 (range 1-8) per patient. Out of the 23 patients whose response couldbe evaluated, 8 partial responses (PR) were observed (34.8%), while 7 patients (30.4%) demonstrated a stable disease (SD). The disease control rate was 65.2%. Progression of cancer was observed in 8 (34.8%) patients. The median progression-free and overall survival time was 5.8 mo (95% CI: 4.5-7.1 mo) and 10.5 mo, respectively. The therapy was well tolerated, with moderate myelosuppression as the main toxicity. Anemia grade 2 was seen in 16.0%, neutropenia grade 3 in 8.0% and thrombocytopenia grade 3 in 24.0% of patients, respectively. Non-hematologic toxicity ranged from mild to moderate. No death occurred due to toxicity. The QOL of patients was improved after chemotherapy, and the scores of QOL were increased by 10 to 20 points. CONCLUSION: GEMOX combined with huachansu (cinobufagin) injection is well tolerated, effective, thus improving the QOL of patients with advanced GBC.
